1. PENDAHULUAN 2.

BAHAN DAN METODE Mutagenesis in silico Visualisasi dari struktur molekul GFPuv ( PDB entri 1B9C ) dilakukan dengan menggunakan PyMOL . Virtual glisin - scanning mutagenesis dicapai dengan mengganti residu masing-masing permukaan dengan glisin sehingga dapat meminimalkan rantai samping besar. Mutan sehingga menimbulkan pelarut terkena analit channel pada permukaan protein setelah asam amino substitusi ( s ) dipilih untuk penyelidikan lebih lanjut .

Mutagenesis situs - diarahkan untuk asam amino substitusi F165G dan H148G pada GFPu Ekspresi dan pemurnian protein Kultur starter dibuat dengan inokulasi suatu koloni tunggal menjadi 5 ml LB broth yang mengandung 100 mg / ml ampisilin dan tumbuh dalam semalam pada 37 C dengan agitasi pada 150-180 rpm. Pengujian stabilitas termal dan pH Semua mutan GFP pori yang mengandung menjadi sasaran perlakuan panas pada suhu tertentu ( 25-90 C ) selama 10 menit dan kemudian didinginkan dengan cepat ke suhu kamar fluorescence kemudian diukur pada 509 nm ( FLX 800 fluorometer , BIO TEK instrumen, USA ) pada saat aktivasi dengan 395 lampu nm. Secara paralel , protein individu diinkubasi dalam Tris - penyangga ( mengandung 5 mM Tris - HCl , 10 mM glisin , 10 mM natrium sitrat atau 10 mM natrium fosfat ) pada berbagai pH selama 10 menit sebelum pengukuran fluoresensi . HASIL DAN DISKUSI Mutagenesis In silico yang mengandung mutan GFPuv Pemodelan molekul mengungkapkan saluran analit potensial pada permukaan GFP barel di Phe165 terletak pada untai kedelapan setelah bermutasi ke glisin (Gambar 2A ) . Prediktabilitas tinggi tersebut juga dapat menjelaskan untuk penemuan pembentukan pori pada His148 (Gambar 2B ) , yang konsisten dengan penyelidikan lain pada YFP dan BFPms1 [ 27 , 28 ] . Hasil dari studi pemodelan menunjukkan bahwa mutan ganda H148G/F165G memberikan diameter ukuran pori yang lebih besar dari 5,5 (Gambar 2C ) , dibandingkan dengan mutan tunggal H148G dan F165G dengan ukuran pori hanya 4,5 dan 4 (Gambar 2B dan 2A ) . His6GFPuv/F165G menunjukkan intensitas fluorescent tertinggi dibandingkan dengan His6GFPuv dan mutannya. Fluorosensi spektrum pori yang mengandung GFPs Eksitasi dan emisi maxima semua pori yang mengandung mutan ditemukan masingmasing pada 403 nm dan 510 nm. Ini mengungkapkan sebuah perubahan non-signifikan pada spektrum fluoresensi (Gambar 4) pada substitusi dari Phe 165 dan glisin 148. Namun, perbedaan besarnya pada emisi fluorosensi terdeteksi terutama pada mutan ganda, H148G/F165G. Selain itu, puncak kromofor anionik (terletak pada 478 nm) adalah khas berkurang setelah penggantian His148 (ditunjuk pada H148G dan H148G/F165G mutan). Temuan kami sangat mendukung peran His148 untuk mempertahankan aktivitas fluoresensi intrinsik melalui ikatan hidrogen kromofor [26, 32]. Proses pergantian residu ini terbukti mengurangi penyerapan anionik kromofor serta mempengaruhi stabilitas konformasi [26]. Semua temuan ini menunjukkan pentingnya F165G menyediakan saluran pelarut tanpa dapat mempengaruhi sifat fluorosensi. Stabilitas pori yang mengandung GFPs Pori yang mengandung mutan lebih peka pada kondisi panas dan asam-basa. Semua mutan mampu berfluorosensi pada suhu 60 0 C dalam waktu 10 menit. yang menyebabkan fluorosensi mutan turun menjadi nol sedangkan His6GFPuv dan His6GFPuv/F165G mampu mempertahankan fluorosensi hingga 40% dan 30 (Gambar 5A). Substitusi pada posisi 148 menyebabkan gangguan secara keseluruhan pada struktur protein destabilisasi dan fluktuasi structural GFP barel pada H148G.[26] Pengurangan aktivitas fluorosensi berkorelasi dengan penurunan pH seperti yang dapat dilihat dari proton dan fluorosensi kurva respon (Gambar 5B) untuk HisGFPuv dan variannya. Pada pH asam yang ekstrim (~ pH 4.0), tidak terjadi fluorosensi pada semua mutan. Namun, perubahan fluorosensi yang relatif pada mutan His6GFPuv dan F165G < dari mutan H148G dan H148G/F165G. His6GFPuv dan His6GFPuv/F165G memiliki pH stabilitas yang sama, sedangkan His6GFPuv/H148G dan His6GFPuv/H148G memiliki pH sensitif. Pada pH 6,0 ,mutan His6GFPuv dan F165G masih mampu mempertahankan aktivitas fluoresensi hingga 90 % sedangkan H148G dan varian H148G/F165G hanya 40 % fluoresensi. Semua protein GFP memberikan aktivitas fluorosensi maksimal ketika pH dinaikkan di atas 7,5.
