ABSTRAK

Citicoline (CDP-choline) merupakan senyawa endogen yang berfungsi
sebagai senyawa intermediate dalam sintesis fosfolipid membran, sintesis asetilkolin
dan sebagai donor metal. Citicoline dapat memberikan efek kolinergik dan
neuroprotektif. Citicoline juga dapat meningkatkan integritas struktural dan
fungsional membran sel saraf serta membantu perbaikan membran sel saat terjadi
cedera sel. Tujuan penelitian ini adalah untuk mengetahui peran citicoline terhadap
perbaikan fungsi motorik pascacedera saraf sciaticus. Penelitian ini merupakan
penelitian eksperimental laboratorik yang menggunakan 40 ekor tikus putih jantan
(Rattus norvegicus.). Tikus dibagi kedalam 4 kelompok percobaan yaitu kelompok I
sebagai kelompok kontrol positif, kelompok II sebagai kelompok kontrol negatif
sedangkan kelompok III, IV sebagai kelompok perlakuan. Kelompok kontrol positif
adalah kelompok yang dibalut dengan spons gelatin yang dibasahi dengan 0.4 cc
larutan saline 0.9%, kelompok kontrol negatif adalah tidak diberikan perlakuan
sedangkan kelompok perlakuan adalah kelompok yang dibalut dengan spons gelatin
yang dibasahi dengan citicoline 0.4 cc dan citicoline 0,8 cc. Penilaian perbaikan
fungsi motorik dapat dilakukan dengan metode extensor postural thrust. Metode ini
dilakukan dengan cara menyentuhkan kaki tikus pada timbangan sehingga kaki tikus
mendorong timbangan tersebut. Dari hasil analisis extensor postural thrust
didapatkan bahwa citicoline berperan dalam proses perbaikan fungsi motorik saraf.
Dengan demikian dapat disimpulkan bahwa citicoline dapat berperan dalam proses
perbaikan fungsi motorik pascacedera saraf sciaticus pada tikus putih (Rattus
norvegicus).

ABSTRACT

Citicoline (CDP-choline) is an endogenous compound that functions as an
intermediate compound in the synthesis of membrane phospholipids, acetylcholine
synthesis and as metal donor. Citicoline can provide cholinergic and neuroprotective
effects. Citicoline can also improve the structural and functional integrity of nerve
cell membranes and help repair cell membranes during cell injury. The purpose of
this study was to determine the role of citicoline for the improvement of motoric
nerve function after sciaticus nerve injury. This study is an experimental laboratory
study using 40 male white rats (Rattus norvegicus.). Rats were divided into 4
experimental groups: the group I as a positive control group, group II as a negative
control group, while group III, IV as the treatment group. Positive control group is
the group that was wrapped with gelatin sponge soaked with 0.4 cc of 0.9% saline,
negative control group was given no treatment, while the treatment group was the
group that was wrapped with a gelatin sponge soaked with citicoline 0.4 cc and
citicoline 0.8 cc. Assessment of motoric function improvement can be done using
extensor postural thrust method. This method is done by touching the mouse feet on
the scales so that the foot of rat pushed the scales. From the analysis of extensor
postural thrust was found that citicoline role in the improvement of motoric nerve
function. It can be concluded that citicoline may play a role in the process of
improvement of motoric function after sciaticus nerve injury in white rat (Rattus
norvegicus).
