IDENTIFIKASI POLIMORFISME PROGINS (INSERSI Alu 306-pb) GEN
RESEPTOR PROGESTERON PADA PENDERITA ENDOMETRIOSIS
DI RS DR. MOHAMMAD HOESIN PALEMBANG (Muhammad Nur Shaffrial, Januari 2014, 64 halaman) ABSTRAK Latar Belakang: Endometriosis merupakan suatu penyakit yang didefinisikan sebagai pertumbuhan jaringan endometrium dengan glandula dan/atau stroma diluar kavum uteri yang umumnya terjadi pada wanita usia reproduksi. Etiologi endometriosis sampai saat ini belum diketahui secara pasti, tetapi terdapat beberapa faktor yang mungkin berperan dalam proses terjadinya endometriosis, termasuk faktor keturunan dan mutasi genetik. Pada polimorfisme PROGINS (insersi Alu 306-pb) gen reseptor progesteron, PR-A yang bermutasi menghambat aktivitas PR-B. Akibatnya, PR-B mengalami penurunan sehingga terjadi peningkatan rasio PR-A/PR-B. Tujuan penelitian ini adalah mengidentifikasi polimorfisme PROGINS (insersi Alu 306-pb) gen reseptor progesteron pada penderita endometriosis di Rumah Sakit Dr. Mohammad Hoesin Palembang. Metode: Penelitian ini merupakan penelitian deskriptif observasional terhadap 48 penderita endometriosis di RS Dr. Mohammad Hoesin Palembang. Identifikasi polimorfisme PROGINS (insersi Alu 306-pb) gen reseptor progesteron dilakukan dengan menggunakan teknik Polymerase Chain Reaction (PCR). Hasil: Genotip T1/T1 (wild type) ditemukan pada 43 penderita (89,6%) dan genotip T1/T2 (heterozigot mutan) pada lima penderita (10,4%). Frekuensi alotip dari 48 penderita bernilai 91 (94,8%) untuk alotip T1 (wild type) dan lima (5,2%) untuk alotip T2 (polimorfik). Simpulan: Genotip T1/T1 (wild type) dan alotip T1 (wild type) lebih banyak teridentifikasi pada penderita endometriosis di RS Dr. Mohammad Hoesin Palembang. The etiology of endometriosis remain unclear until now, but there are several factors that may play a role in the occurrence of endometriosis, including heredity and genetic mutation. In PROGINS (Alu 306-bp insertion) progesterone receptor gene polymorphism, mutated PR-A was shown to inhibit the PR-B activity. As a result, PR-B decreased, therefore increasing the PR-A/PR-B ratio. This study aims to identify PROGINS (306-bp Alu insertion) progesterone receptor gene polymorphism in endometriosis patients at Dr. Mohammad Hoesin General Hospital, Palembang. Method: This study is an observational descriptive study on 48 endometriosis patients at Dr. Mohammad Hoesin General Hospital, Palembang. Identification of PROGINS (306-bp Alu insertion) progesterone receptor gene polymorphism was performed through Polymerase Chain Reaction (PCR). Results: The T1/T1(wild type) genotype was identified in 43 patients (89,6%) and the T1/T2 (heterozygous mutant) genotype in 5 patients (10,4%). The allotype frequencies in 48 patients were 91 (94,8%) for the T1 (wild type) allotype and 5 (5,2%) for the T2 (polymorphic) allotype. Conclusion: The genotype T1/T1 (wild type) and allotype T1 (wild type) were more commonly identified in endometriosis patients at Dr. Mohammad Hoesin General Hospital, Palembang.
