PRELIMINARY ACTIVITY FOR Primary Productivity

Problem
To investigate the amount of dissolved oxygen in water sample.
To study the effect of photosynthesis and cellular respiration in dissolved
oxygen.
To find out the gross productivity of oxygen in water sample.

Purpose
To investigate the amount of dissolved oxygen in water sample.

Hypothesis
If the photosynthesis occurs , then the amount of DO will increase.

Materials
1. 2 sample water
2. Beaker
3. DO probe

4. Data logger
5. DO Vernier
6. Distilled water

Procedure
1. Prepare the Dissolved Oxygen Probe for use following instructions from your
teacher .Place the probe in a beaker filled with about 100 ML of distilled water
2. Connect the Dissolved Oxygen Probe to the interface on open the datacollection program. Allow the probe to stay in the water for 5 minutes as the
probe warms up.
3. Collect DO data.
a. Place the tip of probe into the water sample being tested. Submerge the
probe tip to a depth of 4-6 cm.
b. Start data collection. Gently stir the probe in the water sample. Note: It is
important to keep stirring until you have finished collecting data.
c. Continue stirring and data collection until the reading have been
relatively stable (stable to the nearest 0.2 mg/L)for about 30 seconds , then
stop data collection.
d. Select the stable region of your graph , then display Statistics for that
region. Note and record the mean value for that region as the DO of the water
sample.

Discussion
In dark bottle, the amount of DO would increase a little bit because it has
more cellular respiration than photosynthesis. On the other hand, the amount of
DO in light bottle rapidly increase because there is a lot of photosynthesis. Our
hypothesis is false because generally amount of DO in dark bottle should
decrease because in the dark bottle does not have any photosynthesis and the
Oxygen was left over in the dark bottle would be used for cellular respitration ,
thus the amount of DO in dark bottle before the experiment should be less than
the amount of DO after the experiment. Our next experiment , we will have
more carefulness because we think that the cause of this mistake is that we did
not completely wrap the dark bottle.
