Hydrolysis of Polysaccharides

A. Acid Hydrolysis
In a test tube, 5 drops of conc. HCl
to 5 mL of isolate. The test tube were
covered with cotton and it was boiled
in a water bath for 30 minutes. After
30 minutes, the hydrolysate was kept
for Benedicts test.
B. Enzymatic Hydrolysis
In a beaker, place 10 mL of
isolated carbohydrate. Then, the
mouth was rinse with warm water for
a minute and the washings were
collected in a beaker to collect saliva.
2.3 mL of the washings were added to

the isolated carbohydrate. The


solution was allowed to stand at room
temperature for 30 minutes and the
viscosity of the solution was noted.
The solution was introduced into a
dialyzing bag and was suspended
overnight in a small flask with 50 mL
distilled water. The dialyzing bag was
then removed and discarded. The
solution left in the flask was decanted
to 20 mL for heating. The solution
inside the flask was concentrated by
heating to the volume of 10 mL. The
hydrolysate was tested for the
presence of reducing sugar by
performing Benedicts test.
