(COX-2) PADA MAKROFAG TIKUS PUTIH YANG DIINDUKSI COMPLETE
FREUND’S ADJUVANT (CFA)

INTISARI

Latar belakang : Terapi arthritis rematoid saat ini

umumnya digunakan adalah DMARD(Disease Modifying Anti
Rheumatoid Drugs), OAINS (Obat Anti Inflamasi Non
Steroid), dan Glukokortikoid. Namun penggunaan obat –
obatan ini mengakibatkan banyak efek samping berbahaya.
Mengkudu (Morinda citrifolia L) telah banyak diteliti
sebagai agen anti inflamasi diharpakan dapat menjadi
pilihan untuk terapi RA
Tujuan : Mengetahui efek ekstrak air buah mengkudu
terhadap penekanan ekspresi Enzim Siklooksigenase-2(COX-
2) pada makrofag tikus putih yang diinduksi Complete
Freund’s Adjuvant (CFA).
Metode : Pada penelitian ini digunakan 30 ekor tikus
putih betina galur Wistar dengan berat badan antara 200-
220 gram yang dibuat model tikus arthritis dengan cara
diinduksi 0,1 ml CFA. Tikus dibagi menjadi 5 kelompok,
yaitu kelompok I (kontrol negatif) diberi akuades peroral
1 kali setiap hari selama 2 minggu; kelompok II(kontrol
positif) diberi methotrexate 0,189 mg peroral 1 kali
seminggu selama 2 minggu; kelompok III,IV, dan V masing-
masing diberi ekstrak air yang berasal dari buah mengkudu
2,5 gram, 7,5 gram, 22,5 gram peroral 1 kali sehari
selama 2 minggu. Setelah perlakuan selama 2 minggu, tikus
dikorbankan dan diambil sendi kakinya untuk kemudian
dibuat preparat Patologi Anatomi dengan proses
IHCs(immunohischemical staining). Dari hasil pengecatan
tersebut dilakukan penghitungan ekspresi COX-2 pada
makrofag untuk seterusnya dianalisis menggunakan ANOVA
one way dengan derajat kemaknaan 95%.
Hasil : Tidak Terdapat perbedaan bermakna penekanan
ekspresi COX-2 makrofag pada pemberian ekstrak air yang

berasal dari buah mengkudu 2,5 gram, 7,5 gram dan 22,5
gram terhadap kontrol negatif akuades.
Kesimpulan : Pemberian ektstrak air yang berasal dari 2,5
gram, 7,5 gram dan 22,5 gram buah mengkudu peroral setiap
hari selama 2 minggu tidak menunjukkan perbedaan yang
secara statistik berarti dengan grup kontrol akuades
(kontrol negatif) pada penekanan enzim siklooksigenase II
(COX-2) sel makrofag tikus yang diinduksi Complete
Freund’s Adjuvant (CFA).

ABSTRACT

Background : Rheumatoid Arthritis therapy now rely on

DMARD,NSAID, and Glucocorticoid most of the time. Harmful
side effect of these regimen has been recorded. Research
on less toxic agent now become an alternatives. Noni
(Morinda citrifolia L) has widely known as traditional
herbal remedies. Studies about Inflammatoric suprressor
act of Noni has shown a potential effect as a novel anti-
inflammatoriy agent.

Aim : To revealed an inflamtoric suprresing effect of

Noni fruit water extract on COX2 enzyme at macrophages
cell of rheumatoid arhtritic rat induced by CFA.

Method: This study is an experimental studies. Using 36

female rate, wistar strain, wighted around 200-220gr
induced by 0,1 CFA on rigt knee joint to show Rheumatic
Arthrits pathology. Divided to 6 group, first group, role
as a negative control received distillate water orally
once a day within 2 weeks. Second,third, and fourth group
received noni fruit on different dosages which later
water extracted, each 2.5gr, 7.5gr, 22.5 gr orally once a
day within 2 weeks. Fifth group role as first positive
control group received 0.189mg methotrexat orally once a
week within 2 weeks and sixth group role as second
postitve control group received 0,378 mg orally once a
day within 2 weeks. After 2 weeks, rat slaughtered and

right knee joint taken to make microscopic slide using
COX2 spesific immunohistochemical staining (IHC’s) at
Department of Pathology FM UGM. Positive and negative
COX-2 expresssed macrophages counted on microscopic
picture taken by miroscop and image analyzer then
analyzed using ANOVA one way method with 95% confidence
interval.
Results : There is no significant differences between the
group treated by Noni fruit water extract compare to
negative control group treated with distilatted water on
expression of COX2 at macrophages cell of rheumatoid
arthritic rat.

Summary : Noni fruit water extract failed to show a

inhibitoric action to COX2 at macrophages cell of
rheumatoid arthritic rat compare to the negative group.
