Materials
θ Large liquid nitrogen tank
θ Wrench from Bruce lab
θ Styrofoam box
θ Forceps

Protocol

Twist liquid nitrogen tank

valve closed (clockwise)
Make sure that tank valve is
completely closed before
untwisting the connector

Using wrench untwist the

connector to the liquid
nitrogen tank
CAREFUL: Wear freeze-
proof gloves; otherwise keep
hands clear of liquid nitrogen
stream Place styrofoam box at
nozzle and carefully open
liquid nitrogen tank valve
(counterclockwise)

10 seconds should provide

enough liquid nitrogen for
flash freezing; close valve

Grasping sample tube with

forceps dip specimen in one Very important that the valve

nitrogen tank in order to provide liquid
nitrogen to the deep freezer

Reconnect nozzle of tank to

freezer connector and
tighten with wrench; then re-
open tank valve
