Designing an In-silico proteinprotein interaction profile and Gene regulatory network for Kaposis sarcoma associated Herpes Virus

Herpes Simplex Virus 8 (HSV8) is the causative agent for a cancerous tumor of connective tissue which is widely known as Kaposi's sarcoma. The cancer involves skin, lungs, gastrointestinal tract, and other organs and is often associated with AIDS. Inspired by an urge to design an effective drug against the virus through a systems approach, here we have tried to establish a regulatory network involving key host virus interactions. The network comprises uncharacterized ORFs of the virus as well as interacting host proteins. We collected sequence information from public databases and structurally characterized the protein products of the ORFs using online servers- PSIPRED, CPHmodels- 3.0 and I-TASSER. For functional analysis of the individual ORFs MotifFinder tool was used. Consequently partner profile for each ORFs was predicted using String 9.0. Data mining tool Ali Baba further helped us to integrate predicted networks and interactions. SAGE and Microarray data, signifying gene expression temporally, was useful to quantify association between the nodes.To compare our interaction topology with standard known ones, PathBLAST and NetworkBLAST were used. Moreover, MicroRNAs were predicted applying online tools and databases like RNAFold, MatureBayes, Mipred etc. Targets of the predicted MicroRNAs were also analyzed applying Miranda in the host and Human. Finally, with the help of Cytoskape and CentiBin bioinformatic platform the results were represented as networks. Our study establishes both a protein-protein interaction profile and a gene regulatory network. The topology of the protein-protein interaction profile helped us to predict a pathway whereas the gene regulatory network revealed binary relationships among the genes. This study needs to be further investigated and validated in vitro. The network we propose here can be used to design antivirals in the future against HSV8 by targeting the network.

Science has always been a passion to me since high school days. A basic and fundamental difference with the glimpse of epigenomics, the whole structure of life is a mystery to me. I started finding the answer through the available source, and then I decided to pursue my Undergrad in Zoology and then did my MS in Biotechnology with a thesis in Molecular Biology. I am still on my way searching the answer and the beauty of nature in creating such ways to transform the characters into a multi tasking approach still amaze me.

To build up a greater or significant community in science it is very important to establish an interacting environment to do the sharing and thoughts. I believe interacting with researchers around the globe will surely enhance my ideas and thoughts towards science more precisely. This opportunity will apparently help me in understanding the trends and challenges in the expansion of different applied and classical biological sectors like Cell biology, Genetics, Human biology, Microbiology, and newly developed biological techniques. This knowledge will not only allow me in better understanding biological and natural principles but will also allow me to learn all the breakthrough of new discoveries and the trends & challenges in the applied biology sector. This conference will also give a chance to interact with key experts and leaders from all around the world. Therefore, I believe attending in this conference of FAOBMB will provide the necessary background for me to be to anticipate and to react to ever-changing environments using the experience of the experts.

In this regard I would like to mention that during my undergraduate and graduate studies, emphasis was given entirely to understand the fundamental biological principles that govern modern day of Biological advancements. I also learned about other topics such as bio-safety, laboratory management and recombinant DNA technology. I am highly interested to deal with novel Biological techniques to follow up the regulation and corresponding genetic pathways that lead to specific functional characteristics and want to find out specific regulatory promoters and specific signaling pathways from both the indoor and outdoor influences. My aim in life is to explore and to see the inner beauties of genetic mechanisms through all possible ways. I am very much interested in Molecular Biology, Cell Biology, Drug Designing, Neural Biochemistry and Bioinformatics. Being a part of the program, I will get a more expand area on my thoughts and beliefs. I would really love to be a part of the event and waiting like to see a horizon through it.
