Q Photometry data acquisition method

x Measuring basically is done with gain 1, but when the lamp has become darker or when the sample liquid is darker, the gain is increased (binary to 128 times) for measuring. x There is no gain adjustment for each element, and the degree of scatter is corrected by Measure reset. x Calculation from the raw data of the A/D output to the OD value is performed using a constant as ODO with Measure reset when the PDA output is -6 V. (1) Measure reset (circuit constant measuring) x Measuring is performed for compensation of circuit constant changes from part differences, environment, and aging changes. x No compensation is made at the time of the measuring reset measuring, and offset 0 and the default gain value are used. x This is performed at the time of measuring start and at the start of photo calibration. Gain switching x Gain switching is done according to the following threshold values u2 4.5V (399AH) (3) u4 2.25V (1CCDH) u8 1.125V (0E66H) u16 562.4mV (0733H) u32 281.4mV (039AH) u64 140.7mV (01CDH) u128 70.2mV (00E6H)

(2)

Raw data and OD value Offset Gain Gain x Raw data = {(Measuring value V os1) y A1 Vos2} y A2 x OD value = {42936 (A7B8H) LOG 10 (raw data)} u 10/6 n When the PDA output is 6 V * PDA offset value compensation is not performed

Q Measuring command contents

O This circuit board executes various operations by commands from the ANL CPU. Here, the operation contents for each command will be explained.
(1) Normal photometry x Photometry is performed for the 28 points from P0 to P27. x The photometry wavelengths are as follows Photometry point First half two wavelengths Last half two wavelengths P0, Px, P1-P27 Wavelength specified from main n P1 m O3, O5 for LIH P2 m O7, O8 for LIH P3 m O9, O13 for LIH

x The wavelength setting is put out to OUT21H as 0H to CH for O1 to O13. x Of the specified wavelengths from MV1019 (ANL control circuit board), the main wavelength is measured on the Main side, and the sub-wavelength is measured on the Sub side. x The wavelength for LIH is measured on the Main side for O3, O7, and O9, and on the Sub side for O5, O8, and O13.

(2) One revolution photometry x Measuring is performed consecutively from Cuv. No.48 (P0) for 88 cuvettes. x Measuring with four wavelengths is performed for one cuvette. (First half main: O1, first half sub: O2, last half main: O3, last half sub: O4) x With 88 cuvettes and 13 wavelengths, 44 data are obtained for each wavelength. x After completion of measuring for 88 cuvettes, one revolution (88 cuvettes) is made to return to the origin. (3) DIAG photometry x Measuring is performed from Cuv. No. 48 (P0) consecutively for 13 cuvettes. x Measuring with one wavelength is performed for one cuvette. (Measuring on the Main side and on the Sub side is done with the same wavelength) x The same wavelength as for the first half is used for the last half, but the data are not used. (The data are discarded) x After completion of measuring for 13 cuvettes, one revolution (88 cuvettes) is made to return to the origin. Static photometry x Photometry is performed for the cuvette presently passing through the optical axis. x Photometry is performed for 13 wavelengths, and the raw data, as well as the gain and the OD value at the time of measuring are put out. (The conventional 0% photometry is obtained when the incoming light is cut off. When the wheel is turned by half a pitch, the light is cut off between the holder and the cuvette)

(4)

Q Others
(1) (2) Wheel initialization x Stop is performed 10 pulses after origin detection. Analog Switch relation x Please observe the following for D/A/PDA/GND switching, PGA gain switching, u 1/u 16 Amp switching, and wavelength switching. 1) Always set the total gain to u 1 before wavelength switching. 2) Do not select u 1 Amp, u 16 Amp twice or more, independent of Main side and Sub side.
