Heather Selin High HPV16 Viral Load By the year 2000 cervical cancer had 76,000 cases and

30,000 deaths were estimated. The majority of cervical cancer cases and deaths are in the countries were the women have little or no access to effective prevention services. During research they have noticed that women who are dying from cervical cancer are in the prime of their life. They are either raising children, taking care of their families, or contributing to their town. A lot of the deaths are unnecessary and treatable but with the limited access to health options the cancer progresses to where it is too late. These countries are lacking the programs that help detect cancer early on so it is more treatable. HPV DNA infections are highly influenced by viral number copies meaning measuring HPV load could improve predicting the value of HPV testing. They think that high HPV DNA viral load may be a candidate that could help identity high risk women. This study took place in Tegucigalpa, Honduras from January 2003-December 2005. 760 women were enrolled and 281of these women has cervical dysplasia and 479 women has normal cervical testing. After a formal physical and the proper testing was obtained 118 came out to be positive with HPV 16 who have cervical dysplasia and only 44 came out positive in the women who had normal cervical smears. They were only able to use 110 samples because there was not enough DNA on all the Samples to run the viral load. They used an Ayre Wooden spatula to obtain these samples and then they put them in a saline and tested from there. They took DNA from the thawed cells and added different buffers to wash the samples. They used DEIA to detect the presence of DNA by hybridization of SPF10 amplifiers. They also used the equation below to determine viral load.

Viral load (HPV copies/cell)=Number of HPV copies-cells/Number of G-Globin/2 They determined the highest average viral load of HPV16 were detected in women with HSIL with 75 HPV DNA copies/cells. LSIS groups came back positive for 9 HPV DNA copies/cells and on average 0.7 was found positive in women who had normal pap smears. To determine these results they added a real time PCR-base assay to the positive samples. 25 women with LSIL 72% were found positive for single HPV infection, 28% were found positive for multiple HPV infections. HSIL had 66% single HPV infections and 34% were multiple HPV infections. They found that the HSIL was the highest detected were as the lower vial load were still found in the negative pap smears. The results showed that the single infection category had the highest HPV viral Load than the multiple infections did. They found that women who had high HPV 16 Viral Load should be following up more consistently but money in these countries is a struggle. Doing follow up exams was the biggest limitation in the study because Honduras doesnt have a well-organized national cervical cancer program. They got more information from this study to help them understand the relationship between HPV 16 viral load on respect to cervical cancer in Honduran women.
