Approximate Time Required: 30 minutes Technical References: Aptio Automation Operators Guide Educational References: Aptio Automation Sample Processing Online Training Objectives: Upon completion of this exercise you will be able to perform the following tasks: Identify sample requirements Load and unload samples from the Input/Output Module Describe the sample processing sequence Search for samples in the database Deliver a sample to the Input/Output Module

Supplies Needed: Personal Protective Equipment as determined by your local regulations 15- 20 sample tubes with barcodes and work orders downloaded to Aptio Automation. Use both pre-spun and unspun sample tubes to complete this exercise. If your system is configured to load uncapped samples, use some of these as well. Helpful Hints: To avoid the disruption of laboratory workflow, consult with the Aptio Automation primary operator before starting this practical exercise. Complete the Aptio Automation Sample Processing Online Training before starting this Practical Exercise. If your Aptio Automation does not have a Centrifuge or Decapper module, use the appropriate sample tube type (Ex. spun, uncapped) for your system. Ensure that output racks of the appropriate type are available in the Input/Output Module before beginning the exercise. An answer key is provided at the end of this exercise. Please give these pages to your mentor prior to starting the practical exercise.

Exercise Activity and Questions Identify sample requirements Aptio Automation is configured based on the laboratorys needs. For a list of approved sample tube types, refer to the Aptio Automation Operators Guide. 1. Below are examples of sample containers found in the laboratory. For each one, state if they are acceptable for processing on your system and if not, the reason why. a. Glass tubes

c. Flat bottom plastic tubes

d. Sample cup nested inside of a 13X100 plastic tube

Load and unload samples from the Input/Output Module Note: Consult with the primary operator before performing these exercises so as not to disrupt laboratory operations. Input/Output Module lane configurations vary based on the laboratorys needs. Not all of the LED icons described below may be configured on your system. These exercises are designed for a system with both a Centrifuge and Decapper Module. If your system does not have these modules, load the appropriate sample tube types for your system. 2. When loading samples, it is important that the samples introduced into an input lane have the appropriate centrifugation and cap status designated for that lane. Next to each icon below, write the workorder status, centrifugation, and cap status of samples that are loaded into that lane.

3. When operating Aptio Automation it is important to have output racks in the Input/Output Module to allow the Input/Output Robot to unload samples. In the table below, enter the letter of the description for the icon shown. Answer LED Icon Description A. All sample tube routing is complete.

4. Determine which lanes on your Input/Output Module are designated for output racks and load empty racks into them. a. What action does the robot take after the rack has been inserted?

5. Ensure that all modules and instruments required to process your samples are online. a. Which screen in the User Interface displays all the module and instrument statuses?

b. In this screen, what color indicates that a module or instrument is available to process samples?

6. Select 5 Routine (capped, unspun) samples and record the sample IDs. Load them into a 48 tube sample rack. Note: If your system does not have a Centrifuge or Decapper Module, load the appropriate sample tube type for your system. a. Into which column and row do you place the first sample?

b. What pattern should you follow to load the rest of the samples? (columns and rows)

c. Why is it important to avoid empty spaces between samples in each column of the rack?

d. Should spun and unspun sample tubes be loaded into the same rack? If not, why?

7. Identify the appropriate Input/Output Module lane for loading these samples. a. What is the lane type into which this rack should be loaded?

8. Load the sample rack into a lane and observe the IOM Robot loading the samples. a. From which column and row does the robot start loading samples?

b. Which additional sample rack positions are checked after all sample tubes have been loaded onto the track?

9. Select at least 4 Routine and 2 Priority samples and record the sample IDs. Load the Routine samples into one rack and the Priority samples into another rack. Position both racks in front of the appropriate lanes. DO NOT INSERT. When ready, insert the Routine rack first. Once the robot has started loading the samples, insert the Priority samples into the Priority Input Lane.

a. What action does the IOM Robot take once the rack is inserted into the Priority Input lane?

b. What action does the IOM Robot take once all the samples are loaded from the Priority Input lane?

11. When the samples have completed processing, determine which output racks are available for removal. Check for sample errors before removing the rack. Note: Systems with a Refrigerated Storage Module may not send completed samples to the Input/Output Module. a. List the steps to check for sample errors in the User Interface.

Insert the rack into Input/Output Module and observe the samples as they travel around the track. a. List the sequence of the components encountered in order. Describe the sample processing sequence 12. a. Varies by site and system configuration. Basic sample processing sequence follows: 1. IOM Robot picks up a sample from and Input rack and places it in a carrier on the track. Sample may also be loaded by the Rack Input or Bulk Input Modules. 2. Barcode is read and sample ID is paired with the carrier. 3. Sample arrives at Centrifuge Module where CM Robot picks up the sample and places it into a bucket insert. Sample is weighed, balanced and placed in the centrifuge. 4. After centrifugation is completed, CM Robot places sample into a carrier on the track. 5. Barcode is read and sample ID is paired with the carrier. 6. Sample arrives at the Decapper Module. Cap is removed. 7. Sample arrives at an instrument for analysis. The instrument receives the workorder from the middleware and processes the sample. 8. Sample travels to all instruments required to complete the workorder test requests. 9. Sample arrives at the Sealer Module and is sealed. 10.Sample arrives at the Input/Output, Rack Output or Refrigerated Storage module.
