(Anredera cordifolia(Ten) Steenis) Leaves Extract to the Level of Male Rat Serum TNF- Which is Induced by Formaldehyde Chandra Wahyu Kurniawan description publisher Formaldehyde is one of the hazardous food preservative, its because of the residual metabolite from this compound is carsinogenic if its also generate free radicals very reactive to cause cellular damage of macromolecules and inflammation, then the cells mights eventually die, so the organ dysfunction will be the worst result of it. Binahong leaves (Anredera cordifolia(Ten) Steenis) contains flafonoids as their main secondary active metabolite which is very potential to play a big role as the antioxidant. Therefore, the author wants to prove the effect of binahong leaves extract as an antioxidant to the level of serum TNF- in maximum formaldehyde induced male rat. Laboratory experimental non-randomized post-test only control group is chosen as the design of this research. This research is held in 2 step of action. The first step is the optimization of formaldehide dose, and the second step is the optimization of Binahong leaves extract dose. In the first step of research, rats are treated with 1ml formaldehyde for each rat per day in 7 days, in the range of dosage 0 ppm(as the control), 25 ppm, 50 ppm, and 75 ppm. In the second step of the research, the rats are treated with the effective dose based from the result of the effective dose in the first step, and the rats are also treated with the Bimahomg leaves extract in the range of dose (25%, 50%, 100%) within the result is read by ELISA reader in the wave lenght of 450nm. The data is statistically analized with oneway ANOVA test then further its analyzed with Post Hoc LSD test. The result are catagorized as significan p if p < 0,05. Formaldehyde administration in the dosage 25 ppm, 50 ppm and 75 ppm. Significantly shew the increase of serum TNF- (p 0,05) and the result of serum TNF- respectively are (2197,57 IU/ml; 2458,65 IU/ml; 3065,88 IU/ml. After the administration of Binahong extract, the result of serum TNF- respectively are 25% (2316,00 IU/ml), 50% (2464,00 IU/ml) & (1825,34 IU/ml). The result is that the Binahong leaves extract significantly reduce the serum TNF- of the rat which is incuded by 75 ppm formaldehyde compared to the positive control group (3171,34 IU/ml). The second result is that the administration of 25% extract of Binahong leaves is more effective to reduce serum TNF- compared to the 50% extract of Binahong leaves. The third result is that the maximum reduction of serum TNF- were found at thr serum of rat which are treated with 100% extract of Binahong leaves. Formaldehyde administration is the range of (25 ppm, 50 ppm and 75 ppm) might increase the rat serum TNF- , with the 75 ppm as the optimum dose.
