a) Albumin 2% Solubility in various solvents

1. Prepare 4 test tubes, insert 1 ml of 2 % albumin solution in each test tube.


2. Then add in a test tube :
- First test tube : 1 ml of aquades
- Second test tube : 1 ml of 0,2% NaOH solution
- Third test tube : 1 ml of 0,2% HCl solution
- Fourth test tube : 1 ml of 0,2% NaCO3 solution
3. Each test tube vortexed for one and a half minutes , leave for a moment and
observe what happens.
c. Proving the peptide bond constructed protein through biuret test
1. Prepare 3 kinds of protein solution (albumin, potato, soybean bar)
2. Enter the each kinds of protein solution into a test tube , each 3 mL
3. Add 1 mL of 10 % NaOH solution into each test tube containing any type of
protein solution , and then homogenous with vortex.
4. Enter 3 drops of 0.01 M CuSO4 solution , then stir . If it does not arise color , add
1-2 drops of CuSO4.
5. Observe the color change.
d. Proving the amino acid through Ninhydrin test
1. Prepare 1 mL of protein solutions (albumin, potato, soybean bar) and arginin.
2. Putting 1 mL of albumin, 1 mL extract potato, 1 Ml extract soybean bar and 5
drops of arginin in the test tube, add each solution by 5 drops of 0,1% ninhydrin
solution.
3. Heat until boiling , then wait until the each solution cool and observe the color
change.

Analisys Data of Proving The Amino Acid Through Ninhidrin Test :


a) At 5 drops of Arginin in the test tube and then added 5 drops of 0.1% Ninhydrin, the
solution is colorless. After heated in the water until boil, the color of solution changes
to blue (+++) but there is not precipitate. The blue color is indicate that the solution
containing amino acid.
b) At 1 ml of Albumin in the test tube and then added 5 drops of 0.1% Ninhydrin, the
color of solution is clear yellow (+) . After heated in the water until boil, the color of
solution changes to blueish purple (+) and there is precipitate with white color. The
blueish purple is indicate that the solution containing amino acid.
c) At 1 ml of extract soybean bar in the test tube and then added 5 drops of 0.1%
Ninhydrin, the color of solution is white (+) . After heated in the water until boil, the
color of solution changes to blue (++) and there is precipitate with white color. The
blue color is indicate that the solution containing amino acid.
d) At 1 ml of extract potato in the test tube and then added 5 drops of 0.1% Ninhydrin,
the color of solution is brown (+) . After heated in the water until boil, the color of
solution changes to blackish blue (++) and there is precipitate with blue (+) color. The
blackish blue color is indicate that the solution containing amino acid.

Based on the theory the reduction product obtained from ninhydrin then reacts with
NH3 and excess ninhydrin to yield a blue colored substance. This reaction provides an
extremely sensitive test for amino acids. Ninhydrin test is specific for Amino Acids &
Proteins-to differentiate between Carbohydrates (-ve) and Amino Acids & Proteins(+ve).
In the experiment has been done all treatments with different substances after heated there
are discoloration. Initially before heated the color of the solutions is not blue but after
heated all solutions in the different treatments there are discoloration to blue solution. But
the intensity of blue color of each treatments are different.

The higher the intensity of blue color, the higher the amino acids contained and it is
showed that the amino acid chain is length. In these experiment, Arginin + Ninhydrin
0.1% has higher intensity of blue color so these treatment indicate that arginin containing
much of amino acid and has longest chain amino acid than all treatments. Whereas for
Albumin + Ninhydrin 0.1% treatment showed that intensity of blue color is lowest than
all, this indicate that albumin has the amino acid content slightly. In the albumin treatment
there is pink color, indication formed pink, indicate that the chain of amino acid is short
and have little free amino acids.
