Based on the theory the reduction product obtained from ninhydrin then reacts with
NH3 and excess ninhydrin to yield a blue colored substance. This reaction provides an
extremely sensitive test for amino acids. Ninhydrin test is specific for Amino Acids &
Proteins-to differentiate between Carbohydrates (-ve) and Amino Acids & Proteins(+ve).
In the experiment has been done all treatments with different substances after heated there
are discoloration. Initially before heated the color of the solutions is not blue but after
heated all solutions in the different treatments there are discoloration to blue solution. But
the intensity of blue color of each treatments are different.
The higher the intensity of blue color, the higher the amino acids contained and it is
showed that the amino acid chain is length. In these experiment, Arginin + Ninhydrin
0.1% has higher intensity of blue color so these treatment indicate that arginin containing
much of amino acid and has longest chain amino acid than all treatments. Whereas for
Albumin + Ninhydrin 0.1% treatment showed that intensity of blue color is lowest than
all, this indicate that albumin has the amino acid content slightly. In the albumin treatment
there is pink color, indication formed pink, indicate that the chain of amino acid is short
and have little free amino acids.
