Procedure

a) Sample preparation
- Wash the stems and leaves under the running tap water about 5 minutes until it

clean and weight it.


Moisture content analysis was performed by spreading moist butterfly wings
leaf on a metal tray and dried at atmospheric temperature and pressure

Butterfly wings leaf then proceed to the blending by cup of cold water for
extraction of juice.

4.2 SAMPLE EXTRACTION

a) Hydrodistillation extraction
Put 100g of sample with 1.0 L distilled into a Clevenger-type apparatus.
The extraction process was carried out for 3 hours.
For the first hour, the collected extracted essential oil are decented from the

condensate at 5 minute intervals.


Next, the extraction is decanting for every 30 minute.
In order to remove the water, the extracted oil is dried in anhydrous sodium

sulfate pellets.
Store the products in universal bottles.
Analyze the sample using HPLC.

b) Soxhlet extraction
Put 100 g of the samples inside the thimble.
Install the solvent on the hot plate in the BUCHI Extraction System.
Put the tube holder on the thimble.
Let the hot plate heat up for five minutes.
Allow the extraction process to take place for 2 until 3 hours.
Collect the produtcs after the extraction finish.
The products contain mixture of methanol and essential oil.
Separate the mixture by using the rotary evaporator in order to obtain pure

essential oil.
Analyze the sample using HPLC.

Skin Contact Flush with copious amounts of water for at least 15 minutes. Remove contaminated clothing and wash before reuse. Unless

Inhalation Remove to fresh air, rest and keep warm. If breathing is difficult give artificial respiration and obtain medical attention.
Eye contact Irrigate with copious amounts of eye wash or water for at least 15 minutes. Assure adequate flushing by separating the eyelids
with fingers. Obtain medical attention
Ingestion Do not induce vomiting. If conscious provide water for person to thoroughly wash out mouth (and sip if required). Obtain medical
attention.

Full safety data sheets for the above chemicals must available in file at the laboratory in use.

7a. Specific risk assessment for procedure and precautions to be


taken

Full PPE equipment must be worn everytime handling an experiment and in the laboratory.
A spillage mighy occur, so to avoid any clothing, eye and skin contcact with the chemical
and avoid heavy inhalation in the area as well.
Wear glove and mask if necessary, while rhandling acid and alkaline.
Have to handle with care any fragile glassware .
Wash hand before leave the laboratory.
A full alert need to be considered while handling hot equipment. Wear glove to avoid
burning.
Read manual, labels and tags before use any electrical equipment

After following all the above precautions the overall risk assessment for this procedure is LOW.
