Effect of L. reuteri on the density of enteric bacterial flora SEE FIG. 2
- Focused on the influence of L. reuteri on the density of fecal lactic acid
bacteria and non-specific bacteria.
- Before OVA challenge, density of fecal LAB and NSB was comparable between
the OVA-sensitized and non-sensitized mice.
- Administration of L. reuteri on both mice resulted in the increase of LAB
density but not in NSB.
- After 5th OVA challenge, allergic mice exhibited diminished density of LAB and
increased density of NSB. This is supported by previous findings on
deteriorated enteric flora in sensitized mice.
- These observed alterations were restored with the administration of L.
reuteri.
- The administration of the bacteria significantly increased the density of LAB
and decreased the density of NSB.
Effect of L. reuteri on allergic responses SEE FIG.3
- Successful induction of allergic diarrhea was observed in OVA-sensitized mice
from the 3rd OVA Challenge
- Administration of L. reuteri significantly reduced occurrence of allergic
diarrhea in affected mice from 3rd OVA challenge.
- The infiltration and degranulation of mast cells were also attenuated, as well
as the total and IgE specific serum production.
Effect of L. reuteri on the immune profile of Th cell subsets SEE FIG 4
- How do T-cells play a pivotal role in the immunopathology of food allergies?
- T-cell derived cytokines IFN-y, IL-4, IL-10, TGF-B and transcription factors
Foxp3, GATA-3 and T-bet was observed to be increased in the OVA group, in
comparison to the control group, showing enhanced T-Cell activation in OVA
Sensitized and Challenged Mice.
- L. reuteri down-regulated production of IL-4, a signature cytokine of Th-2.
- Expression of Th1 cytokine IFN-y by splenocytes is also attenuated in bacteria
treated mouse.
- Expression of IFN-y and IL-4 mRNA was down-regulated in the bacteria
treated mouse.
- Expression of IL-10 and TGF-B was upregulated
- Suppressed expression of T-bet an GATA-3 but increased expression of Foxp3.
Effects of L. reuteri on CD11c+ and CD103+ MLN cells see fig 5
- Percentage of CD11c+ and CD103+ cell population significantly increased in
MLNs of mice treated with L. reuteri.
- Production of IL-10 by CD11c+ CD103+ cell was markedly higher than that of
CD11c- CD103- cells.
Supplement with heat-killed L. reuteri prevented the proliferation and
activation of OVA-primed MLN T cells re-stimulated with OVA
- Isolated MLN Cells were directly exposed to heat-killed L. reuteri in culture,
and then re-stimulated with 100uG/mL OVA to induce cell proliferation and
cytokine expression
- Stimulation index and production of IL-2, a cytokine produced by activated T
cells, was attenuated by concentrations greater than 50ug/mL of heat-killed
L. reuteri.
- Direct exposure attenuated the proliferation of and activation of MLN T cells.
4. Discussion

Results from the present study confirm our hypothesis that L.
