ABSTRACT

A research on the effects of test antihyperglycemic drug combination herbal ant


nests ( Myrmecodia pendens Merr . & Perry ) and Virgin Coconut Oil ( VCO ) in male
Wistar rats has conducted . The purpose of this research is to figure out the effect of
the combination of ant nests ( Myrmecodia pendens Merr . & Perry ) and Virgin
Coconut Oil ( VCO ) on antihyperglycemic efficacy in lowering blood glucose levels
of male white rats ( Rattus norvegicus ) of Wistar strain .

The method used in this study was the oral glucose tolerance test with glucose
loading dose of 4.5 g / kg . Test animals used were male Wistar rats aged 2-3
months with 200-300 grams weight . as many as 25 rats which were divided into 5
groups , each group consisted of 5 rats . Group I as a negative control group was
given Na.CMC 1 % , group II as a positive control group was given a dose of the drug
glibenklamide 0.45 g / kg , group III as the first treatment group was given
combination dose of ants nest 37.8 mg / kg ; and VCO 2.03 ml / kg , group IV as a
treatment group II was given combination dose of ants nest 75.6 mg / kg ; and VCO
4.05 ml / kg , and group V as a third treatment group was given combination dose of
ants nest 151 , 2 mg / kg ; and VCO 8.1 ml / kg , 30 minutes later all treatment
groups were given a glucose 4.5 g / kg dose. When treatment was considered as
time 0, glucose is considered as the 30th time . Blood sampling through the whole
rats were taken at 0, 30 , 60 , 120 , 180 , 240 and 300 .

Blood glucose levels were measured by means of EasyTouch Blood Glucose Test
Meters * GCU . Blood glucose lowering effect demonstrated by calculating the value
LDDK0 - 300 ( Regional Area Under the Curve minute 0 to minute 300 of the graph
of time vs. blood glucose levels ) . The data obtained were statistically tested by
One -way ANOVA and LSD with 95% significance level . The results showed that
administration of a combination preparation ants nest dose 75.6 mg / kg and a VCO
of 4.05 ml / kg perfomed as the best in lowering blood glucose levels on mice
compared with other treatment groups in the amount of 31.43 % , but could not
give better result than with the commission of the positive control group
glibenclamide 0.45 mg / kg with percentage of perfomance 53.93 % .
