CHAPTER III

RESULTS

All microorganisms isolated samples were collected from San Pedro College Marine

Station which are sea sand and sea water.

Microorganisms found in sea water and sea sand

In this research, the common microorganisms were identified mainly on bacteria. Among

the sites, microorganisms found in sea water had the highest bacteria isolated.

Colony forming unit

Sample Dillution factor Colony count
(CFU)
Sea sand 3.36E-03 64 2.15E+00/ml
Sea water 3.23E-02 107 3.45E+01/ml

Table 1: Isolated bacteria from sea water and sea sand.

Table 1 showed the most isolated bacteria from different dilutions (3.23E-02 - 3.63E-05)

with their corresponding CFUs. By means, an average of 16 isolated bacteria from sea sand and

27 isolated bacteria from sea water.

Isolation of microorganisms using 3 different medium agars.

Among the isolated samples using Nutrient Agar (NA); Muller-Hinton (MH) Agar; and

Inorganic Salt Starch (ISP4) Agar, the most isolated bacteria were cultivated using the nutrient

medium agar. Bacteria colony count of isolated microorganisms from two samples showed

(Table 2) the highest number of bacteria load was at the sea water followed by sea sand.

Medium agar Sea Sand (CFU/ml) Sea Water (CFU/ml)

Nutrient Agar 2.95E+00/ml or TFTC 4.98E+00/ml
Muller-Hinton Agar 2.79E+00/ml 3.28E+00/ml

Inorganic Salt Starch Agar 2.27E-02/ml of TFTC None

TFTC = Too few to count

Light Absorbance of inocula against E. coli and Staphylococcus aerues

The chosen 7 bacteria isolated from the sites were inoculated to E. coli. The gathered

absorbance of each inoculum were compared to the control. In which the control tube is pure E.

coli. Presented on Figures 1 and 2 are the trials of inocula and their corresponding absorbance.

Turbidity of inocula against E. coli and Staphylococcus aerues

A. for E. coli turbidity (whether there is significant difference among wavelengths absorbance

using ANOVA test).

p-value (at alpha =0.05) Hypothesis Remark

0.096691 Accepted Not Significant

Table 3: Turbidity of inocula against E. coli.

Result showed that the wavelength absorbance on E.coli as far as the control and two

trials were concerned showed no significant difference at all. It simply means all are comparable.

Thus, hypothesis is accepted.

B. for Staphylococcus aerues turbidity (whether there is significant difference among

wavelengths absorbance using ANOVA test).

p-value (at alpha =0.05) Hypothesis Remark

0.12109 Accepted Not Significant
Table 4: Turbidity of inocula against Staphylococcus aerues

Result showed that the wavelength absorbance on Staphylococcus aerues as far as the

control and two trials were concerned showed no significant difference at all. It simply means all

are comparable. Thus, hypothesis is accepted.

C. Comparing both E. coli and Staphylococcus aerues on turbidity (whether there is significant

difference among wavelengths absorbance using t - test).

p-value (at alpha =0.05) Hypothesis Remark
P(T<=t) one- Accepted Not Significant

P(T<=t) two-tail 0.393456938
Table 5: Comparison of inocula against E. coli and Staphylococcus aerues

Result showed that the wavelength absorbance on both E. coli and Staphylococcus

aerues as far as the control and two trials were concerned showed no significant difference at all.

It simply means that both are comparable as far as wavelength absorbance is concerned. Thus,

hypothesis is accepted.
