RELTIF EXPRESSION PROFILE OF MRNA EBNA1 EBSTEIN-BARR VIRUS FROM

FORMALIN-FIXED PARAFFIN-EMBEDDED AS POTENTIAL MOLECULAR


BIOLOGY MARKER TO THE PATHOGENESIS OF NASOPHARYNGEAL
CARCINOMA

Background : Nasopharyngeal carcinoma is a malignant tumor that caused by the infection


of Epstein-Barr virus. In the primary stage of infection the IgG Antibody and IgA anti EBV
EBNA1 titer of the suspect will increase. The activity of mRNA EBV is more representatif as a
pathogenesis marker than serological diagnosis and the EBV DNA measuring in the
cirulation. The measuring of mRNA EBV activity needs to be done from the location of the
primary tumor in the nasopharynx because this gen is expressed in every cycle of EBV
Infection. Goals : To figure out the value of median relatif expression of mRNA EBNA1 from
early stage and late stage of Nasopharyngeal carcinoma patient. Methods :Observasional
analytic with crossectional approach and bivariat analysis. RNA isolation, cDNA sinthesis,
and mRNA EBNA1 quantity expression was done by RT-qPCR from 22 sample of 2014. With
simple random sampling method for the sampling. Mann-Whitney statistical method was
perfomed and boxplot is used for the data presentation. Result : Median value of early stage
cancer is 29.226,71 and for the late stage is 32.429,07. Mann Whitney method show
p=0,249. Conclusion : there is difference of relatif expression of mRNA EBNA1 EBV of
nasipharyngeal carcinoma from FFPE biopsy in the late stage diagnosis by 1.1 times than
the early stage Nasopharyngeal cancer. Statistical method show that relatif expression of
mRNA EBNA1 is not potential as a moluclar biology marker for the pathogensis of
nasopharyngeal carcinoma, especially for the progressivity of the late stage tumor.
