Introduction

General
This document describes the configuration of the Varian ProStar
520 driver. It also explains how to build a control method for this
driver, how to start and view a running acquisition and what
actions are available during an analysis.

The ProStar 520 Column Oven controls the temperature of the


mobile phase and column for HPLC separation on a Helix
system. The column oven controls the temperature of all the
components inserted in the oven (autosampler injector, pre-
saturator column, analytical column, etc.).

By keeping all of these components at the same precise


temperature, more reproducible chromatography is obtained.

The oven has a vapor sensor to detect any organic solvents in


the oven. It will automatically shut down the oven heater if there
is a leak from any of the tubing.

Communications

Ethernet Communications
The ProStar 520 Column Oven uses Ethernet as the method of
communication for instrument control.

All signals are connected through the rear panel of the oven.
Only the J10 connector is used to connect the oven to the
Galaxie Workstation computer. The other connections are not
used.

The column oven is connected using either an Ethernet


crossover cable or a standard Ethernet cable.

Connectors for Peripheral Equipment

The ProStar 520 uses TCP/IP (Transmission Control Protocol /


Internet Protocol) to communicate over the Ethernet network and
needs to be given a unique IP address to identify the instrument.

A BOOTP Server is used to send IP addresses to TCP/IP


devices.

The BOOTP Server lists Ethernet addresses along with the IP


addresses that are to be assigned to the corresponding devices.

To known how to configure the BOOTP server, refer to the


Galaxie installation Guide.

NOTE: The BOOTP server provided by the Galaxie Chromatography Data System is
installed during the installation of the Galaxie software.

Configuring the Driver
The driver software configuration is done in the Galaxie
Configuration Manager when a new chromatographic system is
created.

Configuring the Column Oven ProStar 520

Click on the button corresponding to the Varian ProStar


520 driver and the configuration screen appears.

Four sub-sections allow you to configure the instrument:


Communication, Column Oven, Vapor Sensor, and
Miscellaneous.

The toolbar allows you to easily scroll through the configuration


windows:

to select the first configuration window.

to select the previous the configuration window.

to select the next configuration window.

to select the final configuration window.

to send the configuration to the instrument.

to get the configuration from the instrument.

Communication Window

Enter the IP address of the ProStar 520 Column Oven (declared


in the BOOTP Server) in the GC name or address field or click
on the button to find the instrument previously configured
with the BOOTP Server program and select it.

Press the button to automatically
get the configuration from the instrument.

Column Oven Window

The temperature of the oven can be calibrated to 0.1C in the


lab. Calibrating the temperature has three basic uses:
If you want to adjust the temperature to the exactly
correct temperature, you can calibrate the oven
using a high accuracy temperature measuring
device.

If you want to adjust the temperature set in and read


from the oven based on results for a particular set of
compounds published in the scientific literature.

If you want to adjust the temperature of many


different ovens in the same lab to the same
temperature.

To calibrate the oven, use the following steps:

1. Run one or more standards with setting the temperature to


several values in the range where the separation should
occur (this can be in 0.1 or 0.2 C increments).

2. By looking at the chromatograms, decide what the actual


temperature should be for a particular separation.

3. Select the Column Oven sub-section and choose the


Constant Correction option from the list, if you want to
perform the calibration at one point or select the Linear
Correction option, if you want to perform the calibration at
two points.

4. Enter the temperature that you set in the column oven under
the ProStar 520 heading (Temperature data point 1 or 2
entry).

5. Enter the temperature at which this separation should occur


in the Calibration heading (Temperature data point 1 or 2
entry).

If you are going to run only one set of samples at one


temperature, use the single point calibration (Constant
Correction calibration mode). If you are going to run sample at
several temperatures, you can use a two point calibration. For
this, run two different standards at the two bounds of the
temperature range of interest.

The calibration model is a linear curve with the equation:

Calibrated Temperature = (Oven Temperature x a) + b

Where the coefficients, a and b, are derived from two point


calibration; if a one point calibration was done, a is equal to 1
and b is the only coefficient used.

Vapor Sensor Window

The vapor sensor is located in the column oven. It is designed to


detect leaks of organic chemicals such as acetonitrile in the
mobile phase in the column oven. If it detects a leak, it will
automatically turn off the column heaters, open the column oven
vent, and keep the vent fan running. This will help to clear the
oven of any solvent.

The vapor sensor only responds to organic chemicals. It does


not respond to water or buffer. If there is a water only leak in the
column oven, no alarm will be triggered.

The vapor alarm can trigger three actions besides shutting down
the oven. It can shut down the pumps, turn off the lamps and
sound an audible alarm in the PC which displays the status of
the ProStar 520 Column Oven. Check the required options in the
Vapor Alarm Actions fields.

Miscellaneous Window

The Ready in Contact Closed option allows you to specify what


switch state corresponds to external device Ready. Check the
option if the closed state means Ready.

Configuring the Synopsis

In the Overview part of the system configuration, a synopsis can


be defined for the system. This synopsis depends on the
configured devices.

1. To save the configuration of these devices, click on the OK


button.

2. Right click on the system name in the bottom part of the


Galaxie Configuration Manager screen. A pop-up menu
appears: click on Stop. Then right click on the system name
again, and click on Start.

3. Go back to the System Configuration screen (Properties
wizard). A graphic of the devices that comprise the system is
displayed in the synopsis screen. To configure the synopsis
as the real instrument configuration, move each module,
then connect it appropriately. To do this, select one anchor
for each of the two modules (the selected anchors become

red) and click on the button ; a connection tube will


appear between the two anchors.

Click on the button to delete a selected tube, click on

the button to delete all the tubes.

Click on to define a background color, or on to


make the background transparent.
Click on the icon to change the grid color.

4. Once the synopsis configuration is complete, it will be


displayed in the Galaxie Chromatography Data System, in
the Systems tab. In this tab, the Overview will show the
synopsis of the system with some parameters displayed
(temperature, flow, signal, etc.) depending on the installed
devices.

For example, the following pictures represent the synopsis of


the ProStar 520 Column oven:

NOTE: Once the system has been created and correctly configured, it must be
associated with one or more projects in order to start acquisitions. For more
details about the creation and configuration of a system, refer to the Galaxie
Configuration Manager Users Guide.

Building a Method

Creating a New Method


The method contains parameters for instrument control, data
acquisition, chromatogram processing, and editing of final
results. To initially set up a method, it is not necessary to define
all of the sub-method sections, however, you must fill in the
instrument control section prior to starting an acquisition.

1. To create a new method, select the FILE / NEW METHOD


option from the Galaxie Chromatography Data System main
menu.

A wizard guide will appear and provide assistance during the


first steps of method creation:

2. Choose the system (chromatograph) associated with the
method. A method is created for a particular system. When
starting an acquisition, the name of the system that performs
the acquisition must be correct, since method access is
limited only to those methods associated with this system.

3. Once this field is completed, click on the Next button to


move to the second step of the method creation:

4. Enter the New Method Name in the first field. In the
Description field, enter information concerning the method
(optional).

5. Once these two fields have been completed, click on the OK


button. The new method is created and is opened with
default parameters in the Galaxie Chromatography Data
System. The next step is the definition of each of the method
sections.

The control section must be defined in order to start an


acquisition. In the following part of this guide, only the control
section for the ProStar 520 Column Oven is detailed. Other
parameters such as Injection Volume, Vial and Rack number
may be necessary and are defined in the acquisition section.

Once the new method is created, select the control section in the
lower part of the browser.

The following screen appears:

Click on the button to display the ProStar 520 Column


Oven control method section. It contains two sub-sections: Oven
and Miscellaneous. Click on the required sub-section to define
the corresponding parameters that will be used during the
chromatographic run.

The toolbar allows you to easily scroll through the control method
windows:

to select the first window of the control method.

to move back to the previous window of the control


method.

to move forward to the next window of the control


method.

to select the final window of the control method.

Oven Control

General Parameter Group:

Enter the Stabilization time in the corresponding field.

Column Oven Program Parameter Group:

Check (or uncheck) the Heater option to turn it ON (or OFF).

Use the table to build a temperature ramp program to heat


and/or cool the column oven.

The first row of the table contains the initial temperature and hold
time for the column oven. The column oven will equilibrate to this
setting and stabilize for the specified Stabilization time (defined
in the General field), when the method is activated, and will be
restored to this setting when each chromatographic run is

completed. The rest of the table contains the programmed
settings for each of the ramp segments. Enter the final
temperature of each ramp segment in the Step column, the Rate
(C/min) and the Time the column oven is held at this
temperature in the corresponding column.

Use the following buttons to edit the table:

to add a line to the table.

to delete a line in the table.

to clear the entire table.

The graph shows the column oven temperature program.

NOTE: The browser always displays the current end time of the column oven program.

Miscellaneous Control

Driver Behavior Parameter Group:

Check or uncheck the Reset oven program at the end of the


acquisition to activate or deactivate this option. If it is checked
and the acquisition is stopped from the acquisition window, the
oven temperature program will be reset before continuing with
the next injection (if a sequence is running).

Check or uncheck the Stop oven at the end of the acquisition to


activate or deactivate this option.

Starting and Viewing an
Acquisition

Quick Start and Sequence


Once the instrument control method has been correctly
programmed, an analysis can be started. The next section briefly
describes how to start an acquisition.

Quick Start (Single Acquisition)


1. Press the button.

In the Quick Start window, select the system, the method


and optionally the project if connected in "All project" mode.

2. Press the OK button.

The following screen is displayed:

In the Acquisition Parameters tab, required fields like the


Run Name, the Run ID and the Acquisition length must be
filled before starting an analysis. Furthermore, if the system
is equipped with an autosampler controlled by the Galaxie
Chromatography Data System, the Injection Volume, the Vial
position and the Rack number (if required) must be entered.

Other parameters associated with each acquisition channel


can be entered in the Channel X tab.

3. Click on the Start button to start the acquisition if the system


is a single-injector system or click on the Next button if the
system is a multiple injector system. In a multiple injector
system the same parameters entered for the first injector
must be entered for the other injectors before starting the
acquisition.

Sequence
1. To create a sequence, select FILE / NEW SEQUENCE
menu, and complete the displayed creation wizard screen
(refer to the Galaxie Chromatography Data System Users
Guide), by entering the system name that will perform the
analysis, the number of lines (corresponding to the number
of samples), and the sequence name.

2. The sequence is displayed. Complete the sequence


information columns (refer to the Galaxie Chromatography
Data System Users Guide).

The Method Name, the Run Name, the Run ID, and the
Acquisition length must be filled before starting the analysis.
All these parameters are automatically imported from the
acquisition part if defined in the method.

3. Start the sequence by clicking on the button.

Running Acquisition Status

Viewing a Running Acquisition


To view a running acquisition, click on the Systems tab then
select the running system. The running chromatogram and the
status window where the driver modules are listed.

Click on the button corresponding to the ProStar 520


Column Oven driver and the ProStar 520 status screen will
appear. It contains four sub-sections: Oven, Vapor Sensor,
Miscellaneous, and Overview. Click on the required sub-section
to display the corresponding parameters that will be used during
the chromatographic run.

A toolbar allows you to easily scroll through the status windows:

to select the first status window.

to move back to the previous status window.

to move forward to the next status window.

to select the final status window.

Oven Window

The status display includes the following parameters: :

Ready or Not Ready: the column oven is Ready if the setpoints


have been achieved and stabilized. The column oven is Not
Ready if readiness to start a run has not been achieved or if,
during a run, the oven becomes not ready or goes out of
tolerance.

Fault or No Fault: displays if a fault in the column oven


component has occurred.

Heater: displays if the column oven component is turned OFF or


ON at the ProStar 520.

Actual: displays the actual temperature of the column oven at the


ProStar 520.

Setpoint: displays the column oven programmed temperature


setting in the active method.

Stabilization State: displays the current stabilization state of the


column oven. When the state is ON, the
button becomes active. Click on it to bypass the stabilization
time.

Stabilization Time: shows the ProStar 520 Column Oven


stabilization time in minutes. The stabilization time counts down
from the programmed method value and then displays 0.00
minutes when the stabilization period is over.

Click on to turn off the oven when the oven
is on.

Click on to turn on the oven when the oven


is off.

Vapor Sensor Window

Ready or Not Ready: indicates whether the vapor sensor is OK


or not.

Fault or No Fault: Displays if a fault in the vapor sensor


component has occurred (when it has detected vapor in the
oven).

Click on to turn off the vapor sensor alarm.

NOTE: If a vapor sensor alarm or failure occurs, it can only be cleared by turning the
oven off and turning it back on. If the cause of the alarm was a spill in the oven,
the oven should be carefully cleaned of all spilled material and the vapors should
be allowed to dissipate. This is aided by the fan continuing to run when there is
an alarm. If the alarm turns on after it has been restarted, the procedure should
be repeated. Note, if the atmosphere in the laboratory is so full of vapor that the
alarm continues to go on, the instrument should be moved to a safer
environment.

If the vapor alarm has been triggered because of a failure of the vapor sensor,
the instrument should be turned off and back on.

Miscellaneous Window

The miscellaneous screen contains general information about


the current run and the overall state of the instrument.

The state of the instrument (Ready, Running, Stabilizing,

Equilibrating, or Not Ready) is displayed.

A Fault / No Fault indicator is also shown. When a hazardous or


disabling fault occurs, any run in progress will halt and the
affected component will shut down. If the fault is recoverable,
then the run is not terminated.

The run time bar displays the elapsed time in minutes since the
beginning of the run and the end time.

Line Voltage: displays the voltage setting for the ProStar 520
Column Oven.

Firmware Version: displays the version of the firmware installed


in the ProStar 520 Column Oven.

Press the button, to reset the column


oven. If it is RUNNING, the oven aborts the current run, goes to
the READY state and continues with the next injection in the
sequence.

Overview Window

Click on the button. The Status Overview window is


displayed and contains three tabs:

General: displays the synopsis of the instrument


where some current values can be read (oven
status, column oven temperature, etc.).

Not Ready: contains the not ready information sent


by the instrument.

Events: contains the events that have occurred on


the instrument.

TROUBLESHOOTING:

If the status is reporting that the vapor sensor is giving a fault when there is no
discernable liquid in the oven, check the message carefully. The vapor sensor will report
whether it has failed or whether it can no longer read vapor.

If the vapor sensor reports vapor in the oven when there is no liquid, it is possible that a
previous leak in the oven has saturated the insulation. If that is the case, continue to run
the oven with the fan on (the heaters will be off) and wait until the fault disappears. If the
fault does not go away within a reasonable length of time (1 day), it is possible that the
atmosphere in the lab has a too high saturation of organic vapor. If this is the case, move
the system to an area where the concentration of organic vapor is known to be low and
run it again.
