Diagnosis of dengue fever


By means of a cell count of platelets
Dengue Fever is an infectious disease Dengue virus is transmitted by Aedes aegypti and
Aedes albopictus. Dengue virus belonging to the genus Flavivirus, family Flaviviridae, which
are divided into four serotypes DEN 1, DEN 2, DEN 3 and DEN virus serotypes 4. Fourth,
there are in Indonesia and reported that serotype DEN 3 virus often causes outbreaks, while
in Thailand the cause of the outbreak DEN is the dominant virus 2. the disease is indicated by
a sudden fever 2-7 days, accompanied by severe headache, pain in joints and muscles
(myalgia and arthralgia) and a bright red rash, petechie and usually appears first on the lower
part body spreads to cover most of the body. Stomach ulcers can also appear with a
combination of associated abdominal pain, nausea, vomiting or diarrhea.

1. To determine the number of platelets in the blood cells probandus
2. To determine whether the patient had dengue fever
B. Principle
Blood was diluted using a solution containing the brilliant cresyl blue who
would paint platelets become somewhat colored light blue. then calculated using the
platelet count room
C. tools and materials
1. tools
a) Thoma pipette erythrocytes
b) room count
c) deck glass
d) microscope
e) tube vial
f) 3cc syringe
g) Aspirator

2. Materials
a) Blood + EDTA
b) hayem solution (sodium citrate 3.8 gram ,formaldehyde solution 40% ,
brilliant cresyl blue 30 gram ,aquadest 100 ml
D procedure
a) equipment and materials to be used.
b) do pemipetandarah using a pipette Thoma erytrosit to right on a 0.5 scale and
clean the outside of the pipette with a tissue.
c) Ecker rees continue pipetting solution up to a scale of 101 (do it carefully to
avoid bubbles).
d) homogenize the solution for 15-30 seconds, discard the solution of 3-4 drops.
e) dripping the solution on the surface of the counting chamber were already in
the cover glass deck until blended, on its own capacity.
f) let the counting chamber for 2-3 minutes to allow platelets to settle, then
continue to read the number of platelet cells under a microscope with a
magnification of 40X.

2) patients affected by dengue fever.
