A. Pendahuluan
Bisphenol A (2,2-bis(4-hydroxyphenyl)propane) atau yang biasa dikenal dengan BPA
merupakan salah satu senyawa kimia yang diproduksi dalam skala besar diseluruh dunia dan
memiliki pasar di berbagai produk. Di Negara-negara Eropa produksi BPA sudah melebihi
500,000 ton, sedangkan didunia telah mencapai lebih dari 1 juta ton.
Pada tahun 1936, para ilmuwan menemukan bahwa bisphenol A(BPA) dapat digunakan
sebagai hormon buatan yang menyerupai estrogen, yang dapat digunakan dalam mengatasi
kesulitan kehamilan. Kemudian bisphenol A diketahui memiliki persamaan dengan senyawa
kimia yang disebut diethyl sylbestral (DES). Hingga akhirnya diketahui berbahaya setelah
diketahui DES dapat menyebabkan kanker dan masalah terkait reproduksi (Colborn et
al.1997)Sekitar tahun 1950 para ahli kimia menemukan bahwa BPA dapat dipolimerisasi
menjadi bahan plastik. Saat ini BPA digunakan untuk membuat plastik polikarbonat, suatu
bahan plastik yang bening dan kuat yang banyak digunakan dalam pembuatan botol bayi
merk-merk terkenal. Penggunaan bisphenol sudah cukup luas, bukan hanya untuk kemasan
pangan juga dapat digunakan sebagai bahan penambal gigi.
Sebagian besar BPA digunakan sebagai bahan baku pembuatan plastik polikarbonat (PC) dan
epoxy resin. Plastik Polycarbonat yang diproduksi dengan menggunakan BPA memiliki
beberapa keunggulan, diantaranya memiliki tingkat kecerahan plastik yang baik, lebih kuat,
dan tahan terhadap benturan, serta mudah dibentuk pada suhu ruang. Hal ini membuat
penggunaan plastik polikarbonat lebih disukai untuk diaplikasikan pada berbagai produk
plastik seperti kemasan makanan, botol susu, botol air, bahkan pipa-pipa saluran air.
Sedangkan epoxy resin sering digunakan sebagai bahan pelapis logam seperti kaleng
makanan dan saluran air (Aschberger, 2010).
Pada kemasan pangan, plastik polikarbonat ditandai dengan angka 7 di dalam segitiga daur
ulang yang biasanya terdapat di bagian bawah botol, atau ditandai dengan huruf ‘PC’ dekat
segitiga daur ulang. Tidak hanya pada botol bayi, BPA juga biasa digunakan sebagai
kemasan makanan dan minuman dan botol air, bahkan dapat juga digunakan dalam
pembuatan kepingan CD/DVD atau kacamata dan penambal gigi.
C. Kontroversi BPA
Pada awal tahun 2007, melaui data dari European Food Safety Authority (EFSA) dapat
disimpulkan bahwa BPA memiliki NOAEL sebesar 5 mg /kg bb/hari yang diperoleh dari
penelitian terhadap tiga generasi tikus. Melalui penelitian tersebut ditentukan TDI dari BPA
sebesar 0,05 mg BPA/kg bb. EFSA juga meyakinkan bahwa jumlah paparan pada konsumen
di negaranya, termasuk didalamnya bayi pengguna botol PC, tidak akan terpapar dalam
jumlah yang membahayakan, yaitu kurang dari 30% TDI. Pada bayi, BPA diperkirakan
berasal dari susu formula, yakni dari migrasi botol susu, sedangkan untuk orang dewasa dapat
berasal dari makanan kaleng. Sedangkan di Amerika, melalui hasil penelitian yang dilakukan
FDA diperoleh data paparan BPA pada bayi sebesar 2,42 μg/kg bb/hari, sedangkan pada
orang dewasa sebesar 0,185 μg/kg bb/hari.
Penelitian terkait BPA telah dilakukan lebih dari 10 tahun oleh beberapa lembaga
pemerintahan terkait kesehatan yang berbeda di Eropa, Kanada, USA dan Jepang. Pengujian
dilakukan berdasarkan penelitian yang telah sesuai dengan international guidelines dan Good
Laboratory Practices, menggunakan pengujian secara oral, serta dilakukan pada berbagai
jenis hewan dengan range dosis yang berbeda. Berdasarkan data dari EFSA (2008), ECB
(2008), Food and Drug Administration of the United States of America (US-FDA; 2008) dan
the Japanese National Institute of Advanced Industrial Science and Technology (AIST, 2007)
tidak ditemukan kondisi yang dapat membahayakan manusia.
Sebaliknya, kepedulian terhadap efek dosis rendah ini diungkapkan oleh para ahli dari
pertemuan Chapel Hill pada “Bisphenol A: An Examination of the Relevance of Ecological,
In vitro and Laboratory Animal Studies for Assessing Risks to Human Health” (Vom Saal et
al., 2007), the US National Toxicology Program (Chapin et al., 2008), the Scientific
Committee of the US FDA (2008), beberapa anggota the European Union, dan Kanada
(2008). Keprihatinan ini terutama diungkapkan dalam kaitannya dengan efek neurobehavioral
pada dosis rendah BPA. Oleh karena itu, beberapa negara mempertimbangkan atau melarang
penggunaan BPA terutama dalam kontainer untuk botol bayi dan makanan bayi, seperti
Kanada, Perancis, Denmark dan beberapa dari Amerika Serikat.
Saat ini banyak organisasi melihat potensi resiko kesehatan yang disebakan oleh BPA. Pusat
Riset Toksikologi Nasional FDA saat ini melakukan kajian yang mendalam dalam kerjasama
dengan National Toxicology Program (NTP), untuk mengklarifikasi dugaan tersebut.
Sementara itu, US-FDA mengambil langkah-langkah pencegahan untuk mengurangi paparan
BPA dalam penyediaan makanan dengan misalnya mendorong industri untuk berhenti
memproduksi botol bayi yang mengandung BPA dan cangkir pemberian makan bayi untuk
pasar AS, memfasilitasi pengembangan alternatif untuk BPA dan mendukung upaya untuk
mengganti atau meminimalkan tingkat BPA dalam pelapis kaleng makanan.
Pada January 2010, the French Food Safety Agency (AFSSA, 2010), menyatakan
pendapatnya yang dilanjutkan dengan rekomendasi terkait BPA, yaitu "gejala yang
membahayakan akibat pengaruh BPA dapat diamati melalui pengujian secara in-vitro dan in-
vivo dengan dosis yang lebih kecil dari NOAEL sebesar 5 mg/kg bb/hari dengan kondisi
kesehatan yang telah ditentukan. Kemungkinan ditemukann sumber bisphenol A yang lain
dari material yang kontak langsung dengan makanan. Hal ini dapat memberikan dampak
yang berarti pada penggunaan polikarbonate sebagai bahan pembuat botol bayi serta produk
lainnya di USA dan juga di Eropa. Material ini sepertipolyether sulphone (PES),
polypropylene (PP), pure silicone, polyamide dan co-polyester polymer yang baru.

An international group of researchers led by the Peninsula Medical School and the University of
Exeter have for the first time identified changes in sex hormones associated with exposure to
bisphenol A (BPA) in men, in a large population study.
The study results are published in the latest issue of Environmental Health Perspectives.
Researchers analysed data from the InCHIANTI study, an Italian population sample. The team
measured the amount of BPA excreted per day in urine samples. 715 adults aged between 20 and 74
years were studied. The study aimed to measure the daily BPA loads excreted by adults, and to
examine statistical associations between the amount of BPA exposure and serum estrogen and
testosterone concentrations.
The average BPA daily exposure level in this European study population (over 5 micrograms per day)
was slightly higher than recent comparable estimates for the USA population. The study found that
higher BPA exposure was statistically associated with endocrine changes in men, specifically small
increases in levels of testosterone in the blood.
BPA has a similar molecular structure to estrogen and does cause some disruption of sex hormone
signalling in laboratory animals, but this is the first large human study to suggest that it may have
similar effects in adults at 'background' exposure levels.
Professor David Melzer, Professor of Epidemiology and Public Health at the Peninsula Medical
School (Exeter, UK), commented: "This is the first big study of BPA from a European country and
confirms that 'routine' exposures in the population are not negligible. It also shows that higher
exposure to BPA is statistically associated with modest changes in levels of testosterone in men. This
finding is consistent with the evidence from laboratory experiments. However, this is just the first
step in proving that at 'ordinary' exposure levels, BPA might be active in the human body. This new
evidence does justify proper human safety studies to clarify the effects of BPA in people."
BPA is a controversial chemical commonly used in food and drink containers. It has previously
caused concerns over health risks to babies, as it is present in some baby's bottles. Several nations
have moved to ban BPA from the manufacture of baby's bottles and other feeding equipment. The
US Food and Drug administration has committed itself to reducing BPA residues in food. BPA has
never undergone formal human safety studies: nearly all the safety experiments have been on
laboratory mice or rats, and these may be misleading as BPA is metabolised differently in the human
body.
The main source of BPA in people is thought to be from residues in food, leaching out of certain
types of polycarbonate and resin packaging. The new study therefore used statistical models
adjusted for factors including measures of obesity, to exclude misleading associations in people who
consume more food.
BPA has also been associated with thyroid hormone disruption, altered pancreatic beta-cell function,
cardiovascular disease and obesity.
BPA is used in polycarbonate plastic products such as refillable drinks containers, some plastic eating
utensils and many other products in everyday use. It is one of the world's highest production volume
chemicals, with over 2.2 million tonnes (6.4 billion pounds) produced annually. It is detectable in the
bodies of more than 90% of the population.

Abstract
Concern is mounting regarding the human health and environmental effects of bisphenol A (BPA), a
high-production-volume chemical used in synthesis of plastics. We have reviewed the growing
literature on effects of low doses of BPA, below 50 mg/(kg day), in laboratory exposures with
mammalian model organisms. Many, but not all, effects of BPA are similar to effects seen in
response to the model estrogens diethylstilbestrol and ethinylestradiol. For most effects, the
potency of BPA is approximately 10–1000-fold less than that of diethylstilbestrol or ethinylestradiol.
Based on our review of the literature, a consensus was reached regarding our level of confidence
that particular outcomes occur in response to low dose BPA exposure. We are confident that adult
exposure to BPA affects the male reproductive tract, and that long lasting, organizational effects in
response to developmental exposure to BPA occur in the brain, the male reproductive system, and
metabolic processes. We consider it likely, but requiring further confirmation, that adult exposure to
BPA affects the brain, the female reproductive system, and the immune system, and that
developmental effects occur in the female reproductive system.
