1.2 Proved Security

This device has been constructed under the regulations of EN 61010-1 and
EN 61010-2-101.

This device was examined and left the factory in perfect technical condition. To preserve this and
protect safe and faultless operation, the user must follow the instructions and remarks of this
service manual.

To preserve this condition and to guarantee a safe working, information and working advices,
which are included in these working instructions, have to be heeded by the applier.

This device meets protection 1 (earth conductor wire).

1.3 Precaution and Hazards

All electrical equipment is potentially hazardous. Never remove covers without first ensuring that it
is isolated from the AC supply, unless specific maintenance instructions or repairs are being carried
out by authorized ROBERT RIELE personnel.

The hard- and software is subject to a program of continuous evaluation and improvement and,
therefore, may be changed in the future. This also concerns service requirements.

RIELE BERLIN Photometer 5010 V5+ 1.1 10.05.2006

1 GENERAL NOTES
Warning: All components used on Photometer 5010 V5+ must be regarded as potentially
dangerously contaminated when doing repair work. Use rubber gloves or double gloves whenever
cleaning or sterilizing components. The most frequently used components (tubing, cuvette,
measuring chamber flap, sip. lever and waste container) should be cleaned and treated with a
suitable disinfecting solution (75% alcohol) prior to doing any service work.

Disinfect and wash hands after work is completed.

This device has been constructed and checked in accordance with standards EN 61010-1 and
EN 61010-2-101. When the device leaves our factory it is in perfect condition from the point of view
of work safety. To keep it that way and to ensure safe operation the user must follow the
instructions and warnings given in the operation instruction manual.

The electrical protection of the device corresponds to Class 1 (it has a protective earth).

The power plug must only be inserted into a socket that has a protective contact. The protection
must not be abolished by using an extension lead that does not have a protective earth wire.

Warning:
Any break in the earth wire inside or outside the device and any loose connection of the earth wire
can make the operation of the device dangerous. A deliberate break or interruption is not allowed.

When the housing is opened or the components are being removed (except when this can be done
by hand), live parts may be exposed. Connection may also be live.

Therefore, if the carrying out of an adjustment, service or repair on the open device in the live state
is unavoidable, this must be done by an expert who is familiar with the dangers involved.

Make sure you only use fuses of the specified type and rated amperage when replacing the old
ones. Repaired fuses must not be used and the fuse holder must not be bypassed.

If you have any reason to believe that the device can no longer operate safely, take it out of use
and make sure no one can use it accidentally.

It must be assumed that safe operation is no longer possible when the device:

• shows visible signs of damage

• fails to operate

• has been stored under unfavorable conditions for a fairly long time

• has been transported under rough conditions

The Photometer 5010 V5+ should be used by appropriately qualified persons only.

RIELE BERLIN Photometer 5010 V5+ 1.2 10.05.2006

2 SERVICE CONCEPTS

4.4 CABLE POWER SUPPLY / PRINTER REF 5010-077
Function Description
On devices previous to SN 8002, after a certain time
of operation, may occur error 52 (see chapter 7) due
to the printer controller power supply plug connection
(P23).

To prevent this, it is possible to replace P23 with the

new power supply cable described in this chapter.
The printer controller power supply CN2 will be
connected then directly to the AC / DC power supply,
eliminating the connection through the motherboard.

Beginning with SN 8002 all devices are equipped

with this cable at factory, thus no replacement is
required.

Location
The printer controller cable supply should be placed
as shown here.

Amplifier for photocurrent and ADC
The photocurrent of the photodiode is converted with short circuit application into a voltage signal with the aid
of the amplifier IC5 (AD 711). Potentiometer P3 sets the amplification and potentiometer P4 sets the offset
voltage of the amplifier stage.

A voltage / frequency converter IC9 (AD654) is connected to the output of the amplifier.

The microcontroller can set the amplification of the amplifier stage by the multiplexer IC10 (ADG428). The
eight different settings of amplification are called V0 up to V7. The setting V0 is equivalent to the lowest
amplification.

The microcontroller converts the frequency signal of the voltage / frequency converter to a digital result in a
range between 0 and ~ 600000. This result is called value of the ADC.

The value of the ADC in the case of photocurrent = 0 (darkness) is called offset. The offset is

• dependent on the setting of the amplifier stage,

• positive and <> 0,
• set with potentiometer P4.

The following diagram shows the ADC-counter values versus the photocurrent.

4.40 PCB RR-120 MOTHERBOARD REF 5010-120
Temperature control
General Description
The temperature control takes care of the correct temperature inside the flow-through cuvette or inside the
standard cuvette adaptor. The temperature is selectable by the program at fixed levels, i.e. 25 °C, 30 °C or
37 °C.

The temperature control circuit consists of:

• microcontroller system
• temperature sensor
• analogue-digital-converter (ADC)
• heating unit
• cooling unit

Microcontroller System
The temperature control circuit is a PID-controller, a software module in the microcontroller system. The PID-
controller works in the background all the time. Dependent on the cuvette adaptor the PID-controller uses
different parameters.

The required value of temperature is ascertained by the program of the microprocessor system. The
temperature is controlled according to the set temperature in the selected method.

The PID-controller gives information to the controlling program about the given temperature tolerances and
therefore about the readiness for measuring.

Temperature Sensor and ADC

The voltage signal of the temperature sensor LM335 is converted by the IC2 (LTC1286) to an equivalent
frequency. The microcontroller unit IC3 (87C52PLCC) converts the frequency to a digital value.
The voltage signal of the temperature sensor LM335 is typical 3.1 VDC at 37 °C, 3.03 VDC at 30 °C and 2.98 VDC
at 25 °C.
The ADC converts the input voltage to a digital value between 0 and 32767 as shown on following diagram:

4.40 PCB RR-120 MOTHERBOARD REF 5010-120
Heating
The control variable of the controller has influence on the heating. A switched Darlington transistor in source of
power-switching is used as heating element. The PID-controller produces a voltage at the emitter of the
Darlington transistor by a pulse-width-modulation. This voltage controls the current through the transistor.
Involved components are the microcontroller IC3 (87C52PLCC) and IC7A/IC7B (LM393).

Cooling
For cooling or counter cooling during heating the microcontroller system switches on a Peltier element. There
are four power levels with following terms and meaning:
0 = Cooling out
1 = Cooling (low level)
2 = Cooling (middle level)
3 = Cooling (high level)

In case of temperature changing by method selection cooling level 0 is selected if desired temperature is
higher as selected one.
In case of temperature changing by method selection cooling level 3 is selected if desired temperature is lower
as selected one.

If selected temperature is reached it is switched over to a cooling level depending on the temperature:
• for 37 degrees Centigrade ⇒ cooling level 1
• for 30 degrees Centigrade ⇒ cooling level 2
• for 25 degrees Centigrade ⇒ cooling level 3
The cooling element is a Peltier element. The Peltier element functions as a heat pump and transports heat
out of the cuvette adaptor to the heat sink. The heat sink is provided with air by the fan of the device and
transports the heat outside to the area.
The Peltier element operates with a voltage, switch able on four levels:
• cooling level 0 ⇒ 0 VDC
• cooling level 1 ⇒ 0.45 VDC
• cooling level 2 ⇒ 0.9 VDC
• cooling level 3 ⇒ 1.3 VDC
Involved components are the microcontroller unit IC3 (87C52PLCC), the OP-booster U1A (LM358), the
transistor T4 (BC547), the transistor T5 (TIP132 mounted at the base plate) and the Peltier element.

Thermal adjustment
The PID-controller algorithm works always as follows:

ACTUAL VALUE = REQUIRED VALUE

REQUIRED VALUE = TEMPERATURE VALUE + CORRECTION VALUE

Explanation:
ACTUAL VALUE ⇔ Actual value of the ADC
REQUIRED VALUE ⇔ Sum of TEMPERATURE VALUE + CORRECTION VALUE
TEMPERATURE VALUE ⇔ Value assigned to a temperature level of 37 °C, 30 °C or 25 °C. This
value is automatically selected by the device according to the state of
operating. The temperature value is board-characteristic and cuvette
adaptor specific. The values for the temperature levels of 37 °C, 30 °C
or 25 °C are ascertained in factory and noted during the check of the
board. They are stored within the battery powered RAM.
Dependent on the used cuvette adaptor different service programs can
show and edit the values stored in battery powered RAM. The system
can be adjusted to lower or higher temperature values. A change of
about 25 counts means a temperature change of 0.1 °C.
• Service program S10 - Temperature value
is used for editing the value
(chapter 3.3)
• Service program S12 - Temperature calibration
calibrates the system automatically.
(chapter 3.3 / chapter 9.1)
The value is noted on a sticker. The sticker is shown on the left and put
RR_120 7000
on top of the IC15 (SABC517).
S FTH The sticker shows the serial number of the board (e.g. 7000) and the
25 °C 8255 8230 temperature values of 25 °C, 30 °C, 37 °C of standard cuvette adaptor
30 °C 9495 9430 (S) and flow-through adaptor (FTH).
37 °C 11198 11113
In addition the values are documented in the device card of Photometer
5010 V5+ and stored in a file at the manufacturer.

CORRECTION VALUE ⇔ Correction value in the range of 0 and 9999. The default value is 3000.

Hardware connection: 1:1 data cable with the following pin configuration:
25-pin 9-pin Photometer 5010 V5+
9-pin female
3 2 2 RXD (Receive Data)
2 3 3 TXD (Transmit Data)
7 5 5 GND (Signal Ground)
7 RTS (Request to send)
8 CTS (Clear to send)

The procedure of communication is as follows:

First activate the remote mode of the Photometer 5010 V5+ starting serial interface utilities by
[UTILITIES] -> [MENU SERIAL COM] -> [REMOTE MODE] (OM chapter 7.2.5.2).

The communication starts with a block where the photometer (master) enables the computer (slave) to
send a command (ACK = acknowledge, NAK = not acknowledge):

Photometer 5010 V5+ <=> Computer

STX MØ ETX BCC =>

<= ACK (or NAK)

STX C ETX BCC =>

<= ACK (or NAK)

Within the next block the computer gives the command (for the list of commands see next page):

Photometer 5010 V5+ <=> Computer

<= STX [command] ETX BCC

ACK (or NAK) =>

optional, if an answer is requested:

STX [answer] ETX BCC =>

<= ACK (or NAK)

The two blocks are appearing like a loop alternately one after the other.

9.1 CALIBRATION PROCEDURE OF
PHOTOMETER 5010
SUBJECT: CUVETTE TEMPERATURE
1. Purpose
This calibration procedure describes the calibration of the cuvette temperature of Photometer 5010. The
meaning of calibration is to determine the difference between the true temperature and the set
temperature. The procedure is to carry out for the temperatures which are used by the customer.

2. General description and measuring theory

The temperature is measured with a selected thermistor-probe. The thermistor is supplied by RIELE. The
certified tolerance of this thermistor is ± 0.1 °C.

The photometer-cuvette temperature is measured in case of using the single cuvette adaptor in an
aluminium-dummy cuvette or in case of using the flow-through cuvette adaptor in the original flow-through
cuvette.

The resistance of the thermistor is measured by a 4½ digit digital multimeter (DMM). The requirements of
the DMM are given in chapter 7. The corresponding temperature is listed in chapter 8.

The implementation of the whole procedure lasts approximately 1.5 hours.

3. Operating conditions
The calibration should be performed under normal operating conditions, i.e.

• Room temperature: 15 °C to 30 °C
• Relative humidity: 40 % to 80 %
• No ventilation or draft

Note that the variation of the room temperature should not exceed ± 2 °C during the calibration process.

4. Traceability of the calibration standards

The traceability to the National Temperature Standard is given by selected thermistor and a calibrated
DMM. The last Calibration Certificate of both, the DMM and the thermistor, must not be older than
12 months. If the 24 month accuracy of the DMM fulfils the requirements in chapter 7, the last DMM
Calibration Certificate can be up to 24 months old.

5. Test procedure
a. In case of using the single cuvette adaptor insert the dummy-cuvette instead of a square cell.
In case of using the flow-through adaptor remove the joint inlet tube cuvette / aspirate tubing and
the outlet tube cuvette.

Pipette 200 µl distilled water in the dummy-cuvette or the flow-through cuvette.

b. Connect the thermistor lead to the two-wire resistance input of the DMM.

c. Insert the thermistor-probe into the hole of the photometer dummy-cuvette or the hole of the outlet
tube cuvette of the flow-through cuvette.
Caution: Handle the delicate probe with care! Pay attention to the correct position of the probe.
Avoid bubbles within the aperture of the flow-through cuvette.

d. Switch on the DMM and Photometer 5010 and wait 15 minutes for warming up.

e. Select the two-wire resistance function and appropriate range on the DMM. The resistance of the
thermistor is in the range of 6 kOhm up to 10 kOhm for the temperature range of 25 °C up to
37 °C. The table of resistance of thermistor (REF 090-063) shows the assigned resistance for a
given temperature (see chapter 9 – item 8).

f. Enter the service program level.

9.1 CALIBRATION PROCEDURE OF
PHOTOMETER 5010
SUBJECT: CUVETTE TEMPERATURE
For additional information see chapter 3.3 – SERVICE TOOLS.

g. Perform a system initialization by applying service program item 1 – S1 - System initialization.

h. Enter the service program item 9 – S9 Temperature ON/OFF.

i. Select the temperature 25° C.

j. Check the temperature control by using service program item 9 – S9 Temperature ON/OFF.
As long as temperature is not stabilized, the stability is showing a value different of zero. It needs
minimum five minutes until a stabilized temperature level is reached. Wait 10 minutes
furthermore.

k. Read the displayed measurement of DMM in Ohm. Look up in the table of resistance the
appropriate set of values. Calculate by performing a linear interpolation the assigned temperature
value (see chapter 9 – item 8).

l. Make an automatic temperature calibration by using service program item 12 – S12 -

Temperature calibration.

m. Check the temperature control by using service program item 9 – S9 Temperature ON/OFF.
As long as temperature is not stabilized, the stability is showing a value different of zero. It needs
minimum five minutes until a stabilized temperature level is reached. Wait 5 minutes furthermore.

n. Read the displayed measurement of DMM in Ohm. Look up in the table of resistance the
appropriate set of values. The temperature value should show the desired value.

o. Read the required value and subtract the value 3000. The result is called temperature value. Note
this value in calibration protocol combined with adaptor type and temperature level.
See for additional information in chapter 4.40 – MOTHER BOARD – Thermal adjustment.

p. Repeat above mentioned steps with the temperature levels 25 °C, 30 °C and 37 °C for both types
of adaptors.

6. Documentation
For documentation use a form containing following table:

Single cuvette adaptor Flow-through adaptor

25 °C Temperature value Temperature value
30 °C Temperature value Temperature value
37 °C Temperature value Temperature value

7. Requirements for the Digital Multimeter (DMM)

The DMM for the resistance-measurements has to fulfil the following requirements:

• 12 month accuracy of resistance measurements

within the 0 – 30 KOhm range: 0.05 % + 2 digits
• Current for resistance measurement: ≤ 15 µA
• Latest Calibration Certificate: not older than 12 months

If the 24 months resistance-accuracy fulfils the above accuracy requirements, the latest
Calibration Certificate can be ≤ 24 months old.

9.1 CALIBRATION PROCEDURE OF
PHOTOMETER 5010
SUBJECT: CUVETTE TEMPERATURE
9. Linear interpolation
This procedure describes the performing of a linear interpolation in case of a given resistance R.
The assigned temperature is called T.

If the resistance R is found in the table, get the appropriate temperature value and finish this chapter.

If the actual temperature of the thermistor is in the range of the required temperature ± 1.5 °C, two values
of resistance can be read from table with the relation:

R0 > R > R1

The resistances R0 and R1 form an interval. They are a set of data points next to the resistance R.

The assigned temperature values of the resistances R0 and R1 are called T0 and T1.
The assigned temperature T can be calculated by the formula:

T = T0 + (T1 – T0) × (R - R0) / (R1 – R0)

Example:
In case of a given resistance R with the value 9978 Ohm you can find the interpolated temperature T by:

T = 25 °C + 0.1 °C × (9978 - 10000) / (9956 – 10000)

T = 25.05 °C
