red dragon fruit (Hylocereus polyrhizus) peel contains phenolic compounds that have the potential as a

source of natural antioxidants that can be used as photoprotector. This study aims to determine the
effect of topical aplication of red dragon fruit peel extract to mast cells count as a parameter of
protection to the skin of mice that were exposed to UV-B acute.

This experimental study using 45 female mice strain BALB / C were randomly divided into 5 groups
which KI, KII, KIII, KIV and KV as normal group; negative group; dose of 0.256 mg; a dose of 1.28 mg and
6.4 mg doses. Red dragon fruit peel extract lotion is applied to the backs of mice every day for 7 days.
Mice were then exposed to UVB rays with a single dose of 3 MED (for 24 minutes). The preparat for the
back skin of mice made by using Toluidine blue staining and then counting the average number of mast
cells.

The number of mast cells in the negative control group was observed at 24.48 hours, and 72 to
determine the peak of mast cells increase. The treatment group was observed at a peak time (24 hours)
with a mean number of mast cells in K-I, K-II, K-III, K-IV and K-V respectively of 3.77; 4.67; 5.68; 2.83 ;
and 3.38. One Way Annova test showed a significant difference (p <0.05) in the mean number of mast
cells. Post Hoc LSD test results showed significant difference with K K-II-I, K-III, IV-K and K-V.

Conclusion: The topical application of red dragon fruit peel extracts affect the number of mast cells in
the skin of mice were exposed to UVB acute.
