Tujuan dari pedoman ini ("Pedoman Tes") adalah untuk menjabarkan prinsip-prinsip yang terkandung
dalam Pendahuluan Umum (dokumen TG / 1/3), dan dokumen TGP yang terkait, ke dalam panduan
praktis terperinci untuk pemeriksaan selaras, keseragaman dan stabilitas (DUS) dan, khususnya, untuk
mengidentifikasi karakteristik yang sesuai untuk pemeriksaan DUS dan produksi deskripsi varietas
yang diharmonisasikan.

DOKUMEN ASOSIASI

Pedoman Tes ini harus dibaca sehubungan dengan Pendahuluan Umum dan dokumen TGP

1. Subjek dari Pedoman Tes ini Pedoman

Tes ini berlaku untuk semua varietas Capsicum annuum L.

2. Bahan Yang Diperlukan

2.1 Pihak berwenang yang kompeten memutuskan jumlah dan kualitas bahan tanaman yang diperlukan untuk
menguji varietas dan kapan dan di mana ia akan dikirim. Pelamar yang mengirimkan bahan dari suatu Negara
selain dari tempat pengujian dilakukan harus memastikan bahwa semua formalitas pabean dan persyaratan
phytosanitary dipenuhi.

2.2 Bahan harus disediakan dalam bentuk benih.

2.3 Jumlah minimum bahan tanaman, yang akan dipasok oleh pemohon, harus:

2.500 biji.

2.4 Benih harus memenuhi persyaratan minimum untuk perkecambahan, spesies dan kemurnian analitik,
kesehatan, dan kadar air, yang ditentukan oleh pihak yang berwenang.

2.5 Bahan tanaman yang disediakan harus tampak sehat, tidak kurang kuat, atau terpengaruh oleh hama atau
penyakit penting.

2.6 Bahan tanaman seharusnya tidak mengalami perlakuan apa pun yang akan memengaruhi ekspresi karakteristik
varietas, kecuali jika pihak yang berwenang mengizinkan atau meminta perlakuan semacam itu. Jika sudah dirawat,
detail lengkap dari perawatan harus diberikan.

3. Metode Pemeriksaan

3.1 Jumlah Siklus Tumbuh

Durasi minimum tes biasanya adalah dua siklus pertumbuhan mandiri.

3.2 Tempat Pengujian

Pengujian biasanya dilakukan di satu tempat. Dalam kasus tes yang dilakukan di lebih dari satu tempat,
panduan diberikan dalam TGP / 9 “Memeriksa Perbedaan”.

3.3 Kondisi untuk Melakukan Pemeriksaan

3.3.1 Pengujian harus dilakukan dalam kondisi yang memastikan pertumbuhan memuaskan untuk ekspresi
karakteristik yang relevan dari varietas dan untuk pelaksanaan pemeriksaan.

3.3.2 Metode yang direkomendasikan untuk mengamati karakteristik ditunjukkan dengan kunci berikut di kolom
kedua dari Tabel Karakteristik:

MG: pengukuran tunggal sekelompok tanaman atau bagian-bagian tanaman


MS: pengukuran sejumlah tanaman individu atau bagian-bagian tanaman
VG: penilaian visual dengan pengamatan tunggal terhadap sekelompok tanaman atau bagian-bagian tanaman

3.4 Desain Uji

3.4.1 Setiap tes harus dirancang untuk menghasilkan total setidaknya 20 pabrik yang harus dibagi antara 2 ulangan.

3.4.2 Desain pengujian harus sedemikian rupa sehingga tanaman atau bagian tanaman dapat dipindahkan untuk
pengukuran atau penghitungan tanpa mengurangi pengamatan yang harus dilakukan sampai akhir siklus
pertumbuhan.

3.5 Jumlah Tanaman / Bagian Tanaman yang akan Diperiksa

Kecuali disebutkan sebaliknya, semua pengamatan harus dilakukan pada 20 tanaman atau bagian yang
diambil dari masing-masing 20 tanaman.

3.6 Tes Tambahan Tes

4. Penilaian Perbedaan, Keseragaman dan Stabilitas

4.1 Perbedaan

4.1.1 Rekomendasi Umum

Sangat penting bagi pengguna Pedoman Uji ini untuk berkonsultasi dengan Pengantar Umum sebelum
membuat keputusan mengenai perbedaan. Namun, poin-poin berikut disediakan untuk elaborasi atau penekanan
dalam Pedoman Tes ini.

4.1.2 Perbedaan Konsisten Perbedaan yang

diamati antara varietas mungkin sangat jelas sehingga tidak perlu lebih dari satu siklus tumbuh. Selain itu,
dalam beberapa keadaan, pengaruh lingkungan tidak sedemikian rupa sehingga diperlukan lebih dari satu siklus
tumbuh untuk memberikan jaminan bahwa perbedaan yang diamati antara varietas cukup konsisten. Salah satu
cara untuk memastikan bahwa perbedaan dalam karakteristik, diamati dalam uji coba pertumbuhan, cukup
konsisten adalah untuk memeriksa karakteristik dalam setidaknya dua siklus pertumbuhan independen.

4.1.3 Perbedaan Jelas

Menentukan apakah perbedaan antara dua varietas jelas tergantung pada banyak faktor, dan harus
mempertimbangkan, khususnya, jenis ekspresi dari karakteristik yang diperiksa, yaitu apakah itu dinyatakan dalam
kualitatif, kuantitatif, atau pseudo- secara kualitatif. Oleh karena itu, penting bagi pengguna Pedoman Tes ini untuk
memahami rekomendasi yang terkandung dalam Pendahuluan Umum sebelum membuat keputusan mengenai
perbedaan.

4.2 Keseragaman

4.2.1 Sangat penting bagi pengguna Pedoman Tes ini untuk berkonsultasi dengan Pengantar Umum sebelum
membuat keputusan tentang keseragaman. Namun, poin-poin berikut disediakan untuk elaborasi atau penekanan
dalam Pedoman Uji ini:

4.2.2 Untuk penilaian keseragaman varietas yang diserbuki silang, standar populasi 2% dan probabilitas
penerimaan minimal 95% harus diterapkan. Dalam hal ukuran sampel 20 pabrik, 2 jenis tidak diizinkan diizinkan.

4.2.3 Untuk penilaian keseragaman hibrida, standar populasi 1% dan probabilitas penerimaan minimal 95% harus
diterapkan. Dalam hal sampel ukuran 20 tanaman, 1 off-type diperbolehkan.

4.3 Stabilitas

4.3.1 Dalam praktiknya, tidak biasanya melakukan pengujian stabilitas yang menghasilkan hasil yang pasti seperti
pada pengujian perbedaan dan keseragaman. Namun, pengalaman telah menunjukkan bahwa, untuk banyak jenis
varietas, ketika varietas telah terbukti seragam, itu juga dapat dianggap stabil.

4.3.2 Apabila diperlukan, atau dalam hal keraguan, stabilitas dapat diuji, baik dengan menumbuhkan generasi
selanjutnya, atau dengan menguji stok benih baru untuk memastikan bahwa itu menunjukkan karakteristik yang
sama seperti yang ditunjukkan oleh bahan sebelumnya yang disediakan.

5. Pengelompokan Varietas dan Organisasi dari Uji Coba Tumbuh

5.1 Pemilihan varietas pengetahuan umum untuk ditanam dalam uji coba dengan kandidat varietas dan cara
varietas ini dibagi menjadi kelompok-kelompok untuk memfasilitasi penilaian perbedaan yang dibantu oleh
penggunaan karakteristik pengelompokan.

5.2 Karakteristik pengelompokan adalah karakteristik di mana ekspresi ekspresi yang terdokumentasi, bahkan
ketika diproduksi di lokasi yang berbeda, dapat digunakan, baik secara individu atau dalam kombinasi dengan
karakteristik lain yang serupa: (a) untuk memilih varietas pengetahuan umum yang dapat dikecualikan dari
pertumbuhan uji coba yang digunakan untuk pemeriksaan perbedaan; dan (b) untuk mengatur uji coba penanaman
sehingga varietas yang sama dikelompokkan bersama.

5.3 Berikut ini telah disepakati sebagai karakteristik pengelompokan yang berguna:

(a) Pembibitan: pewarnaan antosianin hipokotil (karakteristik 1)


(b) Tanaman: ruas pendek (di bagian atas) (karakteristik 4)
(c) Buah: warna (sebelum jatuh tempo) (karakteristik 21)
(d) Buah: bentuk dalam longitudinal bagian (karakteristik 28)
(e) Buah: warna (pada saat jatuh tempo) (karakteristik 33)
(f) Buah: capsaicin dalam plasenta (karakteristik 45)

Pendahuluan Umum.

6. Pengantar Tabel Karakteristik

6.1 Kategori Karakteristik

6.1.1 Pedoman Tes Standar Karakteristik

Standar Pedoman Tes karakteristik adalah yang disetujui oleh UPOV untuk pemeriksaan DUS dan dari
mana anggota Perhimpunan dapat memilih yang cocok untuk keadaan khusus mereka.

6.1.2 Karakteristik Asterisk Karakter

Asterisk (dilambangkan dengan *) adalah yang termasuk dalam Pedoman Uji yang penting untuk
harmonisasi internasional dari deskripsi varietas dan harus selalu diperiksa untuk DUS dan dimasukkan dalam
deskripsi varietas oleh semua anggota Perhimpunan, kecuali ketika keadaan ekspresi dari karakteristik sebelumnya
atau kondisi lingkungan regional membuat ini tidak pantas.

6.2 Kondisi Ekspresi dan Catatan yang Berkaitan Kondisi

ekspresi diberikan untuk setiap karakteristik untuk mendefinisikan karakteristik dan untuk menyelaraskan
deskripsi. Setiap keadaan ekspresi dialokasikan catatan numerik yang sesuai untuk kemudahan pencatatan data dan
untuk produksi dan pertukaran deskripsi.

6.3 Jenis Ekspresi

Penjelasan tentang jenis ekspresi karakteristik (kualitatif, kuantitatif dan pseudo-kualitatif) disediakan
dalam Pendahuluan Umum.

6.4 Varietas Contoh

Bila perlu, varietas contoh disediakan untuk memperjelas kondisi ekspresi masing-masing karakteristik.

8.1 Explanations covering several characteristics

Characteristics containing the following key in the second column of the Table of
Characteristics should be examined as indicated below:

(a) Fruit characteristics whi ch should be examined before maturity, ie before the first color change

(b) Fruit characteristics which should be examined at maturity, ie after the time of the first color change

8.2 Explanations for individual characteristics

Ad. 3: Plant: length of stem

The length of the stem is measured from the cotyledons to the first flower branch.

Ad. 4: Plant: shortened internode (in upper part)

Ad. 5: Varieties with shortened internodes only: Plant: number of internodes between the first flower and
shortened internodes

The tests should be done on plants which have not been pruned. The shoot system of pepper consists of
main stems, which are branched off from the main axis and side shoots. Two growth types of the main stems can
be distinguished:

Growth type A: the main stems grow indeterminately; one or two flowers develop per node and shortened
internodes never develop.

Growth type B: after the first branching of the main axis, shorter internodes appear and the growth of the
main stem ends in a bunch of flowers (it appears as if there are more than two flowers per node).

Side shoots develop from the nodes on the main axis and on the main stems.

TSWV is transmitted by thrips (Thrips tabaci and
Frankliniella occidentalis.). TSWV has a broad host
range.

After a few multiplication the virus could be ineffective.
New isolates can be obtained from practice by
harvesting fruits of L4 pepper varieties infected
naturally with TSWV. The fruits are kept at -70°C
temperature. The presence of other viruses must be
checked before using this material.
