I. INTRODUCTION
In order to produce a good dissection, it is necessary: to understand the techniques of
dissection and to dissect carefully; to have available an adequate range of dissection instruments
in good condition; and to have an adequate understanding of the basic anatomy of the animal being
dissected
Much of the skill of dissection is common sense. Before you start a dissection make sure
that you have adequate illumination, and that there is enough space for you to rest the weight of
your arms on the table whilst dissecting. Also, use a new sharp scalpel blade for each dissection.
If you experiment with ways of making a cut, you will find it much easier to cut in some
directions than in others. It is much easier to cut away from yourself or to make a scalpel cut along
a line pointing obliquely to your left. Therefore, whenever necessary, turn the dish, or board, so
that you can cut in these directions.
When dissecting, begin in an area where there is little danger of damaging the structures
you wish to display. Gradually extend your area of dissection outwards from this point. Always
cut upwards with your scalpel into the overlying tissues. Never cut down towards the structures.
If you do, you will be more likely to damage the organs before you see them. Remember that blood
vessels and nerves normally run in connective tissue between blocks of muscle. Dissection is
therefore most sensibly carried out by separating the muscles, and cutting through a block of
muscle only after it has been fully separated from the surrounding tissue. On completing a stage
of dissection, trim back any ragged edges of the tissues you have cut. Pieces of black card can be
placed behind nerves and blood vessels in order to display them.
Most systems in most animals are bilaterally symmetrical. It is therefore sensible to attempt
one side first. If you make mistakes, carefully note what has happened and then use the knowledge
you have gained to make a better dissection on the other side. Use a notebook in which to record
reminders of the difficult phases and danger points of the dissection.
Your notebook containing the drawings of your dissections is important as a record of what
you, as a scientist, have discovered and observed. Adequate materials and care are the first
essentials of a good drawing. Use a reasonably sharp pencil and don't use glossy or lined paper.
The drawing of your dissection should be large enough for the smallest object in it to be
clearly and accurately drawn. If one region has smaller, more detailed structures than the
remainder, it may be useful to make an additional drawing of this region, noting its relationship to
the drawing of the whole dissection.
Techniques which make your drawing easier to understand should be used, for example
pencil shading and the use of color.

III. ACTIVITY
Part A. Pithing Procedure
Before the experiment, you need to render the frog insensitive to pain. Pithing is one
procedure to accomplish this. Pithing will destroy the brain, which is located between the eyes.
Pithing is relatively painless to the frog. Read the entire procedure before beginning so you can
perform the procedure quickly.
1. Hold the frog facing away from your body, with the lower extremities extended.
2. Grasp the frog with your first two fingers: first finger on the nose, second finger under the
jaw. Flex the head forward (away from your body).
3. Move probe down midline until you come to the soft spot. Do not use undo pressure on the
frog skin. Frog skull is fairly hard cartilage; soft spot is at the end of the skull.
4. Insert the probe quickly into the cranial vault and sever the brain and spinal cord.
5. Move the probe into the cranial vault and move it from side to side to destroy the brain.
You can feel inside the cranial vault, but the frog can’t feel anything. Without the brain,
perception of any sensory phenomena is impossible.
6. Test for reflexes to confirm that sensory perception has been destroyed and ensure that the
spinal cord is still intact, although it is now in shock.
7. Keep the frog moist. The frog will still be breathing because frogs breathe primarily
through the skin, not the lungs, so it is important to keep the frog moist
Part B. External Anatomy
1. Make sure frog is right side up so that you can see the face.
2. Identify the eyes, which have a nictitating membrane that serves to moisten the eye.
3. Identify the tympanic membrane, or eardrum. It is located behind each eye.
4. Find the external nares or nostrils.
5. Find the digits, which are like fingers on both the fore and hind limbs
6. Determine if your frog is male or female by looking at the innermost finger of the forelimb.
The male pad is an enlarged darker patched pad that the male uses to grasp a female during
mating.
Part C. Mouth
1. Pry open the mouth. Use the scissors to cut the corner of the mouth where the maxilla
(upper jaw) and mandible (lower jaw) join together.
2. Find the tongue and pull it out.

3. Feel the maxillary teeth that are along the rim of the upper jaw. Notice that only the
upper jaw has teeth.
4. Locate the glottis. It is a slit opening for the voice box on the floor of the mouth.
5. Find the esophagus at the rear of the mouth.
Part D. Internal Anatomy: Dissection
Opening up the frog:
1. Place the frog ventral side up (on its back) and pin its four limbs down.
2. Lift the frog’s skin with forceps between the rear legs.
3. Make a small cut through the lifted skin with the scalpel. Take care to cut only the skin.
You are making a starting place for the scissors.
4. Use the scissors to continue the incision up to the midline all the way through the frog’s
skin.
5. Stop cutting when your scissors reach the frog’s neck.
6. Now, you will cut horizontally. Use the scissors to make sideways incisions in the skin
7. The first incisions are made between the front legs. Then do the same with the rear legs.
8. Be careful to cut only skin, not muscle.
9. Pick up the flap of skin with the forceps.
10. Use a scalpel to help separate the skin from the muscle layer below. Then pin the skin to
the dissection tray.
Cutting the Muscle and Bone:
a. First muscle incision
1. Do the same incisions, this time through the abdominal muscle. You will find it
easier to begin the vertical incision by lifting the muscle layer with the forceps. Do
this between the rear legs of the frog.
2. Make a small cut with the scalpel.
3. Use the scissors to continue the incision up the middle to just below the front legs.
4. Don’t cut too deeply, the muscle is thin. It is easy to damage the organs underneath.
