What muscle do? FUNGSI OTOT
Voluntary vs Involuntary
Pergerakan, jalan, lari, mempertahankan posisi tegak tubuh dll.
Respirasi, pencernaan
Produksi panas (thermogenesis) – menggigil
30-40% tubuh adalah otot daging
KARAKTERISTIK OTOT
Eksitabilitas – mampu menerima & menanggapi rangsangan luar
Kontraktilitas – mampu memendek
Ekstensibilitas – dapat diregang
Elastisitas – mampu kembali ke bentuk awal setelah diregang

TIPE OTOT
1. OTOT POLOS (involuntary)
Umumnya digunakan pada kontraksi organ ( sal. pencernaan,
kantung kemih dan pembuluh darah) untuk pergerakan materi
ke dalam atau luar tubuh
2. OTOT LURIK (striated)
1. Kerangka (voluntary), terdiri dari:
a. Tipe I
b. Tipe IIa
c. Tipe IIb
2. Jantung (involuntary)

Myofibril terdiri dari sarcomer -
subunit yang berulang dan
tersusun secara seri.
Sarcomer adalah unit fungsional
(komponen terkecil yg dapat
menyelenggarakan semua fungsi
organ) otot kerangka, unit
kontraktif terkecil.
Setiap Sarcomere mengandung
proteins panjang- Myofilaments
◦ Tebal - Myosin 1.6 µm
◦ Tipis - Actin 1.0 µm
◦ Filamen tipis dan tebal saling
Thin menumpuk sebagian
membentuk pola pita.
Thick ◦ Protein pengatur: tropomiosin
dan troponin
◦ Protein tambahan: titin dan
nebulin
Sarcomere menyebabkan pola
lurik/striation pada otot
kerangka.

• Panjang filamin aktin dan miosin tidak berubah selama kontraksi (hanya bergeser)
• Lebar daerah tumpang tindih semakin melebar (H zones and I bands are regions of non-overlap.
• Filaments tipis bergeser sepanjang filamen tebal.
• Karena filamen tipis terikat pada lempeng Z maka sarcomer memendek. 21

Pada tahap ini ATP berikatan dengan Miosin.

Twitch: respons otot kerangka terhadap rangsangan tunggal atau potensial aksi:
•Periode latent - no change in length; time during which impulse is traveling along
sarcolemma & down t-tubules to sarcoplasmic reticulum, calcium is being released,
and so on (in other words, muscle cannot contract instantaneously!)
•Periode kontraksi - tension increases (cross-bridges are swivelling)
•Periode relaksasi - muscle relaxes (tension decreases) & tends to return to its

PERBANDINGAN SINGLE UNIT AND MULTI UNIT
Single-Unit smooth muscle – serabut kontraksi bersama dan bekerja sebagai
satu unit (single unit).
Seperti serabut jantung, kecil, memanjang and pipih pd tiap ujungnya.
Dihubungkan dengan lainnya melalui gap junctions
Depolarisasi spontan (myogenic) – disebarkan ke sel dekatnya secara aktif
Kontraksi peristaltik – gelombang aktifitas yang bergerak sepanjang saluran
pencernaan, mendorong makanan. Juga ditemukan di urinary bladder,
ureters, dan uterus
Rangsangan syaraf memodulasi kekuatan frekwensi kontraksi, tapi tidak
menginisiasinya, seperti pada otot jantung.

• Dapat ditemukan pada dinding organ: digestive
tract, urinary bladder, uterus
• Fungsi viseral - gerakan peristaltik untuk
mendorong isi atau squeezing to expel contents
– inervasi oleh sistim syaraf otonom – beberapa
otot polos dapat juga berfungsi secara
independent (myogenic).
• Heterogenous - banyak subtipe
• Tidak ada Sarkomer shg tidak nampak lurik
• Berinti satu
• Retikulum sarkoplasma tidak berkembang
• Tidak ada tubulus T
• Memiliki filamen tipis dan tebal tapi tidak
tersusun dalam sarkomer.
• Bundel filamen tipis melekat pada Dense Bodies atau Attachment Plaques –
dihubungkan dgn penghubung khusus yg juga mengikat sel tetangga - mengandung
alpha-actinin (seperti lempeng Z pd otot kerangka) dan vinculin - khusus pada otot
polos.
• Bundel Bundles of Thin Filaments are interdigitated with the thin filaments.
• Gap junctions pada otot polos single-unit - kontraksi bersama-sama.

Beberapa otot polos kontraksi dan relaksasi sangat lambat.
Mekanisme Eksitasi-kontraktion berbeda dari otot kerangka tapi masih
tergantung pada [Ca2+] dlm sarkoplasma.
Retikulum sarcoplasmic tidak berkembang - Ca2+ datang dari
ekstraseluler dengan cara:

aktin:
• Protein Caldesmon berikatan dg
filamen halus, mencegah ikatan dg
myosin.
• Caldesmon dihilangkan dg:

calmodulin and kompleks
calmodulin/Ca2+ berikatan dg
caldesmon, pembebasan daerah
pengikatan kepala miosin pd aktin.
2. Foforilasi caldesmon by Protein
Kinase C. Phosphorylated
caldesmon does not bind to actin,
so myosin does bind actin.

• Smooth muscles lack troponin. Have
other regulatory mechanisms - for
myosin:
– Binding of Ca2+ directly to myosin
regulatory light chains changes
myosin conformation, allowing it
to bind actin.
– phosphorylation of myosin light
chains by Myosin Light Chain
Kinase. Myosin LC Kinase is
activated by Ca2+/calmodulin.
– Phosphorylation of another site on
the myosin regulatory light chain
by Protein Kinase C induces a
conformational change that
prevents actin myosin binding -
relaxation.
• Slow action of kinases, and slow
changes of Ca2+ levels - slow rate of
contraction of smooth muscle.

MUSCLE HYPERTROPHY AND ATROPHY
A. Repeated, exhaustive stimulation of a muscle will tend to
result in an increase in muscle mass: Hypertrophy
1. addition of more myofibrils (i.e., an increase in diameter)
2. increase in the machinery for energy production
– more mitochondria
– more glycolytic enzymes
note: Hypertrophy does not involve the addition of new
muscle fibers

• Myogenic – dimulai dari otot itu sendiri (berlawanan dg
neurogenic)
– Bioeletrisitas muncul secara spontan pada pacemaker.
Mereka menimbulkan potensial aktion secara ritmis.
– PA menyebar melalui gap junction yang ada di serabut otot.
– serabut otot jantung kontraksi secara bersama-sama.
• Inervasi oleh sistim syaraf sympatis dan parasympatis.
– Tidak menghasilkan potensial aksi pada sinaps
– Berperan sebagai modulator.
– Syaraf symphatis meningkatkan kekuatan & kecepatan
kontraksi
• Parasympathis menurunkan kecepatan dan kekuatan kontraksi.

Tubulus T mengalirkan potensial aksi jauh di dalam serabut otot.
Depolarisasi mengaktifkan reseptor dihydropyridine pada Tubulus T.
Influx of Ca2+ dari ruang extrasellular melalui reseptor
dihydropyridine – tidak seperti otot kerangka dimana yang penting
adalah aktivasi mekanis dari reseptor ryanodine pada retikulum
sarcoplasmic. Tidak ada aktivasi mekanis pada serabut otot jantung.
Ca2+ dari ruang extrasellular memicu reseptor ryanodine pada
retikulum sarcoplasmic untuk terbuka, melepas Ca2+ dari simpanan
intrasellular - Ca2+ yang tergantung dari Ca2+ release.
Ca2+ dikeluarkan melalui Pompa Ca2+ pada retikulum sarkoplasma
dan melalui pertukaran Na+/ Ca2+ di membran plasma.
• Kekuatan kontraksi proposional dengan konsentrasi Ca2+ intraseluler.
• Ca2+ dari ruang extracellular and dari simpanan intracellular bervariasi
antara species – pada hewan (mis: kodok) with serabut otot yang kecil
(larger surface/volume ratio), pada umumnya Ca2+ berasal dari ruang
extrasellular, sedikit daridari SR, yang berkembang jelek.

Tension in isolated frog muscle as a function of
depolarization. The greater the depolarization, the
greater the tension.
Tension also depends on extracellular Ca2+
concentration. With greater extracellular Ca2+, more
Ca2+ enters the cell at any level of depolarization
(greater driving force).
Little or no change seen with changing extracellular
Ca2+ in mammalian heart, where Ca2+ comes from
intracellular stores.
Increase in contraction strength by sympathetic
nervous system stimulation. Catecholamines:
• stimulate release of Ca2+ from SR (a-adrenergic
receptor and innositol phospholipid 2nd
messenger).
• Increase Ca2+ conductance of plasma membrane
(b-adrenergic receptors and adenylate cyclase
