1. Tujuan Penelitian :
Memahami efek dari triploidisasi pada meiosis pada ikan, mempelajari struktur, tingkat metilasi, dan
tingkat ekspresi gen terkait meiosis (Dmc1, Ph1) di diploid Carassius auratus (2n = 100, disingkat
2nCC), Carassius auratus merah (2n = 100, disingkat RCC), 3nCC dan 3nRR.
2. Metode :
Pertama pengumpulan sampel diploid ikan mas (Carassius auratus) dan dari keturunan yang saling
silang dari triploid jantan dan betina dipilih secara acak dan juga ikan mas crucian merah yang dimana
semua sampel tersebut dianestesi sebelum pengumpulan darah. Kedua, hibridisasi fluoresensi in situ
dengan dilakukannya persiapan kromosom dengan menggunakan kultur sel darah perifer dari semua
sampel. Ketiga pengukuran konten DNA dengan aliran sitometer (Partec GmbH) untuk mendeteksi
kandungan DNA dari darah dan sperma. Keempat, struktur gonadal yang dipilih secara acak 10 RCC, 10
2nCC, dan 10 3nCC pada usia 9 bulan untuk pengamatan histologis struktur gonad, sedangkan 10 3nRR
dipilih secara acak untuk observasi. Kelima isolasi RNA dan sintesis cDNA dengan menggunakan a Kit
pereaksi PrimeScript ™ RT dengan gDNA Eraser. Keenam, kloning wilayah CDS dengan cDNA sebagai
tempat, wilayah CDS yang dikloning dari jaringan gonad. Ketujuh, analisis PCR kuantitatif real-time
untuk mempelajari ekspresi Ph1. Dan membandingkan hasil nya dengan menggunakan genom ikan zebra
sebagai referensi serta DNA yang diekstraksi. Dan hasil kloning dikirim ke Sangon untuk diurutkan.
Metilasi urutan yang dihasilkan adalah dengan BiQ analyzer.
3. Hasil :
Fluoresensi dalam situasi hibridisasi, 3nCC menghasilkan anak autotriploid biseksual yang ovum nya
diaktifkan oleh spermatozoa 3nCC. Dua belas 5S lokus rDNA terdeteksi di 88% dari metafase kromosom
3nRR adalah autotriploid dan memiliki tiga set turunan RCC kromosom. Indung telur 3nCC dan 3nRR
mengandung banyak oosit fase III, yang mengindikasikan bahwa ovariumnya dapat subur. Testis 3nCC
subur dan 3nRR tidak subur karena kosong dan rusak. Gen Ph1 memiliki homologi yang signifikan pada
3nRR dan 3nCC, dan gen Dmc1 sangat bagus. Gen Ph1 dan Dmc1 sangat diekspresikan dalam 3nCC
jantan dan betina dan individu betina 3nRR (p <0,05) dibandingkan dengan 2nCC dan RCC, tetapi
diekspresikan secara rendah pada 3nRR individu jantan (p <0,05). Metilasi tingkat daerah promotor Ph1
dan Dmc1 pada individu jantan 3nRR adalah 0,983 dan 0,883. Ada korelasi negatif antara tingkat metilasi
dan ekspresi relatif dari daerah promoter gen Ph1 dan Dmc1.
4. Kesimpulan :
Ikan autotriploid dapat menjadi subur dan dapat menghasilkan gamet normal. Kesuburan ikan dapat
dinilai dengan mengamati perkembangan gonad selama musim kawin, dan fitur histologis gonad
menunjukkan bahwa 3nCC jantan dan betina keduanya memiliki struktur gonad normal dan dapat
berkembang secara normal dan menghasilkan gamet, dan 3nRR dapat menghasilkan telur normal tanpa
menghasilkan sperma normal. Autotriploidation tidak sepenuhnya mengarah pada hilangnya kesuburan
dan tidak menyebabkan infertilitas total.

Abstract
Considering the advantages of gene editing technologies, in recent years, emphasis has been given on three main techniques of
gene editing i.e. zinc finger nucleases (ZFNs), transcription activator-like effector nucleases (TALENs), and the CRISPR/Cas9
RNA-guided endonuclease system. In all of these three technologies one thing is common i.e. the technology utilizes restriction
enzymes to break down the double stranded DNA molecule at a targeted location with the help of another molecule of
homologous binding protein or RNA, which is also called as a guide RNA molecule. This targeted breaking and repair of the
DNA molecule as per our requirement is generally viewed as a great breakthrough in gene therapy methods. In fisheries sector, in
order to promote aquaculture/mariculture activities, aquaculture industries are facing number of challenges, particularly in area of
quality and demand of seed production, control of health and disease management, production of quality traits with
phenotypically improved varieties, and strengthening of immune system. Several efforts are being made both at public and
private sectors to develop scientific technologies to meet these challenges and substantial achievements have also been made. But
still these challenges remained as major constraints in hampering the growth of this industry as per the demand and expectations.
However, with the advent of now recently developed gene editing techniques, we may have to explore and evaluate the
possibilities of applications of this technology as a long lasting solution in addressing at least some of the vital issues of the
aquaculture industry particularly in those related to altering targeted gene structure in the species for positive impact.
