HOEFER SEMI-DRY TRANSFER UNITS use electrophoresis to transfer proteins and nucleic acids from polyacrylamide or agarose gels onto nitrocellulose or nylon membranes.
The HOEFER IMMUNOBLOT AND IMMUNOBLOT XL make it possible to assay the material on a transfer membrane with different antigens or antibody probes simultaneously without cutting the membrane into strips. The membrane remains intact to preserve the correspondence between different lanes or positions on the gel.
SEE PAGE 32
The HOEFER DQ 300 FLUOROMETER is a compact, dual-channel unit that delivers quick, easy, and accurate fluorescence measurements with either longwave UV or blue excitation.

HOEFER CALIBER ELECTROPHORESIS REAGENTS deliver consistently outstanding results. A high standard of purity, rigorous analytical procedures, and carefully controlled manufacturing techniques ensure that Hoefer Caliber reagents are the best quality buffer, gel casting, gel staining and sample preparation chemicals available.
The HOEFER UVC 5000 ULTRAVIOLET CROSSLINKER is excellent for cross-linking DNA or RNA to membranes for multiple probings of Southern, Northern, dot, or slot blots; for DNA nicking in agarose gels; and for sterilizing samples. The UVC 5000 can be stacked on top of the HB 1000 hybridizer to save valuable lab space.
The HB 1000 HYBRIDIZATION OVEN is designed to create the ideal environment for nucleic acid hybridization.
The HOEFER ULTima is a conveniently upgradable imaging platform incorporating advanced electronics and optics. The four models of gel documentation systems can handle the most demanding applications in Proteomics and Genomics without expensive laser scanning systems.

Because of their semi-rigid shape and the regular phosphate charge distribution along their length, DNA fragments separate by size in simple electrophoresis systems. Agarose is the preferred gel matrix used, which is easy to cast and has a good size separation range. For separation of 100 bp to 20 kbp fragments, we recommend the Hoefer horizontal systems, HE 99X MAX SUBMARINE UNIT and HE 33 MINI SUBMARINE UNIT. Nucleic acids can also be separated in vertical acrylamide gels, which have a much higher resolving power than agarose gels but a narrower size range of separation. For vertical separations of small DNA fragments such as oligonucleotides up to 1000 bp, we recommend the HOEFER SE 300 miniVE or the SE 250 and SE 260 MIGHTY SMALL UNITS. For even further increased separation, we recommend the longer gels of the HOEFER SE 600 or SE 400 SERIES STANDARD VERTICAL ELECTROPHORESIS UNITS.

TE Series Transfer Systems
Electrophoretic transfer of proteins is a much faster procedure than simple capillary or contact blotting methods. By directing electrical current through a transfer stack, blotting rates can be greatly accelerated. Assays performed on transfer membranes are sensitive and require only small amounts of material. This is critical when the species of interest is present in low concentrations or represents a minor fraction of the total sample. Moreover, since antibodies and other affinity reagents are often expensive, this can greatly reduce cost. HOEFER TE SERIES TRANSFER TANKS and SEMI-DRY TRANSFER UNITS use electrophoresis to deliver excellent transfers of proteins and nucleic acids from polyacrylamide or agarose gels onto nitrocellulose, PVDF or nylon membranes.

The TE 70X and TE 70XP SEMI-DRY TRANSFER UNITS use minimal buffer to transfer proteins and nucleic acids from polyacrylamide and agarose gels in less than an hour. The TE 70XP has the added convenience of a built-in power supply. Units accept gels up to 14 x 16 cm, or two SE 260 mini gels side-by-side. Transfer two polyacrylamide gels in a stack. Economical to use. Require only a small amount of buffer. Require minimal current. Do not generate the excessive heat that can dry out the transfer stack and halt transfer or damage transfer units. Durable platinum-niobium and stainless steel electrodes assure contamination-free transfers. TE 70XP includes an intelligent built-in power supply which automatically monitors the transfer status, stopping the transfer before the stack overheats.

IMMUNOBLOTTING
Immunoblotting
ImmunoBlot and Deca-Probe Blotting Manifolds
Transfer of proteins or nucleic acids, fractionated by gel electrophoresis, to an immobilizing membrane surface increases the sensitivity of a wide range of detection methods, reduces time of analysis, and makes sequential probing possible. In particular, it has made possible the use of immunological procedures that are not practical to carry out on a gel or in a tube or plate. The HOEFER IMMUNOBLOT AND IMMUNOBLOT XL and HOEFER PR 150 DECAPROBE make it possible to assay the material on a transfer membrane with different antigens or antibody probes simultaneously without cutting the membrane into strips. The membrane remains intact to preserve the correspondence between different lanes or positions on the gel. The HOEFER PR 648 SLOT BLOT MANIFOLD provides a simple, convenient method to immobilize unfractionated samples to a membrane, allowing screening of recombinant clones, DNA, RNA and proteins.

ULTima
Imaging Platform
The ULTima imaging platform is the fastest, most straightforward and readily upgradable unit available incorporating the latest advances in electronics and optics. The four ULTima models can economically handle everything from the most routine to the most demanding applications in proteomics and genomics which have until now required expensive laser scanning systems.
Full line of upgradable instruments (ULTima10si, 16si, 16si Plus and Pro): Allows you to upgrade to higher specifications as research needs change. Unique patented image positioning: Makes positioning of gels accurate and quick with a mininum of handling of fragile gels. Slant-correction: Permits the optical rotation of the sample for best straight visualization without touching the gel. True multi-fluorophore capabilities: Supports a wide range of applications and is particularly well suited for proteomics (Cy-dye imaging) Super Sensitivity: Performs fast chemiluminescent imaging. Enables faint-band fluorescent detection. Electronic 5 position emission filter changer: Allows automatic application setup for all imaging applications. Large image format of 21 x 26 cm: Captures full large-format proteomic gel images in a single pass. All acquisition parameters stored: Facilitates GLP use. Smaller footprint with door that slides up: Uses absolute minimum working space and eliminates the need to step back to open the door. The optional ULTima Optix illumination module provides high intensity excitation of your sample with visible and nearinfrared light.

Slant Correction Correction Slant correction is a unique feature that allows the user to turn the image optically 5 degrees. In this way the sample can be positioned so the analysis software is better able to perform lane detection.
Image Positioning Imag Positioning Unique image positioning using patented optics. Every user of an imaging system has experienced the problem that the gel is never in the right place initially. With most commercially available systems the user has to open the drawer or door to reposition the gel for the best visualization. The ULTima however is the only imaging system with patented optics for image positioning without loss of optical resolution. This also ensures that lens adjustments are not required. The image is positioned with the positioning keys without having to open the cabinet or adjust a fragile gel.
User Interface In order to be able to acquire a high quality image, full control of the optics is indispensable. The Image Control screen is always visible at the bottom of the image, providing ultimate control over the imaging process. The user has the choice of either setting the exposure time manually, or making use of the fully automatic capabilities of the ULTima. Full lens control allows optimization of a broad range of applications. Robot Ready The UV 26 transilluminator unit is fully accessible. Samples for automated 2D analyses can be loaded by robotic arm/system. Quantifiable Data The demanding ULTima user can be certain of producing high quantifiable data. The flat fielding function performs a uniform calibration for every individual image pixel. GLP Documentation of results and the integrity of data becomes much more important for drug discovery and genetic engineering. GLP (Good Laboratory Practice) can only be achieved if all image parameters and conditions are known and archived. ULTima is fully equipped for GLP validation and verification.
Templates Templates allow the user to define the settings for image acquisition, including the excitation and emission filters. The parameters are stored per specific application. Superior Dynamic Range Since every pixel within the image represents intensity data, it is important to achieve the highest possible conversion resolution. In most commercially available systems this is 8, 10 or 12 bit, representing 256, 1024 or 4096 grey levels. The ULTima 16si, ULTima 16si Plus and the ULTima Pro have real 16 bit dynamic ranges which means 65536 real different grey levels. This offers much more precise quantification but also allows for shorter exposure times for chemiluminescent images. The exposure time is normally much shorter than the exposure time needed for film with comparable image quality. Broad Range of Applications The emission filter changer accommodates a maximum of 5 filters. The filters are selected from the drop-down list or from free definable templates. As standard the ULTima is equipped with an emission filter for the detection of ethidium bromide and other fluorophores within in the range from 560 to 640 nm (e.g. SYPRO Ruby). Filters for most other fluorophores are available (for example SYBR Green II, Cy3, Cy5, FITC, etc.).

Optix
TotalLab
Analysis Software
The Optix module is a patented epi-illumination option for the excitation of fluorophores within the VIS/NIR range. When Optix is used for ethidium bromide stained gels, it offers the following advantages: DNA remains intact for applications downstream. Lower background means higher detection limits. Selection of excitation wavelength is fully computer controlled and, in combination with the automatic emission filter changer, true multi fluorophore capability is achieved. Multiple images with different excitation/emission filter sets allow the detection of multiple fluorophores within the same sample. The Optix can accommodate a maximum of five different excitation filter sets. The Optix module can be added to all models of the ULTima line. Applications that are possible with the addition of the Optix module include: CyDye imaging, array detection, in vivo GFP detection and GFP tissue and organ screening.
