Intended Use Baird-Parker Egg Yolk Agar

Using Baird-Parker Egg Yolk Agar, the ISO-GRID and NEO-GRID Baird-Parker Agar
Membrane Filter Systems are used to provide a method for enumerating Tryptone 10 g/L
Staphylococcus aureus in foods. Beef Extract 5 g/L
Yeast Extract 1 g/L
The Test Sodium Pyruvate 10 g/L
The ISO-GRID/NEO-GRID systems are based on the principle of Glycine 12 g/L
hydrophobic grid membrane filtration (HGMF). Staphylococcus aureus is Lithium Chloride-6-Hydrate 5 g/L
detected through the use of a unique membrane filter containing a grid Agar 17 g/L
of 1,600 squares. First, a diluted sample is prefiltered to eliminate any Final pH: 7.0 ± 0.2 @ 25°C
food particles present. The sample is then filtered through a hydropho-
bic grid membrane filter, and the membrane is placed on Baird-Parker Egg Yolk Tellurite Emulsion (100 mL)
Egg Yolk Agar. Baird-Parker Egg Yolk Agar is specifically formulated Chicken egg yolk 100% solution
to detect Staphylococcus aureus. Potassium Tellurite 0.21 g
After incubation the membrane filter is examined for the presence of
black or dark grey colonies. Any black or dark gray colonies are a pre-
sumptive positive for Staphylococcus aureus, and should be confirmed Ordering Information
using procedures detailed in the Methods Manual.

Test Procedure
7112 Baird-Parker Agar
1. Prepare a sample homogenate. 7983 Egg Yolk Tellurite Emulsion (100 mL)
2. Filter 1 mL of homogenate through an ISO-GRID or 6810 1-Place Filtration Manifold
NEO-GRID hydrophobic grid membrane filter. 6800 3-Place Filtration Manifold
3. Place the membrane filter on Baird-Parker Egg Yolk Agar. 6801 6-Place Filtration Manifold
4. Incubate for 48 hours at 35 - 37°C. 6821 Rubber Tubing (10 feet)
