Struktur Light & Heavy Chain
Analisa struktur
Pemecahan oleh enzim Papain (Porter,1959)
Enzim Papain memecah pd hinge region
3 fragments (MW = 150,000 Da), 45,000 Da/each
Fab fragments (Fragmen antigen binding)
Tetap memiliki sifat Antibodi, mengikat Ag
Bereaksi dengan determinan Antigen dan hapten
Univalent
2 fragmen yang identik
Fc fragment (fragment crystallizable)
Dapat dikristalkan dari larutan
Tidak dapat mengikat Ag
Menunjukkan fungsi biologis sesudah Ag diikat oleh Fab
Struktur Light & Heavy Chain
Pemecahan oleh enzim Pepsin
Memecah pada asam amino sebelum ikatan disulfid
Divalent fragment disebut sebagai F(ab)2
Terbentuk beberapa Fc subfragments
Antibodies disrupt
function of pathogens

Antigen Presentation
Antigen yg masuk melalui usus akan
dihancurkan oleh gut-associated lymphoid
tissues.
Antigen yg masuk melalui kulit atau
respiratory tract akan dihancurkan secara
local atau ditransport ke local lymph nodes.
Antigen yg memasuki darah akan disaring di
dalam spleen.
Immune Response-
Step by Step
1. Pathogen (carrying foreign antigens) enters
and survives the inflammatory response
2. Some pathogens remain exposed in tissues
where their antigens may be recognized
by circulating B cells
OR
3. Macrophages engulf pathogens and display
their antigens on MHC (major
histocompatibility complex) receptors.
Macrophage has now become an Antigen
Presenting Cell (APC)
Antigenic Determinants
(Epitopes)
Tempat spesifik dari antigen yang berikatan
dengan antibodi disebut antigenic
determinant atau epitope.
Kebanyakan antigen memiliki berbagai jenis
epitope yang merangsang antibodi yang
berbeda disebut sebagai polyclonal.
Immune respons tunggal terhadap suatu
antigen disebut sebagai monoclonal.
Monoclonal Antibodies
Laboratory produced antibodies
Penyerapan IgG ibu dlm colostrum diperoleh akibat berikatan IgG dgn
Fc receptors dlm jaringan intestinal
Kehamilan 3-4 bln, tjd peningkatan yg cepat dari IgG ibu
Setelah umur 5 bln, fetus mulai menghasilkan IgM & IgA dlm jumlah yang kecil
3-4 bln setelah kelahiran, level IgG ibu mulai menurun infant mulai
menghasilkan Ig G sendiri
CH4
Properties of IgM
Ig pertama yang terbentuk setelah immunisasi
Macroglobulin (M), dengan MW = 900,000 Da
Memiliki extra CH domain
Pentamer molecule (tdd 5 rantai polipeptida)
Memiliki ikatan disulfid tambahan antara Fc portions dengan rantai
polipeptida J chain
Valence of 5 (5 Ag combining sites)
Ditemukan lebih banyak pada ruang intravascular
Half life dalam 5 hari
Ditemukan pd permukaan B cells yg matang bersama dg IgD berfungsi sbg Ag
specific B cell receptor (BCR) setelah aktivasi B cells terjadi pergantian klas utk
membentuk isotypes lainnya.
Biological Properties of IgM (continued)
Tidak menembus placenta
Ig yang hanya disintesis oleh fetus (5 months
gestation)
Peningkatan level IgM pd fetus menunjukkan
congenital atau perinatal infeksi
Isotype pertama yang disintesis setelah imunisasi
terhadap T-dependent Ags
Kadar IgM yang meninggi biasanya menunjukkan
infeksi yang baru terjadi atau paparan thd Ag

IgE
Biological Properties of IgE
Ig dengan jumlah paling sedikit dalam serum
Berikatan dgn basophils and mast cells (tidak
memerlukan ikatan Ag)

ANTIBODY FUNCTIONS
The Specific Defense System
Very complex system of cells and
chemicals
Interacting and collaborating with the non-
specific defenses
Activity carefully controlled
malfunctioning of system: allergies,
autoimmune diseases
Immunological memory immunity
Immunity
Immune system primed to eliminate antigen after a
previous exposure (active immunity)
passive immunity: antibodies administered to
individual (medical treatment or mother-fetus across
placenta)
tetanus, snake venoms, hepatitis, botulism, rabies,
etc.
Vaccination: artificially acquired immunity
weakened pathogen, recombinant DNA produced
vaccines (artificial pathogen proteins)
irradicated small pox, polio, whooping cough, etc.
