SCENARIO

Salwa is a colleger who is learning about the signal pathways

that control the activity of gene. Salwa get a task to make a
journal review about the role of TGF-β in the tumor. From
that journals, she gets an information that TGF-β is known to
mediate the activity of systolic suppression of NK cell. This is
because induction of microRNA (mir-183) that made protein
DAP 12 not expressed and make there is no signal to
mobilize granulitik. How is the role mechanism of TGF-β
against NK cell and the linkages with the tumor cell.
DIFFICULT WORDS

• microRNA
is a family of RNA that does not encode (non-coding RNA) which controls the expression of genes
in cellular signal transduction pathways
• DAP 12
DNAX activating protein of 12KDA
• TGF – β
Regulations that has multifunctional properties that can enhance or inhibit many cell functions,
including disrupt the production of other cytokines and increase collagen deposition. It exists in
multiple subtypes and produced by platelets and macrophages but can be done with many other
cell types.
• Supresion
In genetics, a second mutation that occurs at a different site from the first mutation site and were
able to mask or suppress the expression of phenotypic first mutation; organism seems to be
restored but in reality the double mutant.
DIFFICULT WORDS

• NK cell
A lymphocyte that activated by double-stranded RNA or lymphokines and fight against viral infections
and tumors without apparent antigenic specificity.
• Gene
A functional unit of heredity that occupies a specific place (locus) on the chromosom, is able to reproduce itself
exactly at each cell division, and direct the formation of enzymes or other proteins.
• Tumor
abnormal swelling or enlargement; This is one of the cardinal signs of inflammation.
KEYWORDS

• TGF – β
• microRNA
• NK cell
PROBLEM’S IDENTIFICATION

• 1. How is the signal path can control the

activity of a gene?
• 2. ow is the function of micro rna
• 3. How MicroRNA works with DAP12 in the
granulitic mobilization process?
• 4. How can normal cells into abnormal cells?
• 5. How is the role of TGF-β in the tumor cells?
• 6. How Cells NK association with tumor cells
MIND MAPPING

TGF β

DAP 12

Non active NK
cell
LEARNING OBJECTIVE

• To know and understand the signaling pathways that control the activity
of a gene
• To determine the role of microRNA and DAP12 and also associations
between them
• To determine the mechanism of the role of TGF-β against NK cell and its
association with tumor cells
SIGNALLING PATHWAYS THAT
CONTROL THE ACTIVITY OF GENE
MICRORNA FUNCTION

• miRNA that has been identified has a function that can be

seen that the molecules that regulate a variety of processes
important role in the growth, differentiation, apoptosis,
adhesion, and other cellular processes.
• abnormal miRNA expression can promote tumorigenesis,
metastasis, and a variety of other cancer properties
COOPERATION BETWEEN MICRORNA AND
DAP12 IN THE GRANULITIC MOBILITATION
PROCESS

in that cases, TGF β become mutated so that TGF β

mediate mikrorna to inhibit DAP12 work
And so it will make a NK cell become non active and
then make a tumor cell. And then, because of the DAP
12 that being obstructed, it also make a granulitic
mobilitation process become obstructed too
NORMAL CELL BECOME ABNORMAL
CELL

Transformation of normal cells to become abnormal

cell caused by there is a mistake in the checkpoint G2
/ M which is in that phase, abnormal cell is still go
through division and thereby causing tumor
ROLE OF TGF-Β IN TUMOR CELL

in that cases, TGF β become mutated so that

TGF β mediate mikrorna to inhibit DAP12 work
And so it will make a NK cell become non active
and then make a tumor cell
