Isolasi Enzim
alfa-Glukosidase
 Isolasi dan penentuan pH buffer optimum
ekstrak kasar enzim α-glukosidase (Risma, 2012)

01 Beras ketan putih bersih

• Dihaluskan dengan blender

• Diayak ukuran 100 mesh
02 Tepung beras ketan putih
• Ditambahkan 50 mL buffer variasi pH 5,0;
5,5; 6,0; 6,5; 7,0; 7,5; 8,0
• Diaduk dengan stirrer selama 1 jam
• Didiamkan ± 12 jam dalam lemari
pendingin
• Disentrifuge 3500 rpm selama 20 menit
suhu 4 °C
• Disaring dengan kain kasa
 Isolasi dan penentuan pH buffer optimum
ekstrak kasar enzim α-glukosidase (Risma, 2012)

• Diuji aktivitasnya

setelah memperoleh pH buffer optimum

yang baik digunakan, isolasi diulangi
dengan skala yang lebih besar, 400 g
tepung beras dalam 1000 mL buffer

1. Fraksinasi dengan amonium sulfat

01 Ekstrak kasar enzim

• Ditambahkan garam amonium sulfat 0-20%

sedikit demi sedikit di atas
• magnetic stirrer dilengkapi icebath
• Setelah penambahan garam terakhir,
pengadukan dilanjutkan 20 menit
• Didiamkan selama ± 20 jam pada suhu 4 oC
• Disentrifugasi 8000 rpm 20 menit pada suhu 4 oC
• Dipisahkan supernatan dan endapan

2. Dialisis dengan membran selofan

01 10 cm membran selofan

• Direbus dalam campuran NaHCO 2% dalam EDTA

1 mM selama 10 menit
• Larutan diganti dengan akuades dan kembali
direbus selama 10 menit

 Pemurnian enzim α-glukosidase
(Bollag dan Edelstein, 1991)
Fraksi amonium sulfat dengan aktivitas enzim terti
01 nggi
• Dimasukkan ke dalam membran selofan, bagian atas dan bawah membran
selofan diikat
• Direndam dalam larutan buffer dengan konsentrasi sepuluh kali lebih encer
daripada buffer larutan enzim
• Dilakukan dialisis selama 10 jam pada kondisi dingin, sambil diaduk dengan
magnetic stirrer
• Setiap 2 jam dialisat diganti, setiap dialisat yang dikeluarkan diuji dengan larutan
Ba(OH)2.8H2O 2% untuk mengetahui apakah masih terdapat garam
• Dialisis dihentikan bila dialisat sudah tidak mengandung garam
• Dilakukan pengukuran volume larutan enzim, pengujian kadar protein, dan
pengujian aktivias enzim

Alfa-Glukosidase dalam Pengobatan
Diabetes
Inhibitor alfa-glukosidase bekerja secara kompetitif dan reversibel dalam menghambat
alfa-glukosidase, enzim usus. Ini akan memperlambat pencernaan karbohidrat serta
menunda penyerapan glukosa yang juga memperlambat mengurangi peningkatan dalam
kadar glukosa darah setelah makan secara efektif sepanjang hari.
