Hepatosit Hepar pada Tikus Putih Jantan (Rattus Novergicus Strain Wistar). Tugas

Latar belakang: Kopi sebagai minuman, mengandung acrylamide terbentuk pada proses
roasted. Paparan acrylamide pada hepar meningkatkan ROS, TNF, IL-1 yang mengaktivasi
sel stelata menjadi miofibroblas menjadi jaringan fibrosis dan akan menjadi sirosis. Sirosis
adalah faktor resiko karsinoma sel hati.
Tujuan: Membuktikan pengaruh acrylamide yang terkandung dalam seduhan kopi robusta
(Coffea canephora var. Robusta) terhadap perubahan morfologi sel hepatosit hepar pada tikus
putih jantan (Rattus novergicus strain wistar).
Metode: True experimental post test only control group design. Sampel 24 ekor tikus dibagi
6 kelompok; K1 Negatif, K2 Positif diberi acrylamide, K3,K4,K5, dan K6 diberi dosis
seduhan kopi 0.36ml/200gBB, 0.72ml/200gBB, 1.44ml/200gBB, dan 2.16ml/200gBB.
Analisis data menggunakan uji One-Way ANOVA, Post Hoc Tukey, Korelasi dan Regresi.
Hasil penelitian dan diskusi: One-Way ANOVA menunjukkan nilai sig. sebesar 0.000
(P<0.05) berarti berpengaruh yang bermakna. Uji Tukey nilai sig. (P<0.05) pada K4,K5,K6
berbeda signifikan terhadap K1 negatif sedangkan K3 tidak berbeda signifikan terhadap K1
negatif. Uji korelasi pearson didapatkan nilai 0.899. Kekuatan seduhan kopi berpengaruh
80.9% terhadap jumlah sel hepatosit normal.
Kesimpulan: Seduhan Kopi Robusta (Coffea canephora var. Robusta) dapat menyebabkan
perubahan morfologi sel hepatosit hepar pada tikus putih jantan (Rattus novergicus Strain
wistar).

(Coffea canephora var. Robusta) on Morphological changes of Hepatocytes Cell of
Liver in Male Rats (Rattus novergicus Strain Wistar). Final Assignment, Faculty of

Background: Coffee beans as beverages that contains acrylamide that formed in roasted
process. Acrylamide exposure in liver increase ROS, TNF, IL-1 that activated stellate cell
into myofibroblast that produce fibrosis tissue and will be form as liver cirrhosis. Liver
cirrhosis is the risk factors of hepatocellular carcinoma.
Objective: The research aims to prove the influence of acrylamide contained in steering of
Robusta Coffee (Coffea canephora var. Robusta) on Morphological changes of Hepatocytes
Cell of Liver in Male Rats (Rattus novergicus Strain wistar).
Methods: True experimental post test only control group design. 24 samples of male rats
clustered in 6 groups; K1 Negative, K2 Positive was given Acrylamide, K3,K4,K5 and K6
were given doses of coffee infusion of 0.36ml/200gBB, 0.72ml/200gBB, 1.44ml/200gBB,
and 2.16ml/200gBB. Analysis of the data using One-Way ANOVA, Post Hoc Tukey,
Correlation, and Regression.
Results: One-Way ANOVA result shows with significant value 0.000. That value is (P<0.05)
that mean that there is a significant effect. Tukey test shown that significant value is
(P<0.05) in the group K4,K5,K6 significantly different to the K1 negative while K3 shows no
significant difference to K1 negative. Pearson correlation test obtained value of 0.899. The
intensity of the correlation shows the effect of coffee steeping is 80.9% correlated of the
number of normal hepatocytes in the liver.
Conclusion: Steeping of robusta coffee (Coffea canephora var. Robusta) could cause
morphological changes of hepatocytes cell of liver in male rats (Rattus novergicus Strain
Wistar).
