KULTUR JARINGAN BEBERAPA KULTIVAR
BUAH PISANG (Musa paradisiaca L.)
DENGAN PEMBERIAN CAMPURAN NAA
DAN KINETIN
bertujuan untuk mengetahui pengaruh
interaksi antara campuran NAA dan
Kinetin terhadap pertumbuhan tiga
kultivar buah pisang secara teknik
kultur jaringan.
A.Latar belakang
Kendala pengadaan bibit unggul secara konvensional adalah sulit mendapatkan bibit yang berkualitas
dalam jumlah besar dalam waktu yang singkat. Salah satu keunggulan perbanyakan tanaman melalui
teknik kultur jaringan adalah sangat dimungkinkan mendapatkan bahan tanam dalam jumlah besar dalam
waktu singkat (Priyono et al., 2000).

Dari sekian banyak jenis media dasar yang digunakan dalam teknik kultur jaringan, tampaknya media MS
(Murashige dan Skoog) mengandung jumlah hara organik yang layak untuk memenuhi kebutuhan banyak
jenis sel tanaman dalam kultur (Gunawan, 1990).

Dalam kultur jaringan, dua golongan zat pengatur tumbuh yang sangat penting adalah sitokinin dan auksin
(Gunawan, 1990).
B.Tujuan penelitian
(1) pengaruh interaksi antara NAA dan kinetin terhadap pertumbuhan eksplan tiga kultivar bakal buah
pisang yang ditanam dengan teknik kultur jaringan,

(2) pengaruh masing-masing konsentrasi campuran NAA dan kinetin dengan kultivar pisang yang terbaik
terhadap pertumbuhan eksplan bakal buah pisang yang ditanam dengan teknik kultur jaringan,

(3) pengaruh campuran NAA dan kinetin yang terbaik pada kultivar pisang, dan

(4) pengaruh kultivar pisang terhadap tingkat keberhasilan kultur jaringan bakal buah pisang.

1.Kintekin
dalah sejenis sitokinin, kelas hormon tanaman yang mendorong pembelahan sel. Kinetin pada awalnya
diisolasi oleh Miller [1] dan Skoog et al. [2] sebagai senyawa dari DNA sperma herring autoklaf yang
memiliki aktivitas pembelahan sel. Itu diberi nama kinetin karena kemampuannya untuk menginduksi
pembelahan sel, asalkan auxin hadir di media. Kinetin sering digunakan dalam kultur jaringan tanaman
untuk menginduksi pembentukan kalus (bersama dengan auksin) dan untuk meregenerasi jaringan tunas
dari kalus (dengan konsentrasi auksin lebih rendah)

2.NAA
Hormon NAA adalah senyawa kimia yang termasuk dalam golongan auksin sedangkan BAP termasuk
dalam golongan sitokinin yang berperan dalam pertumbuhan tunas.
Naphthaleneacetic Acid (NAA) merupakan golongan auksin yang berfungsi dalam menginduksi
pembentangan sel dan inisiasi pengakaran. Sementara itu, 6- Benzylamino Purin (BAP) berfungsi
merangsang pembelahan sel dalam jaringan eksplan dan merangsang pertumbuhan tunas (Wattimena et
al., 1992).
