Night At The Museum 2 Larry Daly ( Ben Stiller) yang sudah meninggalkan pekerjaan lamanya sebagai penjaga museum,

kini menjadi pencipta barang keperluan sehari hari. Suatu hari, Larry berkunjung ke Museum tempat kerjanya dulu, dan menemukan bahwa teman temannya sudah masuk ke dalam kardus, siap dipindahkan ke museum di Washington D.C. Lalu, suatu malam, teman museumnya menelpon, bahwa mereka membutuhkan Larry, di rumah baru mereka tersebut. Ternyata karena pendapatan museum yang menurun, maka sebagian besar dari koleksi di museum tersebut akan dipindahkan ke museum di kota lain. Dan itu berarti nasib dari teman teman Larry menjadi tidak jelas. Larry bersama anaknya pun bertekad akan melakukan sesuatu untuk membantu mereka. Di museum baru yang lebih besar, Larry pun bertemu dengan lebih banyak lagi tokoh tokoh yang hidup, seperti Amelia Earhart (Amy Adams) si penerbang wanita pertama dari Amerika, juga Napoleon Bonaparte, Al Capone sampai raja raja Mesir yang berantem karena memperebutkan warisan leluhur mereka.

Seluruh penumpang pesawat Air Force One disandera termasuk istri Presiden, Grace Marshall (Wendy Crewson) dan putri mereka, Alice (Liesel Matthews). Namun Presiden Marshall berhasil lolos dari tangkapan para teroris dan menyembunyikan diri dalam pesawat yang luas itu. Walau sempat kesal dengan lolosnya sang presiden, Egor langsung menghubungi Gedung Putih di Washington untuk menuntut pembebasan pemimpin mereka, Jenderal Radek dari penjara. Jika tidak dipenuhi, para teroris akan mengeksekusi seorang penumpang setiap setengah jam sampai permintaan mereka dipenuhi. Sementara Presiden Marshall yang disangka para teroris telah meloloskan diri dari pesawat dengan menggunakan sekoci penyelamat, ternyata diam-diam berusaha mengambil kendali terhadap pesawat. Para teroris yang akhirnya tahu usaha Marshall, berusaha menangkapnya hidup-hidup. Tetapi mereka telah meremehkan kemampuan sang presiden yang merupakan mantan prajurit pemenang penghargaan tertinggi negara "Medal of Honor" di medan perang Vietnam. Dengan fisiknya yang masih fit dan kemampuan bela diri, Marshall mencoba menghabisi para teroris satu persatu di samping berusaha membebaskan istri, putrinya dan para penumpang lainnya. Sementara ketegangan di dalam pesawat kepresidenan makin mencekam, situasi yang tidak kalah tegang pula dialami oleh Wakil Presiden Kathryn Bennett (Glenn Close) yang langsung mengadakan pertemuan darurat untuk menyelesaikan pembajakan tersebut. Sesuai kebijakan pemerintah Amerika Serikat, mereka tidak boleh memenuhi tuntutan teroris walau harus membahayakan pemimpin mereka. Selagi Kathryn masih bingung mengambil keputusan, ia didesak oleh Menteri Pertahanan Walter Dean (Dean Stockwell) untuk mengambil alih kekuasaan karena sang presiden dapat dianggap berhalangan secara tetap secara teknis. Wakil Presiden tersebut masih ragu-ragu juga walau permintaan Walter Dean didukung oleh Jaksa Agung Amerika (Philip Baker Hall).
