ABSTRACT
Newcastle Disease (ND) is a disease that is spread widely and become one
of the diseases to wary at the poultry farm. The disease caused by a
Paramyxovirus type 1 (APMV-1). NDV causes the death of more than 90%. The
incidence of this disease can occur throughout of the years. In Indonesia, ND
virus is known to have two forms, velogenic viscerotropic newcastle disease
(VVND) and velogenic neurotropic newcastle disease (VNND). Proper diagnosis
became one of the success factors in controlling the disease. This study was
purpose to identification the NDV with the gel presipitation test (AGP). This
research used four samples of the suspected ND, Layer/Hari/P-2011, Layer/Afkir-
2013, Pullet/Hari-2012 and Pullet/Vera-2013 that showed symptoms of torticollis.
Virus samples was inoculated in 11 days old embrionated chicken eggs with the
negative antibody of ND. Then, allantoic fluid was identificated by using
hemagglutination test (HA) and hemagglutination inhibition test (HI). The
principle of HI test is inhibit reaction between antigen and red blood cell by using
anti ND serum. Positive viruse samples was tested to gel precipitation test (AGP)
as the confirmaion test. AGP test used four wells for each isolate and a vaccine for
the control wells. The gel was incubated at room temperature and not exposed
directly to the light. The line of precipitation will be observed after 24 -48 hours
of incubation, indicated by the white line between the allantoic fluid and anti ND
serum on the plat of gel agar presipitation test (AGP). The result of HA and HI
test was showing positive interpretation. The precipitation line formed between
wells on gel agar precipitation test (AGP). From this result can be concluded that
four samples are positive NDV.
