Abstrak
Penanaman bakteri atau biasa disebut juga inokulasi adalah pekerjaan memindahkan
bakteri dari medium yang lama ke medium yang baru dengan tingkat ketelitian yang sangat
tinggi. Untuk melakukan penanaman bakteri (inokulasi) terlebih dahulu diusakan agar
semua alat yang ada dalam hubungannya dengan medium agar tetap steril, hal ini agar
menghindari terjadinya kontaminasi. Metode yang digunakan pada praktikum ini adalah
metode gesek dengan agar miring. Agar ini mempunyai permukaan luas sehingga sering
digunakan untuk menumbuhkan atau menyimpan biakan murni. Dari hasil yang didapat
terlihat bahwa bakteri yang ada pada medium terlihat buram namun jelas bila disinari
dengan cahaya. Terlihat bentuk dari koloni mikroorganisme yang tampak pada agar miring
berbentuk seperti akar. Hal ini dikarenakan oleh beberapa faktor misalnya lamanya waktu
inkubasi, kontaminasi, dan lain-lain. Sifat koloni pada agar miring, yaitu dilihat pada bentuk
dan tepi koloninya dan sifat-sifat itu dinyatakan dengan bentuk serupa pedang, serupa duri,
serupa tasbih, serupa titik-titik, serupa batang dan serupa akar.

Abstract
Planting bacteria or also called inoculation is the work of transferring bacteria from the old
medium to the new medium with a very high level of accuracy. In order to carry out
bacterial planting (inoculation) it is first damaged so that all tools that are in contact with
the medium in order to remain sterile, this is in order to avoid contamination. The method
used in this lab is the swipe method with sloping agar. For this to have a broad surface, it is
often used to grow or store pure cultures. From the results obtained it appears that the
bacteria present in the medium look blurry but clear when exposed to light. Visible form of a
microorganism colony that appears on the sloping to form like a root. This is due to several
factors such as the length of time of incubation, contamination, and others. The nature of
the colony on sloping agar, which is seen in the shape and edges of the colony, and the
properties are expressed in the form of a sword, like a thorn, like a rosary, like a dots, like a
stem and like a root.
