A. TITRIMETRI
Satu teknik analitik yang memungkinkan
penetapan kuantitatif dari satu senyawa
spesifik (analit) yang terlarut dalam sampel.
Didasarkan kepada satu reaksi yang sempurna
antara analit dan satu reagen yang telah
diketahui konsentrasinya yang ditambahkan
kepada sampel.
Karena pengukuran volume memainkan
peranan kunci dalam titrasi, maka dikenal
sebagai analisis volumetrik.

1. Titrasi Asam-Basa:
Didasarkan kepada reaksi netralisasi antara
analit dan satu titran asam atau basa.
2. Titrasi Redoks:
Didasarkan kepada reaksi oksidasi-reduksi
antara analit dan titran.
3. Titrasi Kompleksiometri:
Didasarkan kepada pembentukan satu
kompleks antara analit dan titran. Chelating
agent EDTA sangat umum digunakan untuk
mentitrasi ion logam dalam larutan.

4. Titrasi Presipitimetri:
Didasarkan kepada pembentukan endapan
sebagai hasil reaksi antara analat dengan titran
5. Titrasi Karl Fisher (Coulometry)
Digunakan dalam penetapan kadar air dalam
sampel yang kadarnya sangat rendah
Teori Titrasi
Prosedur Titrasi
Titrasi dimulai dengan satu Erlenmeyer berisi reaktan
yang telah diketahui volumenya dan sedikit indikator
diletakkan di bawah satu buret berisi reagen.
Dengan mengontrol jumlah reagen yang ditambahkan ke
dalam reaktan, memungkinkan untuk mendeteksi titik
dimana indikator berubah warna.
Sepanjang indikator tersebut telah dipilih dengan benar,
titik saat terjadinya perubahan warna harusnya juga mrpk
titik reaktan dan reagen saling menetralkan.
Dengan membaca skala buret, volume reagen dapat
diukur. Begitu konsentrasi dari reagen diketahui, jumlah
mol dari reagen dapat dihitung (karena konsentrasi=mol).
Dari persamaan kimia yang melibatkan dua senyawa,
jumlah mol yang ada dalam reaktan dapat ditemukan.
Dan dengan membagi jumlah mol dari reaktan dengan
volumenya, konsentrasi bisa dihitung.
Titrasi Kompleksiometri:
Titrasi ini umumnya memerlukan indikator
khusus yang membentuk kompleks yang lebih
lemah dengan analit.
Contoh yang umum adalah Eriochrome Black T
untuk titrasi ion kalsium dan magnesium.
Satu bentuk dari titrasi juga dapat digunakan
untuk menentukan konsentrasi virus atau bakteri.
Kurva Titrasi
Titrasi sering direkam pada kurva titrasi,
yang komposisinya umumnya identik.
independent variable merupakan volume
titrant, dan dependent variable merupakan
pH larutan (yang berubah tergantung
kepada komposisi dari dua larutan).
Titrasi Balik
Istilah titrasi balik (back titration) digunakan bila
satu titrasi dilakukan secara terbalik
("backwards) bukan mentitrasi analit asal.
Satu standar dalam jumlah berlebih
ditambahkan ke dalam larutan, kemudian
kelebihannya dititrasi
Titrasi balik berguna
bila titik akhir dari titrasi kebalikan lebih mudah
diidentifikasi daripada titrasi yang normal.

1. Dalam penetapan vitamin C secara titrimetri
dengan metode dichloroindophenol, 100 g
sampel ditimbang dan diencerkan sampai 500
ml dengan larutan pengekstrak. 25 ml filtrate
tersebut kemudian dititrasi dengan larutan dye
(dichloroindophenol. Jumlah titran yang
digunakan adalah 9.1 ml dan konsentrasi asam
askorbat dalam larutan dye adalah 0.175
mg/ml. Hitunglah konsentrasi vitamin C dalam
sampel tersebut?

2. 25,023 gram sampel ikan segar dikeringkan,
kemudian diabukan dan akhirnya dianalisis
kadar garamnya dengan metode titrasi Volhard.
Berat dari sampel setelah pengeringan adalah
5.001 gram dan berat abunya adalah 1,002
gram. Kemudian 30 ml AgNO3 0.1 N
ditambahkan kedalam sampel yang telah
diabukan. Endapan yang dihasilkan disaring
dan sejumlah kecil ferric ammonium sulfat
ditambahkan ke dalam filtrat. Filtrat kemudian
dititrasi dengan 3 ml KSCN 0.1 M sampai titik
akhir berwarna merah dicapai.
B. Spectroscopy Theory
Spectroscopy Theory
1. Spectroscopy deals with the production, measurement,
and interpretation of spectra arising from the interaction
of electromagnetic radiation with matter.
2. Many different spectroscopic methods are available for
solving a wide range of analytical problems.
3. The methods differ with respect to
1. the species to be analyzed (such as molecular or
atomic spectroscopy),
2. the type of radiation-matter interaction to be monitored
(such as absorption, emission, or diffraction), and
3. the region of the electromagnetic spectrum used in the
analysis.

4. Spectroscopic methods are very informative and
widely used for both quantitative and qualitative
analyses.
5. Spectroscopic methods based on the absorption
or emission of radiation:
1. ultraviolet (UV),
2. visible (Vis),
3. infrared (IR), and
4. Radio (nuclear magnetic resonance, NMR)
6. Each of these methods is distinct in that it
monitors different types of molecular or atomic
transitions.

Emission of Radiation
Emission is essentially the reverse of the absorption
process, occurring when energy from an atom or
molecule is released in the form of a photon of radiation.
A molecule raised to an excited state will typically remain
in the excited state for a very short period of time before
relaxing back to the ground state.
There are several relaxation processes through which
an excited molecule may dissipate energy.
The most common relaxation process is for the excited
molecule to dissipate its energy through a series of small
steps brought on by collisions with other molecules.
Emission of Radiation
The energy is thus converted to kinetic energy, the net
result being the dissipation of the energy as heat. Under
normal conditions, the dissipated heat is not enough to
measurably affect the system.
In some cases, molecules excited by the absorption of
UV or Vis light will lose a portion of their excess energy
through the emission of a photon.
This emission process is referred to as either
fluorescence or phosphorescence, depending on the
nature of the excited state.
In molecular fluorescence spectroscopy, the photons
emitted from the excited species generally will be of
lower energy and longer wavelength than the
corresponding photons that were absorbed in the
excitation process.
UV-VIS Spectroscopy
One of the most commonly encountered
laboratory techniques in food analysis.
Electromagnetic radiation in the UV-Vis portion
of the spectrum ranges in wave-length from
approximately 200 to 700 um.
The UV range runs from 200 to 350 nm. and the
Vis range from 350 to 700 run
The UV range is colorless to the human eye,
while different wavelengths in the visible range
each have a characteristic color, ranging from
violet at the short wavelength end of the
spectrum to red at the long wavelength end of
the spec-trum.
UV-VIS Spectroscopy
Spectroscopy utilizing radiation in the UV-Vis
range may be divided into two general
categories, absorbance and fluorescence
spectroscopy, based on the type of
radiation-matter interaction that is being
monitored.
Each of these two types of spectroscopy may be
subdivided further into qualitative and
quantitative techniques.

Teknik ini digunakan untuk menganalisis
komponen-komponen kimia dalam
bahan bahan yang bila dieksitasi oleh
radiasi elektromagnetik akan
memancarkan radiasi fluoresent.
Instrument ini akan mengukur radiasi
fluorescent dari komponen bahan yang
dianalisis tersebut.
Proses pemancaran radiasi oleh suatu
zat setelah zat tersebut diradiasi
sebelumnya disebut sebagai
fluoresensi.

1. Why is it common to use absorbance values rather than
transmittance values when doing quantitative UV-Vis
spectroscopy?
2. For a particular assay, your plot of absorbance versus
concentration is not linear. Explain the possible reasons
for this.
3. What criteria should be used to choose an appropriate
wavelength at which to make absorbance measurements,
and why is that choice so important?
4. In a particular assay, the absorbance reading on the
spec-trophotometer for one sample is 2.033 and for
another sample 0.032. Would you trust these values?
