 Praktikum Biokimia gizi Fermentasi
1) Judul Pratikum : Fermentasi Peragian

3) Tujuan Praktikum :

● 1. Mahasiswa dapat melakukan analisis kerja enzim dalam pemecahan

glukosa dengan cara fermentasi
● 2. Mahasiwa dapat mengamati kerja enzim dalam pemecahan glukosa
 Praktikum Biokimia gizi Fermentasi

4) Prinsip Kerja : Enzim-enzim yang terdapat pada ragi roti akan membantu
pemecahan glukosa,fruktosa,mannosa,maltosa,atau sukrosa menjadi etil alkohol
dan CO2.
 5. Tinjauan Pustaka
Fermentasi timbul sebagai hasil metabolisme tipe anaerobik. Untuk hidup semua organisme membutuhkan sumber
energi-energi diperoleh dari metabolisme bahan pangan dimana organisme ada di dalamnya. Bahan baku yang paling banyak
digunakan di antara mikroorganisme adalah glukosa. Dengan adanya oksigen beberapa mikroorganisme mencerna glukosa
dan menghasilkan air, karbondioksida dan sejumlah bersar energi (ATP) yang digunakan untuk tumbuh. Ini adalah tipe
metabolisme aerobik. Akan tetapi beberapa mikroorganisme dapat mencerna bahan baku energinya tanpa ada oksigen dan
sebagai hasilnya bahan baku energi ini hanya sebagian yang dipecah.

Gula adalah bahan yang umum dalam fermentasi. Beberapa contoh hasil fermentasi adalah etanol, asam laktat dan
hidrogen. Akan tetapi beberapa komponen lain dapat juga dihasilkan dari fermentasi seperti asam butirat dan aseton. Ragi
dikenal sebagai bahan yang umum digunakan dalam fermentasi untuk menghasilkan etanol dalam bir,anggur dan minuman
beralkohol lainnya. Respirasi anaerobik dalam otot mmalia selama kerja yang keras (yang tidak memiliki akseptor elektron
eksternal), dapat dikategorikan sebagai bentuk fermentasi yang menghasilakn asam laktat sebagai produk sampingannya.
Akumulasi asam laktat inilah yang berperan dalam menyebabkan rasa kelelahan pada otot.

Pembahasan :

Dalam praktikum yang dilakukan hanya sampai langkah pencampuran 20 ml larutan gula

dengan 20 gr ragi yang kemudian di diamkan selama 1 jam, karena praktek yang dilaksanakan

dirumah masing-masing (online). Dari hasil praktikum dapat dilihat bahwa terdapat gelembung

pada permukaan. Ragi yang digunakan dalam praktikum ini adalah fermipan. Fermipan

merupakan ragi instant yang biasa dipergunakan dalam pembuatan roti dan kue. Fermipan atau

ragi digunakan agar bahan kue atau roti menjadi mengembang ketika dipanggang. Pada

praktikum yang telah dilakukan, kita mengetahui bahwa ragi yang dicampur dengan gula

menjadi mengembang. Maka setelah kita tahu bahwa dengan adanya gelembung, berarti ada

reaksi dari fermipan atau ragi dengan gula dan air.

 Reaksi fermipan sebagai berikut :

Ragi atau fermipan itu sendiri merupakan zat yang menyebabkan fermentasi. Ragi mengandung mikroorganisme yang
melakukan fermentasi dan media biakan ini dapat berbentuk  butiran-butiran kecil atau cairan nutrient. Mikroorganisme yang
digunakan di dalam ragi umumnya terdiri atas berbagai bakteri dan fungi, yaitu Rhizopus aspergillus,
Mucor,Amylomyces, Endomycopsis, Saccharomyces, Hansenula anomala, Lactobacillus, Acetobacter , dan sebagainya.

Setelah melihat adanya gelembung pada hasil praktikum seharusnya dilanjutkan dengan menambahkan NaOH encer untuk
mengetahui apakah gelembung tersebut merupakan CO2, tetapi karena keterbatasan alat dan bahan cukup sampai mengetahui adanya
reaksi ragi,gula dan air.
KESIMPULAN

Jadi kesimpulannya, fermipan dapat digunakan untuk fermentasi

yang dilakukan oleh sel ragi. Beberapa monosakarida seperti
glukosa,fruktosa dan manosa diragikan akan terbentuk etil alkohol dan
CO2. Dapat dilihat dari adanya gelembung gas pada hasil praktikum.
Glukosa di fermentasikan menjadi :
