Prinsip Kerja :
Pd kromatografi kolom, campuran yg akan
dipisahkan berupa pita pd bagian atas kolom
penjerap yang berada dalam tabung kaca, tabung
logam atau bahkan tabung plastik.
Kromatografi kolom/ tabung utk pengaliran krn gaya
tarik bumi (gravitasi) atau sistem bertekanan rendah
biasanya terbuat dr kaca yg dilengkapi keran jenis
ttt pd bag bawahnya utk matur aliran pelarut.
Pelarut (fase gerak) dibiarkan mengalir mll kolom,
krn aliran yg disebabkan ol gaya berat atau didorong
dg tekanan.
Pita senyawa sampel bergerak mll kolom dg laju yg
bbeda, memisah dan dikumpulkan mjd brp fraksi
ketika keluar dari kolom.

Adsorpsi mrp penyerapan pd
permukaan yg melibatkan interaksi2
elektrostatisk spt ikatan hidrogen,
penarikan dipol2 dan penarikan
induksi oleh dipol. Solut akan
bersaing dengan fase gerak utk
berikatan dg sisi aktif pada
permukaan adsorben.
Adsorption Chromatography
The silica gel is surrounded on all sides by
mobile phase. The solvent occupies all
active sites to a greater or lesser extent.
A sample molecule can only be adsorbed if
it interacts more strongly than the solvent
with the adsorbent.
All sample molecules (also the solvent) are
arranged on the silica gel surface so that
their functional group or double bond is
close to the silanol group.

kolom
Memisahkan campuran dari dua senyawa
yang berwarna, yaitu kuning dan biru.
Warna campuran yang tampak adalah
hijau.
Anda akan membuat larutan jenuh dari
campuran dengan menggunakan pelarut
yang lebih disukai dalam kolom.
Pertama anda membuka kran penutup
untuk membiarkan pelarut yang sudah
berada dalam kolom mengering sehingga
material terpadatkan rata pada bagian
atas, dan kemudian tambahkan larutan
secara hati-hati dari bagian atas kolom.
Lalu buka kran kembali sehingga campuran
berwarna akan diserap pada bagian atas
material terpadatkan, sehingga akan
tampak seperti gambar dibawah ini:

RP-Chromatography is the term used to

phase is less polar than the mobile phase.
Chemically bonded octadecylsilane (ODS) is
the most frequently used stationary phase.
In reverse-phase systems water cannot wet
the non-polar alkyl groups and does not
interact with them in any way.
The greater the amount of water in the
eluent, the longer is the retention time.
