PENDAHULUAN
Dalam desain bioreaktor ada tiga faktor penting yang harus
menjadi pertimbangan:
Faktor engineering.
Faktor biologis (karakteristik sel, maximum specific
growth rate, konstanta Monod, koefisien yield, range pH
dan range temperatur).
Cost/biaya juga menjadi faktor penting. Semakin besar
reaktor semakin tinggi kebutuhan agitasi dan biaya yang
dibutuhkan juga akan semakin besar.
Pada bab ini, kita akan melihat bagaimana bioreaktor
didesain untuk memenuhi ketiga hal tersebut diatas.

Foto dibawah ini adalah "Fernback" flask 3 liter yang
mengandung 1 liter medium dan Erlenmeyer flask 250 ml
mengandung 100 ml medium.

Baffled flask
Untuk aerasi volume liquid besar dari 200 ml, beberapa opsi
tersedia. Non-sparged STR dapat memberikan aerasi yang cukup
untuk volume liquid s.d. 3 liter. Namun perlu untuk menaikkan
kecepatan pengadukan s.d 600 rpm agar kultur tidak kekurangan
oksigen.
Pada non-sparged STR, oksigen ditransfer dari headspace diatas liquid fermentasi. Agitasi secara kontinu
memecah permukaan liquid dan meningkatkan
surface area untuk transfer oksigen.
Efek kecepatan stirrer/pengadukan pada entrainment
gas pada bioreaktor laboratorium 2 liter ditunjukkan
oleh gambar:

Agitasi biasanya digunakan untuk memecah
bubble sehingga menaikkan nilai kLa.
Fermenter dengan sparging membutuhkan
kecepatan agitasi yang jauh lebih rendah untuk
efisiensi aerasinya dibandingkan dengan
fermenter yang non-sparged. Air-sparged
fermenters dapat mempunyai volume kerja
liquid lebih dari 500,000 liter.
Disengagement zone
Fungsi dari Disengagement zone adalah untuk tambahan
volume pada reaktor, mengurangi foaming/busa dan
meminimalkan resirkulasi bubble melalui down comer.
Pelebaran bagian atas reaktor menurunkan kecepatan bubble
sehingga melepaskan bubble dari aliran liquid.
Aliran Carbon-dioxide yang banyak bubblenya maka akan
tercegah dari masuk ke aliran downcomer.
Kurangnya kecepatan bubble pada Disengagement zone
juga dapat mengurangi kehilangan medium akibat
pembentukan aerosol.
Peningkatan area juga akan mendorong bubble kedalam busa
dan membantu dalam proses pemecahan bubble dalam
foam/busa. Sirkulasi aliran axial akibat adanya draft tube juga
menurunkan pembusaan didalam reaktor.
Laju transfer massa antara sel dan medium tergantung dari flow rate/laju alir dan
ketebalan biomass film diatas/dekat permukaan solid particles.
Packed bed reactors sering mengalami masalah laju transfer massa yang rendah dan
penyumbatan/clogging. Diluar hal itu, reaktor ini secara komersial sudah dipakai
dengan katalis enzimatis dengan menggunakan sel yang slowly/non-growing.

Note how superficial gas velocity and thus the bubble rise velocity slows in
the disengagement zone.
