1. PENDAHULUAN
Oksidasi sangat penting bagi banyak makhluk hidup untuk memproduksi energi yang
dapat digunakan sebagai bahan bakar proses biologis (UNAL dan AKALIN, 2006). Radikal
bebas dan spesies oksigen aktif adalah metabolit fisiologis yang terbentuk sebagai hasil
respirasi pada organisme aerob. Jumlah radikal reaktif yang berlebihan dapat merusak semua
jenis makromolekul seluler, termasuk protein, karbohidrat, lipid, dan asam nukleat, yang
menyebabkan kematian sel dan kerusakan jaringan (UNAL dan AKALIN, 2006; FURST,
2009). Kerusakan sel ini dapat memicu risiko beberapa penyakit termasuk aterosklerosis,
radang sendi (arthritis), diabetes, dan kanker (SARMADI dan ISMAIL, 2010; ANDRÉ et al.,
2010). Juga dikenal bahwa oksidasi lipid yang terjadi dalam produk pangan menyebabkan
penurunan kualitas makanan, seperti rasa tengik, rasa yang tidak dapat diterima, dan
pemendekan umur simpan (PIHLANTO, 2006).
Dalam beberapa tahun terakhir, konsumsi yoghurt telah meningkat karena memenuhi
banyak kebutuhan makanan. Selain efek kesehatannya, yoghurt telah terbukti menjadi produk
yang sangat baik untuk produksi pangan fungsional, terutama yang mengandung bakteri
probiotik dan/atau bahan-bahan berbasis protein susu. Karena tidak ada perbedaan dalam
komposisi kotor antara yoghurt dan susu yang dapat menjelaskan kapasitas antioksidan yang
tinggi, diasumsikan bahwa peptida dan asam amino yang dibebaskan selama fermentasi
memainkan peran penting (FARVIN et al., 2010b). Aktivitas antioksidan telah dilaporkan
untuk protein susu (WOO et al., 2009), natrium kaseinate dan konsentrat protein whey
(SUGIART O et al., 2009) dan yoghurt (MCCUE dan SHETTY, 2005; FARVIN et al., 2010a;
2010b) . Kemampuan antioksidan bakteri asam laktat telah ditunjukkan oleh banyak peneliti
(KAIZU et al., 1993; SANDERS et al., 1995; LIN dan YEN, 1999; KUDOH et al., 2001; LIU
et al., 2005; SAIDE dan GILLILAND, 2005; KIM et al., 2005; VIRT ANEN et al., 2007; LIU
dan PAN, 2010; WANG et al., 2011; MONAJJEMI et al., 2012). Aktivitas antioksidan bakteri
probiotik, terutama Bifidobacteria, juga telah diselidiki melalui penelitian in vitro dan in vivo
(KANSAL, 2009; SHEN et al., 2010; 2011; HUANG et al., 2011; XU et al., 2011; RAJPAL
dan EJTAHED et al., 2012). Sebagian besar peneliti menyimpulkan bahwa pengembangan
aktivitas antioksidan adalah karakteristik spesifik-regangan. Selanjutnya, WANG et al. (2006)
menyelidiki aktivitas antioksidan susu kedelai yang difermentasi dan menyimpulkan bahwa
susu kedelai yang difermentasi dengan bakteri asam laktat dan bifidobacteria menunjukkan
aktivitas antioksidan yang secara signifikan lebih tinggi dibandingkan susu kedelai yang
difermentasi bakteri asam laktat atau bifidobacteria saja. Sejauh pengetahuan kami, aktivitas
antioksidan yoghurt probiotik dan pengaruh fortifikasi dengan bahan-bahan berbasis protein
susu pada aktivitas ini belum diteliti. Selanjutnya, aktivitas penangkapan hidrogen peroksida
yoghurt, sebagai indikator kapasitas antioksidan, belum diselidiki sebelumnya.
Tujuan dari penelitian ini adalah untuk mengevaluasi aktivitas antioksidan yoghurt
probiotik, yang mengandung Bifidobacterium animalis subsp. Lactis, dan pengaruh fortifikasi
dengan natrium kalsium kaseinate dan protein whey. Untuk melakukan penelitian ini, satu set
uji antioksidan in vitro, termasuk 2,2,-diphenyl-1-picrylhydrazyl (DPPH ·) efek penangkapan
radikal, penghambatan hidrogen peroksida (H2O2), dan digunakan aktivitas chelating Fe2+.
4. KESIMPULAN
Penelitian ini menunjukkan bahwa fortifikasi yoghurt probiotik, mengandung B.
animalis subsp. lactis, dengan bahan-bahan berbasis protein susu meningkatkan aktivitas
antioksidan dibandingkan dengan fortifikasi dengan susu bubuk skim. Penentuan aktivitas
penangkap radikal bebas dan H2O2 serta efek pengkhelat besi adalah parameter penting dalam
mengevaluasi kapasitas antioksidan in vitro. Aktivitas antioksidan dari sampel ditentukan
dengan metode yang berbeda tidak dapat dibandingkan. Karena metode ini berbeda satu sama
lain dalam hal mekanisme reaksi, oksidan, kondisi reaksi, perbandingan mungkin tidak sesuai
(EL dan KARAKAYA, 2004; KARADAG et al., 2009). Menggunakan metode penangkapan
radikal DPPH ·, fortifikasi yoghurt probiotik dengan konsentrat protein whey (WPC) lebih
menguntungkan daripada fortifikasi dengan natrium kalsium kaseinate (SCC). Di sisi lain,
yoghurt probiotik yang difortifikasi dengan natrium-kalsium kaseinate (SCC) memiliki
aktivitas khelat Fe2+ yang lebih tinggi daripada difortifikasi dengan konsentrat protein whey
(WPC). Fortifikasi dengan natrium kalsium kaseinate (SCC) atau konsentrat protein whey
(WPC) menyebabkan hal serupa efek pada aktivitas penangkapan H2O2. Karena aktivitas
antioksidan terjadi oleh mekanisme yang berbeda, menggunakan metode yang tergantung pada
satu mekanisme mungkin tidak mencerminkan kapasitas antioksidan yang sebenarnya. Oleh
karena itu, untuk mendapatkan pemahaman yang baik tentang sifat antioksidan dari produk
susu, penggunaan beberapa metode yang berbeda diperlukan (CHEN et al., 2003). Dalam
semua metode, yoghurt yang difortifikasi dengan campuran natrium kalsium kaseinate dan
konsentrat protein whey (SCC-WPC) menunjukkan aktivitas antioksidan setinggi yang
difortifikasi dengan hanya satu bahan. Oleh karena itu, menggunakan campuran kedua bahan
ini dapat direkomendasikan dalam fortifikasi yoghurt untuk mendapatkan peningkatan
aktivitas antioksidan.
