ABSTRAK
Tujuan : Tujuan dari penelitian ini adalah untuk mengetahui positivitas gen
KRAS pada jaringan dalam fiksatif formalin dengan menggunakan metode
PCR , sehingga diharapkan metode PCR dapat digunakan sebagai deteksi
molekular yang dapat membantu dalam diagnosis kanker.

TISSUE : STUDY OF RAT (Rattus norvegicus) WISTAR STRAIN
AFFECTED KRETEK SMOKEEXPOSURE
ABSTRACT
Background: Polymerase Chain Reaction has been widely used as one of the
most important invention of the 20th century in molecular biology. PCR has been
used to identify and manipulate DNA, detecting the organism infections (HIV,
hepatitis, tuberculosis, HIN5, H1N1), detecting genetic variation (SSR, RAPD,
AFLP) and others. Smoking is one part of lifestyle. Nearly a third of Indonesia's
population amounted to 237.56 million people were active smokers. Smoke
cigarettes was caused in gene or sequence of DNA bases chain that will affect the
growth of cancer cells, accelerate malignancy, and help metastasis.
Objective: The aim of this study was to determine the positivity of KRAS gene in
colorectal tissue in formalin fixative using the PCR method, which is expected
PCR method can help as a molecular detection in the diagnosis of cancer.
Methods: The study was conducted using a descriptive cross sectional (cross-
sectional). Researchers conducted DNA isolation and detection of KRAS gene
using the PCR method in 20 tissue samples of rat (Rattus norvegicus) in
formaldehyde, consisting of 10 control tissue and 10 tissue are exposed smoke
kretek (5 tissue termination until the 14th week and the 5 tissue termination until
28 th week for each group)
Conclusions: PCR positivity is about 75% for the detection of KRAS gene from
formalin fixative tissue.
