1. Sampel ikan diambil dari TPI (Tempat Pelelangan Ikan) Pantai Baron, Gunung Kidul,
D.I.Y.
2. Sampel ikan diidentifikasi dan dimasukkan ke dalam ice box dengan suhu 4ᵒC.
3. Kemudian sampel ikan dibawa ke laboratorium Biokimia Fakultas Biologi UGM untuk
diambil tulangnya dan dibuat gelatin
4. Sampel tulang hiu dipotong dan dibersihkan
5. Tulang ikan hiu sebanyak 200 gr dilarutkan dengan asam asetat 1,5% selama 12 jam
untuk ekstraksi dengan pelarut asam
6. Untuk ekstraksi dengan pelarut basa, tulang ikan hiu direndam dalam NaOH 0,3% selama
48 jam.
7. Kemudian kolagen yang diperoleh dari kedua pelarut dicuci dengan air mengalir sampai
pH netral
8. Setelah proses pencucian, kolagen diekstrak dengan air panas suhu 80ᵒC selama 2 jam
(perbandingan tulang dan air adalah 1 : 2)
9. Larutan gelatin yang diperoleh kemudian disaring dengan saringan ukuran 40 mesh,
dipekatkan dengan rotavapor (Buchi)
10. Selanjutnya larutan gelatin dikeringkan dengan oven dan kemudian digiling. Gelatin yang
diperoleh dari pelarut asam disebut dengan gelatin tipe A, sedangkan yang diperoleh
dengan pelarut basa disebut gelatin tipe B
11. Kemudian dilakukan pengujian kualitas gelatin

Ways of working
1. Fish samples taken from TPI (Fish Auction Place) Baron Beach, Gunung Kidul, D.I.Y.
2. Samples of fish and heat into the ice box with a temperature of 4ᵒC.
3. Then the fish samples are taken to the Biochemistry laboratory of Faculty of Biology UGM to
be taken bones and made gelatin
4. Sample shark bone cut and clean
5. Shark bone as much as 200 grams dissolved with 1.5% acetic acid for 12 hours for extraction
with acid solvent
6. For extraction with basic solvents, shark bone is immersed in 0.3% NaOH for 48 hours.
7. Then the collagen produced from both solvents is washed with active water until the pH is
neutral
8. After the washing process, collagen is extracted with hot water temperature 80ᵒC for 2 hours
(bone and air ratio is 1: 2)
9. The gelatin solution undergoes then filtered with a mesh size of 40 mesh, concentrated with a
rotary evaporator (Buchi)
10. Next the gelatin solution is dried with oven and then ground. The gelatin produced from the
acid solvent is called the type A gelatin, while the one obtained with the basic solvent is called
gelatin type B
