Pengertian Vitamin C
Vitamin C adalah salah satu
vitamin yang larut dalam air
dan memiliki peranan penting
dalam menangkal berbagai
penyakit. Vitamin ini juga
dikenal dengan nama kimia
dari bentuk utamanya yaitu
asam askorbat. Vitamin C
termasuk golongan vitamin
antioksidan yang mampu
menangkal berbagai radikal
bebas ekstraseluler.

Ada beberapa metode yang dikembangkan untuk penentuan kadar vitamin C

diantaranya adalah metode spektrofotometri UV-Vis (panjang gelombang 265
nm). Metode Spektrofotometri dapat digunakan untuk penetapan kadar
campuran dengan spektrum yang tumpang tindih tanpa pemisahan terlebih
dahulu. Karena perangkat lunaknya mudah digunakan untuk instrumentasi
analisis dan mikrokomputer, spektrofotometri banyak digunakan di bidang
analisis kimia.

Spektrofotometri adalah suatu metode analisis yang berdasarkan pada

pengukuran serapan sinar monokromatis oleh suatu lajur larutan
berwarna pada panjang gelombang yang spesifik dengan menggunakan
monokromator prisma atau kisi difraksi dan detector vacum phototube
atau tabung poto hampa.

Cara kerja spektrofotometeri ini dimulai dengan dihasilkannya cahaya

monokromatik dari sumber sinar. Cahaya tersebut kemudian menuju ke kuvet
(tempat sampel/sel). Banyaknya cahaya yang diteruskan maupun yang diserap
oleh larutan akan dibaca oleh detektor yang kemudian menyampaikan ke layar
pembaca.
