Ikan merupakan salah satu bahan makanan yang
mengandung berbagai macam zat, selain harga yang
murah, absorpsi protein ikan lebih tinggi dibandingkan
dengan produk hewani lain seperti daging sapi dan ayam,
PENDAHULUAN karena daging ikan mempunyai serat-serat protein lebih
pendek dari pada serat-serat protein daging sapi atau
ayam (Pandit,2008).
Ikan tenggiri (Scomberomorus commersoni) adalah
jenis ikan air laut yang merupakan kelompok ikan laut
pelagis yang memiliki citarasa khas sehingga digemari
oleh masyarakat. Menurut Depkes gizi protein yang
dihasilkan oleh ikan tenggiri cukup tinggi
yaitu21,4gr/100gr ikan (Depkes, 2008). Kualitas protein
ikan perlu dijaga agar tetap tinggi maka diperlukan proses
pengawetan ikan yang baik agar tidak merusak protein
yang ada dalam ikan tersebut.
PENDAHULUAN
Proses pengawetan ikan terbagi menjadi 2 yaitu: Menggunakan
pendingin seperti kulkas dan pengawetan secara tradisional yaitu
dengan metode penggaraman. Pada dasarnya terdapat tiga cara
penggaraman dalam pembuatan ikan asin, yaitu penggaraman kering,
penggaraman basah, dan kombinasi keduanya.
Analisis profil protein daging dilakukan dengan pemisahan protein menjadi
molekul yang lebih sederhana dengan menggunakan teknik elektroforesis SDS-
PAGE, selanjutnya dilakukan pengukuran jarak perpindahan (Rf) untuk
mengidentifikasi profil protein pada masing-masing sampel (Sandra Hermanto,
2009). Tujuan penelitian yaitu menganalisis profil protein padaikan tenggiri
sebelum dan sesudah penggaraman dengan variasi konsentrasi penggaraman
10% b/b, 20% b/b, dan 30% b/b dan lama penggaraman 12 jam, 24 jam, dan 36
jam dengan menggunakan metode SDS-PAGE.
SDS-PAGE (Sodium Dodecyl Sulphate Polyacrylamid Gel Electrophoresis)adalah suatu metode
elektroforesis yang digunakan untuk analisa pita protein secara kualitatif. Metode ini sering
digunakan untuk menentukan berat molekul suatu protein disamping untuk memonitor pemurnian
protein. Protein dalam gel dapat ditampakkan oleh pewarnaan Coomasie Brilliant Blue(Sudjadi,
2012).
METODE PENELITIAN
Metode penelitian ini adalah kualitatif dengan desain penelitian berupa penelitian eksperimen. Variabel
penelitian ini yaituprofil protein ikan tenggiri sebelum dan sesudah penggaraman dengan konsentrasi
penggaraman 10%, 20% dan 30% b/b dan lama perendaman selama 12 jam, 24 jam, dan 36 jam. Profil
protein ikan tenggirimerupakan sub-sub unit protein pada ikan tenggiriyang diperoleh dengan menggunakan
metode SDS-PAGE.
Objek penelitian ini adalah ikan tenggiri yang dibeli di pasar kobong semarang kemudian dilakukan
penggaraman dengan variasi konsentrasi 10%, 20%, dan 30% b/b dan direndam selama 12 jam, 24 jam, dan
36 jam.
Alat yang digunakan dalam penelitian ini yaitublender, labu ukur, pipet volume, beaker glass,timbangan
analitik, pot, cawan mortir,vortex,centrifuge, microtube, mikropipet, spektrofotometer,chamber elektroforesis,
power supply,waterbath,rotator, boxplastik, plastik pressdan kaca press
METODE PENELITIAN
Bahan yang digunakan dalam penelitian ini adalah ikan tenggiri, garam krosok, polyakrilamid 30%, TEMED,
APS 10%, SDS 10%, 1,5 M Tris pH 8,8 dan 6,8, staining 0,1% Coomasie Brilliant Blue (CBB) R-250,
destaining, asam asetat glasial 10%, butanol, alkohol 70%, running buffer 1x, biorad assay, PBS pH 7,4,
sampel buffer, dan marker protein.
