ANTIOXIDANT ACTIVITY TEST OF ETHANOL EXTRACT OF
TALOK LEAF (Muntingia calabura L.) WITH THE DPPH
METHOD (1,1 Diphenyl 2 - Picrylhydrazil)
ABSTRACT
Cherry plant (Muntingia calabura L.) is one of the plant that is widespread
in Indonesia. Utilization of cherry plants, especially the leaves are not widely known
by the public. Cherry leaves are known to have antioxidant activity that can be used
to treat various types of degenerative diseases. This research was conducted in order
to determine the antioxidant potential of extracts and fractions of cherry leaf and
secondary metabolites responsible as an antioxidant. The extraction method used is
the method of sonification using ultrasonic waves. The powder was extracted using
ethanol 96% and then evaporated to obtain a dry extract with a yield of 13.88%.
Extract and fractions of the cherry leaf were tested qualitatively using TLC method
and quantitatively using spectrophotometry UV - Vis method. Preliminary test
showed antioxidant activity on TLC plates were marked with yellow spots with the
purple background after spraying DPPH. The test results of antioxidant activity using
spectrophotometer UV - Vis showed that the ethanol extract has antioxidant activity
with IC50 values 4.98 ppm greater than fractions with IC50 values respectively -
participated in fraction F7, F6, F2, F5, F4, and F3 are 6.21; 7.17; 9.81; 11.78; 24.22
and 26.11 ppm. Quercetin as a comparison natural flavonoid compound have greater
antioxidant activity with IC50 values of 2.70 ppm compared to extract and its
fractions. Results of phytochemical screening by TLC were sprayed with the spray
reagent group is known that the cherry leaf extract and the fractions containing
secondary metabolites such as flavonoids and tannins.
