AKTIVITAS ANTIBAKTERI METABOLIT SEKUNDER

BAKTERI ENDOFIT DAUN PEGAGAN (Centella asiatica)


TERHADAP Escherichia coli PENYEBAB

Intisari
Latar belakang: Infeksi saluran kemih (ISK) merupakan istilah umum untuk
menunjukan keadaan infeksi pada saluran kemih, umumnya disebabkan oleh
uropatogenic Escherichia coli (UPEC) dan dapat diterapi dengan antibiotik.
Penggunaan antibiotik yang terlalu sering pada kasus ISK mengakibatkan
berkembangnya resistensi antibiotik, dimana penggunaan bakteri endofit daun
pegagan (Centella asiatica) menjadi solusi dalam mengatasi resistensi antibiotik.
Tujuan: Mengetahui aktivitas antibakteri metabolit sekunder bakteri endofit daun
pegagan terhadap Escherichia coli penyebab ISK. Metode: Penelitian ini bersifat
deskriptif yaitu isolat bakteri endofit daun pegagan diujikan dengan metode difusi
cakram terhadap Escherichia coli penyebab ISK. Isolat yang paling berpotensi
memiliki aktivitas antibakteri dilakukan uji metabolit untuk mengetahui senyawa
antibakteri yang dihasilkan. Identifikasi bakteri endofit berdasarkan morfologi
koloni, morfologi sel dan uji biokimia. Hasil: Sebanyak 2 dari 37 isolat yaitu I23
dan I24 mampu menghambat pertumbuhan Escherichia coli dengan zona hambat
sebesar 9,0 mm dan 9,1 mm (kategori resisten). Hasil uji metabolit menunjukan
bahwa isolate I23 dan I24 mengandung senyawa saponin. Kesimpulan: Isolat
bakteri endofit daun pegagan (Centella asiatica) tidak memiliki aktivitas
antibakteri terhadap Escherichia coli penyebab ISK.

ENDOPHYTIC BACTERIA FROM PEGAGAN LEAVES
(Centella Asiatica) AGAINST Escherichia coli THAT
CAUSES URINARY TRACT INFECTIONS

Abstract
Background: Urinary tract infections (UTI) is a general term to indivate the state
of infection in urinary tract, generally caused by uropathogenic Escherichia coli
(UPEC) and can be treated with antibiotics. The overly often usage of antibiotics
to UTI causes the development of antibiotic resistance, where the usage of
endophytic bacterial from Pegagan leaves (Centella asiatica) can resolve
antibiotic resistance. Objective: To determine the antibacterial activity of
secondary metabolite endophytic bacteria isolate from Pegagan leaves against
Escherichia coli that causes UTI. Method: Descriptive research where
endophytic bacterial isolates of Pegagan leaves were tested with disk diffusion
method against Escherichia coli that causes UTI. The most potential isolates with
antibacterial activity were performed metabolite test to determine the
antibacterial compounds produced. Endophytic bacteria identification based on
colony morphology, cell morphology and biochemical tests. Results: 2 out of 37
which are I23 and I24 were able to inhibit the growth of Escherichia coli with
obstacles zone of 9.0 mm and 9.1 mm (resistant). Metabolites test results showed
that isolate I23 and I24 contained saponins.. Conclusion: Endophytic bacterial
isolates of Pegagan leaves (Centella asiatica) does not have antibacterial activity
against Escherichia coli that causes UTI.
