Rumput laut memiliki peran

penting dalam ekosistem
laut. Alga merah berperan
sebagai produsen yang
secara ekonomis penting
sebagai penyedia makann
dan gel.

Alga merah atau Rhodophyta
mengandung klorofil a yang tertutupi
oleh pigmen biru yang terikat pada
protein. Kloroplas dalam alga merah
mirip dengan Chianobacteria baik
secara biologis maupun struturnya.
Cadangan makanan alga ini disimpan
di luar kloroplas sebagai floridien.

Terdapat lebih dari 6.000 spesies alga

merah yang sebagian besar terdapat
di laut dan sisanya di air tawar.
LATAR BELAKANG

ROS dikenal sebagai penyebab utama dari patogenesis katarak. Dalam

rumput laut merah kandungan protein bersama dengan karotenoid
memiliki aktivitas anti-oksidan yang tinggi untuk menghilangkan
Stres oksidatif. Alga merah pun dipilih kemudian dimaserasi dalam
berbagai sistem pelarut.

Hal ini menyebabkan diukurnya parameter fitokimia. Ekstrak Aqueous

dari Hypnea valentiae menunjukkan adanya kandungan protein,
karbohidrat, tannin dan cartotenoid. Ekstrak yang didaftar
menunjukkan adanya efek antigluconesgenesis dalam mata kambing
terisolasi.
 Berdasarkan beberapa hal yang telah diuraikan
ekstrak Hypnea valentiae diduga dapat
menghambat terjadinya difusi dan atau mencegah
perkembangan katarak. Dalam hal ini, anti-katarak
potensi dapat dikaitkan dengan adanya protein
tinggi dan karotenoid yang terdapat pada alga
merah.

METODE
Hypnea valentiae dikering, kemudian ditumbuk menjadi bubuk
kasar. Sekitar 500 gram bahan bubuk diekstraksi secara terpisah
dengan prosedur maserasi dingin berturut-turut dengan pelarut
yang dapat meningkatkan polaritas (Hexane, Kloroform, Etil
Ekstraksi asetat, Etanol, Methanol dan Aqeous). Pelarut disaring dan
disuling, kemudian dimurnikan menggunakan vakum.

Skrining Bola mata kambing segar diperoleh dari rumah

Farmakolog penyembelihan kambing dalam waktu dua jam
i setelah penyembelihan dan lensa diisolasi,
kemudian dibawa ke laboratorium pada 0-4°C.
Lensa diisolasi dan diinkubasi dalam aqueous
buatan pada 37°C dan pH 7,8 selama 72 jam.
Aktivitas katarak Glukosa pada konsentrasi 55 mM digunakan untuk
in-vitro menginduksi katarak.
 Sebanyak 42 lensa mata kambing yang digunakan dan dibagi menjadi tujuh
kelompok eksperimen yang terdiri dari 5 dalam setiap kelompok.
Kelompok I: aqueous buatan

Setelah inkubasi, lensa dihomogenisasi dengan 10 volume
buffer kalium fosfat 0,1 M, pH 7,0. Homogenat disentrifugasi
pada 10.000 g selama 1 jam dan supernatan digunakan untuk
estimasi parameter biokimia. Setelah 72 jam inkubasi dalam
Persiapan Lensa glukosa 55 mM, lensa menjadi benar-benar buram. kontrol
Homogen normal (Gambar 1a).
Inkubasi lensa dengan HV 100 μg / 200μg (Uji 1 dan 2),
pada konsentrasi yang digunakan, tampaknya menghambat
perkembangan kekeruhan lensa, dibandingkan dengan lensa yang
diinkubasi dalam glukosa 55 mM (kontrol negatif, Gambar 1b).
Efek vitamin E, kelompok kontrol positif menunjukkan retardasi
yang cukup besar dalam perkembangan kekeruhan lensa yang
mendekati normal jika dibandingkan dengan kontrol negatif.
