Apa itu Non-Homologous End Joining?

Non-Homologous End Joining (NHEJ) atau

Penggabungan ujung non-homolog adalah jalur
yang memperbaiki kerusakan untai ganda pada
DNA atau Double-Strand Break (DSB).
Disebut "non-homolog" karena ujung yang
putus diikat secara langsung tanpa memerlukan
template homolog.

Mekanisme Non-Homologous End Joining (NHEJ)

 Ketika untai ganda DNA yang rusak dan

terputus atau mengalami DSB, ini
menunjukkan sekuens-sekuens DNA yg
hilang karena kerusakan yg terjadi.
 Proses perbaikan DNA oleh nuclease yaitu
protein KU 70 dan KU 80. Protein KU 70
dan KU 80 mengikat ujung untai DNA yang
terputus, dan DNA-PKcs akan direkrut dan
melekat pada KU protein.
 Selanjutnya protein artemis yang bergabung
dalam kompleks akan membersihkan
nucleotide yang tersisa pada sekuens DNA
yang terputus.
 Selanjutnya DNA polymerase akan
bergabung dalam kompleks.
 Pada tahap akhir yaitu DNA ligase IV akan
menggabungkan kembali untai DNA double
helix yg terputus sehingga akan terbentuk
DNA baru.
